Unlocking Performance: Pharmacogenomic Insights for Athletes.

Pharmacogenomics is the study of how genetic differences influence drug responses, offering a safer and more effective prescribing model. While pharmacogenomics is well-established in specialties such as oncology and psychiatry, its application in sports medicine remains limited. Athletes frequently use medications such as nonsteroidal anti-inflammatory drugs, opioids, and selective serotonin reuptake inhibitors, and these medications have well-documented gene-drug interactions. This review explores pharmacogenomics considerations for these commonly used substances in athletic populations, emp

Genomic Diversity of Human Respiratory Syncytial Virus (HRSV) Type A in a Pediatric Cohort in the United States After the Onset of the COVID-19 Pandemic.

Human Respiratory Syncytial Virus (HRSV) is a leading cause of respiratory infections in young children. HRSV cases declined dramatically in the initial stages of the COVID-19 pandemic, followed by a resurgence in 2022/2023. To investigate patterns of viral genetic diversity and evolutionary dynamics during this period, HRSV sequences were analyzed at national and global levels, including newly sequenced samples from pediatric patients at a major Florida medical center in Alachua County. Our Florida strains clustered within multiple clades, with no clear association between specific clades and

Habitat Mosaic Limits Gene Flow and Promotes Morphological Adaptation in a Generalist Mammal.

Many habitat generalist species exploit habitat patches of differing types and quality, yet the influence of such habitat mosaics on genetic structure remains poorly understood. Here, we tested whether fine-scale habitat heterogeneity affects the population structure of the European wood mouse (Apodemus sylvaticus) by sampling three matched forest parks in Northern Ireland across hedgerow, forest edge, and inner forest habitats. Microsatellite analysis revealed strong genetic differentiation among sites and consistent divergence between habitat types within sites. Stable isotope data showed th

Identification of Interactive Genetic Loci Linked to Insulin Resistance in Metabolic Syndrome-An Update.

Metabolic syndrome is a metabolic disorder characterized by hypertension, dyslipidemia, impaired glucose tolerance, and abdominal obesity. Impaired insulin action or insulin resistance initiates metabolic syndrome. The prevalence of insulin resistance is increasing all over the world. Insulin resistance results in the defective metabolism of carbohydrates and lipids, in addition to low-grade chronic inflammation. Insulin resistance is associated with metabolic syndrome, which is a risk factor for a number of pathological conditions, such as Type 2 diabetes (T2D), cardiovascular disease (CVD),

Implementing a New Algorithm for Reinterpretation of Ambiguous Variants in Genetic Dilated Cardiomyopathy.

Dilated cardiomyopathy is a heterogeneous entity that leads to heart failure and malignant arrhythmias. Nearly 50% of cases are inherited; therefore, genetic analysis is crucial to unravel the cause and for the early identification of carriers at risk. A large number of variants remain classified as ambiguous, impeding an actionable clinical translation. Our goal was to perform a comprehensive update of variants previously classified with an ambiguous role, applying a new algorithm of already available tools. In a cohort of 65 cases diagnosed with dilated cardiomyopathy, a total of 125 genetic

Polygenic risk score and age: an extra help in the cardiovascular prevention of the young?

All major guidelines recommend assessing the risk of atherosclerotic cardiovascular disease (ASCVD) using risk scores. In fact, it has been shown that their use at the population level increases the accuracy of event prediction and facilitates the choice of strategies to be adopted in primary prevention. In fact, their use in clinical practice is far from optimal and their predictive ability on an individual level is not excellent. Our genetic heritage is substantially stable from birth and determines a 'baseline risk' on which external influences act. Genetic information therefore has the pot

Relationship between rs7586085, GALNT3 and CCDC170 gene polymorphisms and the risk of osteoporosis among the Chinese Han population.

Osteoporosis (OP) has plagued many women for years, and bone density loss is an indicator of OP. The purpose of this study was to evaluate the relationship between the polymorphism of the rs7586085, CCDC170 and GALNT3 gene polymorphisms and the risk of OP in the Chinese Han population. Using the Agena MassArray method, we identified six candidate SNPs on chromosomes 2 and 6 in 515 patients with OP and 511 healthy controls. Genetic model analysis was performed to evaluate the significant association between variation and OP risk, and meanwhile, the multiple tests were corrected by false discove

The impact of the COVID-19 preventive measures on influenza transmission: molecular and epidemiological evidence.

OBJECTIVE: We quantify the impact of COVID-19-related control measures on the spread of human influenza virus H1N1 and H3N2. METHODS: We analyzed case numbers to estimate the end of the 2019-2020 influenza season and compared it with the median of the previous 9 seasons. In addition, we used influenza molecular data to compare within-region and between-region genetic diversity and effective population size from 2019 to 2020. Finally, we analyzed personal behavior and policy stringency data for each region. RESULTS: The 2019-2020 influenza season ended earlier than the median of the previous

BDNF Genetic Variant and Its Genotypic Fluctuation in Major Depressive Disorder.

Major depressive disorder (MDD) still has an unknown etiology and mechanisms. Many studies have been conducted seeking to associate and understand the connection of different genetic variants to this disease. Researchers have extensively studied the brain-derived neurotrophic factor (BDNF) Val66Met genetic variant in MDD; yet, their findings remain inconsistent. This systematic review sought to verify the GG (Val/Val) genotype frequency fluctuation in different populations with MDD. For this, we searched in different databases and, after applying the eligibility criteria, selected 17 articles.

Genetic and environmental factors related to the development of myopic maculopathy in Spanish patients.

High myopia and the subsequent degenerative changes of the retina, choroid, and sclera, known as myopic maculopathy (MM), are a serious visual problem in many Asian countries, and are beginning to be so in the south of Europe, especially in the Mediterranean. It is therefore necessary to carry out genetic and environmental studies to determine the possible causes of this disease. This study aims to verify if the genetic factors that have been most related to Asian populations are also associated in two Spanish cohorts. Eight SNPs from six genes (PAX6, SCO2, CCDC102B, BLID, chromosome 15q14, an

No association between dysplasminogenemia with p.Ala620Thr mutation and atypical hemolytic uremic syndrome.

Atypical hemolytic uremic syndrome (aHUS), a form of thrombotic microangiopathy, is caused by the uncontrolled activation of the alternative pathway of complement on the cell surface that leads to microangiopathic hemolytic anemia, thrombocytopenia, and renal failure. A recent genetic analysis of aHUS patients identified deleterious mutations not only in complement or complement regulatory genes but also in the plasminogen gene, suggesting that subnormal plasminogen activity may be related to the degradation of thrombi in aHUS. Dysplasminogenemia, which is caused by a genetic variant in the pl

Implementation of an optimized strategy for genetic testing of the Chinese patients with oculocutaneous albinism.

BACKGROUND: Oculocutaneous albinism (OCA) is a relatively common inherited disorder in all populations worldwide. The mutational spectra of OCA are population-specific. OBJECTIVE: Based on our previous molecular epidemiological studies, we have implemented an optimized strategy for the genetic testing of Chinese OCA patients. METHODS: Genomic DNA was extracted from the blood samples of 52 clinically diagnosed OCA patients and 100 unaffected subjects. The amplified DNA segments were screened for mutations of TYR, OCA2, TYRP1, SLC45A2 and HPS1 by direct sequencing. To exclude the previously un

Heritability of human hookworm infection in Papua New Guinea.

Hookworms infect approximately 740 million humans worldwide and are an important cause of morbidity. The present study examines the role of additive genetic effects in determining the intensity of hookworm infection in humans, and whether these effects vary according to the sex of the host. Parasitological and epidemiological data for a population of 704 subjects in Papua New Guinea were used in variance components analysis. The 'narrow-sense' heritability of hookworm infection was estimated as 0.15+/-0.04 (P<0.001), and remained significant when controlling for shared environmental (household

HIV-1 subtypes among blood donors from Rio de Janeiro, Brazil.

The prevalence of HIV infection in Brazil is one of the highest in the world. In addition, transfusion-transmitted HIV accounts for 2.3% of all AIDS cases in Brazil. The objective of this study was to evaluate genetic diversity and distribution of HIV-1 strains circulating in the blood-donor population. We characterized 43 seropositive blood units collected from volunteer blood donors residing throughout Rio de Janeiro, Brazil. Viral RNA was extracted from plasma, reverse transcribed, and amplified by nested polymerase chain reaction (PCR) using HIV group M degenerate primers. Genetic heteroge

Age-dependent expression of familial risk in Alzheimer's disease.

There is growing evidence for genetic causes of Alzheimer's disease; therefore, consideration of its age at onset among genetically predisposed individuals is timely. The authors studied the onset of Alzheimer-like dementia in families of 79 probands ascertained clinically for the Bronx Veterans Administration/Mt. Sinai School of Medicine longitudinal studies of Alzheimer's disease. Analyses among their 379 first-degree relatives, living and decreased, suggested that 1) early onsets form a distribution that is distinct from that of later onsets, and thus, different causes or mechanisms may ope

Idiopathic cholestasis in adults: Genetics as another lens for liver pathologists.

Chronic liver disease (CLD) is a significant global health problem, responsible for approximately two million deaths annually. Despite extensive diagnostic evaluations, a proportion of patients with CLD remain undiagnosed, making the study and quantification of these cases challenging. Genetic testing has emerged as a critical diagnostic tool, identifying monogenic causes in a substantial portion of undiagnosed cases. In addition, nearly 50 % of known genetic liver diseases were identified in the past 20 years, underscoring the rapid expansion of genetic knowledge. Notably, recent data demonst

Committee Opinion No. 691: Carrier Screening for Genetic Conditions.

Carrier screening is a term used to describe genetic testing that is performed on an individual who does not have any overt phenotype for a genetic disorder but may have one variant allele within a gene(s) associated with a diagnosis. Information about carrier screening should be provided to every pregnant woman. Carrier screening and counseling ideally should be performed before pregnancy because this enables couples to learn about their reproductive risk and consider the most complete range of reproductive options. A patient may decline any or all screening. When an individual is found to be

Congenital myasthenic syndromes: achievements and limitations of phenotype-guided gene-after-gene sequencing in diagnostic practice: a study of 680 patients.

Congenital myasthenic syndromes (CMSs) are clinically and genetically heterogeneous disorders characterized by a neuromuscular transmission defect. Even though CMSs are genetic disorders, they are highly treatable, and the appropriate drug treatment depends on the underlying genetic defect. This highlights the importance of genetic testing in CMS. In recent years, the molecular basis of CMS has constantly broadened and disease-associated mutations have been identified in 14 genes encoding proteins of the neuromuscular junction. In the dawn of novel sequencing strategies, we report on our 14-ye

Emergence of Circulating Recombinant Form 56_cpx in Pakistan.

In Pakistan, HIV has converted from outbreak to concentrated epidemic and has also bridged into the low-risk population. The HIV epidemic in Pakistan mainly comprises subtype A. Here, we present the first case and genetic analysis of a circulating recombinant form 56_cpx in a Pakistani HIV-infected patient. Genetic analysis of the sequence indicated that Pakistani 56_cpx sequence had more drug resistance mutation than the other 56_cpx sequences available in the database.

Higher plasma vitamin D levels are not causally associated with less age-related macular degeneration.

PURPOSE: Higher levels of plasma 25-hydroxyvitamin D have been associated with a lower risk of age-related macular degeneration (AMD). We investigated observational and potential causal associations of plasma 25-hydroxyvitamin D with the risk of AMD in two large contemporary population-based prospective studies and a genetic case-control consortium. METHODS: Observationally, we studied 25 445 individuals with measured baseline plasma 25-hydroxyvitamin D (536 incident AMD events) in the Copenhagen General Population Study (CGPS) and 372 477 individuals (7842 incident AMD events) in the UK Biob
