Purification of a non-enveloped virus using sequential aqueous two-phase extraction.

Virus-based vaccines and therapies require a purification method that is both cost-effective and easily scalable. An aqueous two-phase system (ATPS) consisting of polyethylene glycol (PEG) and citrate salt has been proven to deliver high virus recoveries along with high impurity removal. However, these systems often place the virus into a viscous PEG-rich phase or at the two-phase interface, leading to difficulties in subsequent downstream processes. This study explored a second ATPS to extract the virus product back into the citrate-rich phase by changing the chemical conditions, a required s

Exploring the Role of Aqueous Buffered Saline Solutions on the Macroscopic Properties of PEG/Carbomer/Agarose Hydrogels.

Nowadays, during hydrogel formulation design, the focus is primarily on polymers and cross-linkers. It is crucial to ensure a high level of biocompatibility and degradability, along with appropriate physico-chemical properties. However, other ingredients can also influence hydrogel formation and performance. Therefore, this study examines the effect of different salt molarities in phosphate buffered saline (PBS) solutions, used as reaction solvents, on the properties of hydrogels mainly composed of polyethylene glycol, agarose, and carbomer. To conduct the study, two PBS solutions are prepared

Double-Helix Duality: Rods Bow, Toroids Wow in the Nuclease Arena.

Utilizing polyion complexation, the formation of rod-like DNA condensates is driven by the intrinsic rigidity of supramolecular plasmid DNA. Upon interaction with polycationic block copolymers of poly(ethylene glycol)-polylysine (PEG-PLys), these macromolecules undergo a regular self-folding process, during which double-stranded DNA (dsDNA) transitions into single-stranded DNA (ssDNA) at the kinked junctions. Our investigations, employing transmission electron microscopy (TEM), unprecedentedly reveal the absence of a PEG coating at these critical junctions, rendering them susceptible to nuclea

Unraveling the time course of interaction between DNA nanopores and lipid bilayers using QCM-D: role of cholesterol anchors and bilayer supporting substrates.

Lipid membranes are fundamental elements of cells, serving as barriers that protect the cell interior from the external environment. DNA nanostructures, which can engineer lipid membranes' signal transduction and substance exchange properties, attract interest for their potential applications in the biomedical field. The interaction between DNA nanostructures and lipid membranes is of scientific and technological interest. Here, we investigate the interaction between DNA nanopores (DNPs) and model supported lipid bilayers (SLBs) using real-time quartz crystal microbalance with energy dissipati

Surfactant additives in water-based metalworking fluids lead to strong biophysical inhibition of pulmonary surfactant film.

As a class of very common and important industrial liquid materials, metalworking fluids (MWFs) are easy to be vaporized into the air and cause pulmonary toxic effects. The inhaled MWF aerosols should first interact with the pulmonary surfactant (PS) film that plays an essential role in maintaining the normal respiratory mechanics and pulmonary immunology in a human body. Here, to probe any potential adverse impacts of airborne MWFs on the biophysical and physiological functions of PS film that may help achieve a deep and comprehensive understanding of the pulmonary toxicology of MWFs, a syste

From in vitro to in vivo: The Dominant role of PEG-Lipids in LNP performance.

Lipid nanoparticles (LNPs) are widely employed for delivering nucleic acids, such as mRNA, in both vaccines and therapeutic applications. These LNPs generally include an ionisable lipid (typically ∼ 50 %) to facilitate nucleic acid encapsulation and a PEG-lipid (typically ∼ 1.5 %) to enhance nanoparticle stability. To examine how the choice of PEG-lipid impacts LNP performance, we investigated the physicochemical characteristics and potency of LNPs prepared using two PEG-lipids with different acyl chain lengths: DMG-PEG 2000 and DSG-PEG 2000, containing 14 and 18 carbon tail lengths, respectiv

rhGALNS Enzyme Stability in Physiological Buffers: Implications for Sustained Release.

Morquio A syndrome is a rare genetic disorder where deficiency in N-acetylgalactosamine-6-sulfate sulfatase (GALNS) enzyme prevents breakdown of glycosaminoglycans (GAGs). Recombinant human GALNS (rhGALNS) is currently administered by intravenous infusion, but the treatment is costly and time-consuming and provides limited efficacy. Patient quality of life could be improved by an injectable sustained rhGALNS release device that would eliminate weekly multi-hour infusions. Polyethylene glycol (PEG) hydrogels can be employed as a hydrophilic, tunable, non-toxic, and biodegradable drug delivery s

Development of CS/PLA Composites with Enhanced Ductility via PBS Elastomer Reinforcement.

Polylactic acid (PLA) exhibits remarkable biocompatibility and biodegradability, rendering it a highly promising material for applications in packaging and disposable products. However, its inherent brittleness, low melt strength, and slow crystallization rate significantly restrict its practical uses. Our previous studies have shown that incorporating the ADR chain extender can yield chitosan-polylactic acid-ADR (CS/PLA-ADR) composites with outstanding antibacterial properties, enhanced biodegradability, and the capability to effectively block water vapor and oxygen. However, the low elongati

Charge microenvironment and bioactivity of in situ-formed PEG-RGD dual hydrogel dressings promote wound healing.

Healing of large skin wounds involves a complex biological process with overlapping phases, facing challenges from fibroblast proliferation, immune response, and extracellular matrix (ECM) remolding. Hydrogel dressings serve as temporary barriers protecting injured tissue from exogenous infections while providing an advantageous microenvironment for cellular regeneration. However, traditionally molded hydrogels through catalyzed or triggered crosslinking into fixed size and strength prior to treatment struggle to integrate tightly with irregular wound surfaces, leading to dressing detachment a

Spontaneous formation of protein microcapsules using water-in-water emulsions stabilized by protein microgels.

Protein microgels tend to adsorb at the surface of dispersed droplets in water-in-water (W/W) emulsions. It was hypothesized that suspensions of stable protein microcapsules (MC) can be produced by spontaneous crosslinking of the microgels at the interface in a process akin so-called cold gelation. Rapeseed or whey protein microgels added to W/W emulsions formed by mixing polyethylene oxide (PEO) and Dextran (DEX) adsorbed at the W/W interface. The rapeseed protein microgels were found to spontaneously crosslink and formed MC suspensions that were stable to dilution and washing. The size of th

Overcoming biological inertness: multifaceted strategies to optimize PEEK bioactivity for interdisciplinary clinical applications.

Polyether ether ketone (PEEK), characterized by a comparable elastic modulus to human bone with high wear resistance, radiolucency, and biocompatibility, demonstrates considerable promise for clinical applications. However, due to the significant limitations in clinical applications caused by the biological inertness of PEEK, it should first be modified to meet clinical needs. Currently, the field of PEEK modifications is rapidly advancing, with a particular emphasis on enhancing its biological properties. Most of the previous reviews have separately discussed strategies like antibacterial, os

Conjugated crown ether lipid liposomes: enhancing integrity in serum through novel surface modifications.

This study explores the potential of 18-crown-6 ether to enhance liposomal integrity in biological media. A novel 1,2-Dipalmitoyl-sn-glycero-3-phosphoethanolamine (DPPE)-crown ether conjugate was synthesized and incorporated into phosphatidylcholine/cholesterol liposomes at varying concentrations (0.5-8 mol%). The physicochemical characteristics, serum integrity, and response to divalent cations were systematically evaluated. Although, these crown ether-modified liposomes do not provide the same level of protection as PEGylation at 48 hours, they clearly exhibited improved integrity in serum a

Formulation methods for peptide-modified lipid nanoparticles.

Lipid nanoparticles (LNPs) are a promising non-viral gene carrier, but one significant unmet challenge is cell-specific delivery in extrahepatic organs. Peptides are one class of targeting ligand that have been used to target nanoparticles, including LNPs. Herein, we compared two formulation approaches that use the polyethylene glycol (PEG)-lipid to display targeting peptides: (1) post-conjugation targeted (PCT), in which LNPs were formulated with PEG-lipid with chemical handles and subsequently modified with peptides, and (2) in-line targeted (ILT), in which peptide-PEG-lipid conjugates were

A coarse-grained model for disordered proteins under crowded conditions.

Macromolecular crowding may strongly affect the dynamics and function of proteins, with intrinsically disordered proteins being particularly sensitive to their crowded environment. To understand the influences of crowding on chain compaction and phase separation (PS) behavior of disordered proteins, both experiments with synthetic crowders-like polyethylene glycol (PEG) and ficoll-and theoretical models and molecular simulation approaches have been applied. Here, we developed a residue-based coarse-grained model for PEG that is compatible with the protein CALVADOS model. To achieve this, we op

Engineering Bonding Types and Macromeric Cross-Linkers in the Network To Modulate the Structures and Properties of Polydextran/Chitosan Hybrid Hydrogels.

Hybrid hydrogels are versatile biomaterials, and cross-linkers are the key for tailoring their characteristics. However, the bonding types in the network and the hydrophobic chain lengths of cross-linkers remain underexplored. This study introduces formylbenzoic acid-modified PEG (PEG-FA), which further reacts with adipic acid (AA), sebacic acid (SA), and tetradecanedioic acid (TDA) to create cross-linkers with varying hydrophobic segments. PEG-FA and its derivatives are used as aldehyde-functionalized macromeric cross-linkers to react with polydextran hydrazide (PDH) and quaternized chitosan

Enriching Subcellular Proteins in Leptospira Using a Triton X-114-Based Fractionation Approach.

Prokaryotes lack precise subcellular organelles but exhibit distinct regions such as cytoplasm, inner membrane, periplasm, and outer membrane, where most biochemical and physiological functions are organized. Thus, understanding the functional characteristics of proteins necessitates elucidating their subcellular localization. However, extracting subcellular proteins from gram-negative bacteria poses challenges due to their complex phospholipid bilayer. Although Triton X-114 has shown promise in outer membrane protein (OMP) extraction, a concise protocol remains elusive. This protocol demonstr
