[Expression of metalloproteinase MMP-9 and tissue inhibitor of metalloproteinase TIMP-2 in placenta of pregnant women with intrauterine growth restriction].

INTRODUCTION: The development of pregnancy depends on many factors, among other the implantation and the development of the placenta and placental vascularity The correct course of these processes depends on the proper activity of the enzymes degradating elements of extracellular matrix and basal membrane - metalloproteinases. OBJECTIVES: Assessment of the expression of MMP-9 and TIMP-2 in the placenta of women with intrauterine fetal growth restriction. MATERIAL AND METHODS: Two study groups were distinguished - with untreated IUGR and with IUGR treated with the acetylsalicylic acid and L-a

Expression of the homeobox genes OTX2 and OTX1 in the early developing human brain.

In rodents, the Otx2 gene is expressed in the diencephalon, mesencephalon, and cerebellum and is crucial for the development of these brain regions. Together with Otx1, Otx2 is known to cooperate with other genes to develop the caudal forebrain and, further, Otx1 is also involved in differentiation of young neurons of the deeper cortical layers. We have studied the spatial and temporal expression of the two homeobox genes OTX2 and OTX1 in human fetal brains from 7 to 14 weeks postconception by in situ hybridization and immunohistochemistry. OTX2 was expressed in the diencephalon, mesencephalon

Somatic cell nuclear transfer is associated with altered expression of angiogenic factor systems in bovine placentomes at term.

Low efficiency of somatic cell cloning by nuclear transfer has been associated with alterations of placental vascular architecture. Placenta … ctors are involved in abnormal placental development in cloned gestations, contributing to impaired fetal development and poor survival rates.

MAMLD1 (CXorf6): a new gene involved in hypospadias.

MAMLD1 (mastermind-like domain containing 1), previously known as CXorf6 (chromosome X open reading frame 6), has been shown to be a causative gene for hypospadias. This is primarily based on the identification of nonsense mutations (E124X, Q197X, and R653X), which undergo nonsense-mediated mRNA decay, in patients with penoscrotal hypospadias. Subsequent studies have shown that (1) the mouse homolog is transiently expressed in fetal Sertoli and Leydig cells around the critical period of sex development; (2) transient knockdown of MAMLD1 results in significantly reduced testosterone production

Cardiac manifestations of neonatal lupus erythematosus: guidelines to management, integrating clues from the bench and bedside.

One of the strongest clinical associations with autoantibodies against components of the SSA/Ro-SSB/La ribonucleoprotein complex is the development of congenital heart block in an offspring, an alarming prospect facing 2% of primigravid mothers with these reactivities. This risk is increased tenfold in women who have had a previous child with congenital heart block. Accumulated evidence suggests that anti-SSA/Ro and anti-SSB/La antibodies are necessary but insufficient for fetal disease. Basic and clinical research is heavily focused on identifying fetal and environmental factors that convert

Cysts of PRKCSH mutated polycystic liver disease patients lack hepatocystin but express Sec63p.

Polycystic liver disease (PCLD) is an inherited disorder caused by mutations in either PRKCSH (hepatocystin) or SEC63 (Sec63p). However, expression patterns of the implicated proteins in diseased and normal liver are unknown. We analyzed subcellular and cellular localization of hepatocystin and Sec63p using cell fractionation, immunofluorescence, and immunohistochemical methods. Expression patterns were assessed in fetal liver, PCLD liver, and normal adult liver. We found hepatocystin and Sec63p expression predominantly in the endoplasmic reticulum. In fetal tissue, there was intense expressio

Connexin expression in renin-producing cells.

Absence of connexin 40 (Cx40) leads to ectopic juxtaglomerular renin expression and abrogates recruitment of renin-expressing cells in the adult kidney but does not disturb renin expression during kidney development. To find an explanation for these observations, we aimed to analyze the expression pattern of major vascular Cxs in normal juxtaglomerular epithelioid cells, in recruited renin-expressing cells, and in fetal renin-expressing cells. We found that during kidney development, the appearance of renin-producing cells paralleled the expression of Cx40 and, to a lesser extent, Cx45 but not

Association between prenatal sonographic findings and post-natal outcomes in 30 cases of isolated spina bifida aperta.

This study presents 30 cases of fetal isolated spina bifida aperta (SBA) to identify prenatal ultrasound findings that could predict the prognosis. Comparisons between surviving patients who had normal (group 1) and abnormal (group 2) post-natal neurological outcomes were made for three different prenatal signs, that is, site of vertebral lesion, presence and degree of ventriculomegaly and presence of talipes. The site of the lesion was the most significant outcome predictor, as high spinal dysraphisms were observed in 2 patients (2/7-28.6%) in group 1 and in 15 patients (15/19-79.0%) in group

Infection and preterm birth: evidence of a common causal relationship with bronchopulmonary dysplasia and cerebral palsy.

Subclinical genital tract infection has been clearly established as a significant cause of spontaneous preterm birth, particularly in early gestations. Bacterial vaginosis organisms rank highly among the pathogens involved in preterm labour and there is considerable beneficial evidence from the use of prophylactic antibiotics for women at high risk of preterm birth. The pathogenesis involves activation of macrophages and the generation of pro-inflammatory cytokines. Bronchopulmonary dysplasia is seen in the most immature of survivors and appears to be secondary to interruption of normal develo

HIV-1 infection of primary human neuroblasts.

Central nervous system (CNS) disorders are frequent in HIV-1-infected individuals, particularly in newborns and children, and are accompanied by histological alterations resulting in neuronal loss. Although several tumor-derived neuroectodermal cell lines can be infected by HIV-1, it has been reported that primary neural cells cannot be infected after they differentiate. However, pediatric AIDS is often the result of HIV-1 infection occurring during fetal development and early postnatal life, when neural cells are not yet differentiated. Here we show that primary cell cultures derived from the…

Trichloroethylene-induced pneumotoxicity in fetal and neonatal mice.

Trichloroethylene (TCE) has previously been shown, in adult mouse lung, to alter the distribution of phospholipid associated with the pulmonary surfactant and to affect the activity of phospholipase A2, an enzyme involved in synthesis of the surfactant. However there are no data available on the effects of maternal exposure to TCE on fetal lung development. To determine if maternal TCE exposure may affect fetal pulmonary development, pregnant mice were treated with TCE (3000 mg/kg body wt.) administered intraperitoneally on day 17 of pregnancy. Fetuses were examined on the 18th and 19th gestat

Emulation of seizure induced brain damage in neural tissue transplants to the anterior chamber of the eye.

We have developed a model system in which the mechanisms of neuronal damage due to hyperexcitation can be studied in isolation and where extended observation periods can be used. Substantia nigra pars reticulata (SNPR) develops a hypermetabolic necrosis following status epilepticus (Nevander et al. 1985; Auer et al. 1986). We transplanted rat fetal nigral area alone or together with fetal frontal neocortex to the anterior chamber of the eye in adult rats. Following 3 months of transplant maturation the hosts were subjected to status epilepticus for 60 min. In single nigral transplants no sign

The effect of ten years of regionalized perinatal health care in Iowa, U.S.A.

In 1973, Iowa, a rural, midwestern state in the U.S.A., established a Statewide Perinatal Care Program. A major objective was to develop and maintain a regionalized system of care. Such a system has been developed but differed from traditional systems by using regional level II centers. Iowa's low population density necessitated this modification. The development and maintenance of Iowa's system is described. Evaluation is provided by analysis of commonly available birth and mortality data. Improved birth-weight-specific neonatal and fetal mortality rates suggest an overall improvement in care

A fetal lamb model of partial urethral obstruction: experimental protocol and results.

The functional effects of bladder outlet obstruction in the developing urinary tract are well recognized in patients born with posterior urethral valves, in whom a spectrum of bladder dysfunction has been described. To better understand the changes occurring in the partially obstructed developing lower urinary tract, a fetal lamb model of partial urethral obstruction was developed. Fetal lambs at 90 days' gestation underwent surgical placement of a silver ring (ex utero) at the level of the proximal bladder neck, with concomitant ligation of the urachus. Control animals underwent urethral liga

Cesarean Delivery in Fetal Triploidy: Clinical Considerations and Case Study Insights.

BACKGROUND Fetal triploidy is a rare, lethal disorder characterized by an extra set of haploid chromosomes resulting in 69 chromosomes (69, XXX; 69, XXY; or 69, XYY). Fetal anomalies and occasionally maternal complications such as hypertension result in a high fetal loss rate during gestation. It is estimated to be present in 1: 250 000 pregnancies at 20 weeks, and very few survive to term. Diagnosis is suspected by antepartum ultrasound and confirmed by invasive testing of fetal cells by karyotype. Management includes pregnancy termination or pregnancy continuation based on the patient's choi

Pregnancy-related listeriosis in Spain.

BACKGROUND: Pregnant women are at high risk of acquiring listeriosis, resulting in severe fetal and neonatal outcomes. METHODS: All hospitalizations with a listeriosis diagnosis in pregnant women (obstetric listeriosis) and/or newborns (neonatal listeriosis) in Spain from 2000 to 2021 were examined using the National Registry of Hospital Discharges, employing ICD-9 and -10 coding lists. RESULTS: A total of 540 and 450 hospital admissions for obstetric listeriosis and neonatal listeriosis were identified, respectively, with 146 adverse fetal-neonatal outcomes (miscarriage, fetal loss, stillbi

Randomized controlled trial of prenatal zinc supplementation and fetal bone growth.

BACKGROUND: Maternal zinc deficiency is relatively common in developing countries, but its consequences for fetal growth are not established. OBJECTIVE: The goal was to examine whether improvement in maternal gestational zinc status is positively associated with fetal growth as assessed by ultrasonography. DESIGN: We conducted a double-masked, randomized trial among 242 pregnant Peruvian women in an impoverished shantytown in Lima, Peru. At 10-16 wk of gestation, the women were randomly assigned to receive daily supplements containing 60 mg Fe and 250 microg folic acid, with or without 25 mg

New approach to document cannabis toxicity during pregnancy: a low abundance molecular species detection in long-term formalin-fixed paraffin embedded fetal tissues. A technological report.

BACKGROUND AND AIMS: Cannabis is a widely abuse substance during pregnancy. However, reports of effects on human fetal brain development and implication in brain malformations are scarce in the literature. This study aims to determine the usability of long-term formalin-fixed tissues, highly degraded as a result of long post-mortem delays, collected during post-mortem medical examinations of human fetuses for molecular identification of low-abundance molecular species, namely the cannabinoid receptors, as a prerequisite for studying cannabis role in human fetal brain malformations. METHODS: Th

Syncytin-1 deficiency impairs placental nutrient transport via PI3K/Akt/mTOR signaling.

The placenta, a vital organ bridging the fetus and mother, governs nutrient exchange. Syncytin-1, an endogenous retroviral envelope protein specifically expressed in placental trophoblasts, is diminished in preeclampsia and fetal growth restriction. This study aimed to investigate the effects of low syncytin-1 expression on placental transport of amino acids, fatty acids and cholesterol, and its implications for fetal and placental development, contributing to fetal growth restriction pathogenesis. Pregnant C57BL/6J mice received tamoxifen-induced conditional syncytin-a gene knockout at embryo
