Triple-negative breast cancer (TNBC) is highly malignant with limited effective targeted therapies, leading to poor patient outcomes. Although immune checkpoint inhibitors (ICIs) have shown promise in cancer treatment, most patients with TNBC exhibit primary or secondary resistance to ICIs, restricting their clinical use. Interferon-induced transmembrane protein 3 (IFITM3), an interferon-induced protein, is highly overexpressed in various tumors and is associated with immune suppression; however, its role in TNBC progression and immune escape remains unclear. In this study, we investigated IFI

Acute ischemic stroke is often complicated by infections and extracerebral organ dysfunction, which compromise the benefits of reperfusion therapy and independently predict poor outcomes including disability and death. Accumulating evidence indicates that these complications arise from the disruption of the bidirectional central-peripheral immune communication network, rather than being isolated organ-specific events. As the earliest and most abundant innate responders to acute ischemic stroke, neutrophils act as pivotal integrators in this intersystem crosstalk, orchestrating cellular, molecu

BACKGROUND: Infections are frequent in critically ill patients with earthquake-associated crush injury (CI) and contribute to poor outcomes, yet the immune pathways underlying posttrauma susceptibility remain poorly defined. We investigated longitudinal changes in damage-associated molecular pattern (DAMP)/PAMP sensing and downstream inflammatory programs in CI patients requiring intensive care.
METHODS: Nineteen CI patients admitted to the ICU after the February 6, 2023 Kahramanmaraş earthquake and 18 age- and sex-matched healthy controls were enrolled. Peripheral blood was collected on hosp

The gut microbiota, acting as a critical extrinsic endocrine organ, is profoundly involved in the pathological evolution and therapeutic response of hormone-dependent malignancies. This review elucidates the core mechanisms governing the microbiota, endocrine, and immune triple-axis. Multi-omic and biochemical evidence demonstrates that microbial metabolic networks, comprising the estrobolome, androbolome, and progestobolome/corticobolome, rely on enzymatic systems such as β-glucuronidases (GUS) and steroid-17,20-desmolases to execute hormone deconjugation and structural modification, thereby

Lupus nephritis (LN) is a kidney injury caused by systemic lupus erythematosus (SLE) and can lead to serious impairment of renal function. Glucocorticoid (GC) combined with cyclophosphamide (CTX) is currently a commonly used treatment for LN; however, it is associated with several limitations, including a high proportion of refractory cases, a high recurrence rate after remission, and a long treatment cycle. The purpose of this study was to evaluate the safety and efficacy of Tacrolimus (Tac) combined with GC in the treatment of lupus nephritis. This retrospective cohort study included 112 ped

BACKGROUND: Immune thrombocytopenia (ITP) and antiphospholipid syndrome (APS) are autoimmune disorders that may coexist, creating a challenging clinical scenario characterised by bleeding risk alongside paradoxically increased thrombotic risk. Catastrophic antiphospholipid syndrome (CAPS) is a rare but life-threatening manifestation of APS. The occurrence of CAPS in a patient with ITP receiving thrombopoietin receptor agonist therapy is uncommon and highlights important diagnostic and therapeutic challenges, particularly in the presence of concurrent bleeding manifestations.

Myopathies are a heterogeneous group of skeletal muscle disorders caused by genetic mutations or acquired insults, including inflammation, infection, endocrine imbalance, and toxic exposure. Myopathies affect a substantial number of individuals worldwide and are a significant cause of chronic muscle weakness and disability. Despite diverse etiologies, progressive myofiber injury and degeneration underlie the functional decline across disease subtypes. Accumulating evidence indicates that innate immune activation and regulated myofiber death pathways, including apoptosis, necroptosis, and pyrop

BACKGROUND: Immune checkpoint inhibitors (ICIs) are widely used in cancer therapy but may cause cardiovascular immune-related adverse events (irAEs) with substantial morbidity and mortality. The impact of biological sex on ICI-associated cardiotoxicity remains incompletely defined in real-world populations.
OBJECTIVES: To evaluate sex-based differences in cardiovascular immune-related adverse events following immune checkpoint inhibitor therapy.
METHODS: We conducted a retrospective, propensity score-matched cohort study using the TriNetX global research network. Adults initiating ICI therap

Deoxynivalenol (DON), a Fusarium-derived mycotoxin, disrupts immune homeostasis and induces immunotoxicity. Macrophage extracellular traps (METs) are key effectors in innate immunity and inflammation, yet whether DON triggers METs formation and the underlying mechanisms remain unclear. This study aims to investigate the molecular mechanism by which DON induces METs generation through circadian rhythm immune checkpoints involving the PER2-c-Myc axis. Using RAW264.7 and THP-1 macrophage models, we combined siRNA-mediated Per2 knockdown, and pharmacological inhibitors to dissect the mechanisms of

The discovery of Tertiary Lymphoid Structures (TLS) within tumors has reshaped our understanding of cancer immunity. Unlike the classical view that immune responses are solely initiated in lymph nodes, TLS, ectopic lymphoid aggregates resembling secondary lymphoid organs, can form in the tumor microenvironment (TME). These structures contain T cells, B cells, dendritic cells (DC) presenting antigenic peptides to T cells in the T cell zone of TLS, and follicular dendritic cells (FDC) which are stromal cells involved in the formation of germinal centers (GCs) and presenting antigens, under the f

Immunoglobulin T (IgT) is a class of Ig unique to teleost, with current research largely supporting its role in mucosal immunity. However, the precise function of IgT in many kinds of teleost remains poorly understood due to a lack of appropriate antibody tools. In this study, we prepared the recombinant protein of Oreochromis niloticus IgT heavy chain (rOn-IgT) and generated a monoclonal antibody (mAb). Through enzyme-linked immunosorbent assay (ELISA) and flow cytometry screening, the 1B3H4 hybridoma was identified as producing an IgG1-type antibody, which can recognize IgT and identify IgT

Helicobacter pylori is a gram-negative bacterium that persistently infects the human stomach, leading to peptic ulcers, gastritis, and an increased risk of gastric cancer. The extremophilic characteristics of this bacterium make it resistant to current drug treatments, and there are no licensed vaccines available against H. pylori. Computational approaches offer a viable alternative for designing antigenic, stable, and safe vaccines to control infections caused by this pathogen. In this study, we employed an immunoinformatic strategy to design a set of candidate multi-epitope subunit vaccines

The immune response to malaria vaccines is generally stronger in malaria-naive individuals than in those with lifelong exposure. The immunological basis for this is unclear. IgM, total IgG, and IgG subclass (IgG1, IgG2, IgG3, and IgG4) antibody responses against 21 pre-erythrocytic and erythrocytic Plasmodium falciparum proteins before and after controlled human malaria infection (CHMI) via the direct venous inoculation of 3,200 P. falciparum sporozoites (PfSPZ) were compared in three groups of volunteers: 1) malaria-naïve (n = 22); 2) malaria-naïve immunized with a PfSPZ chemoattenuated vacci

Blindness or vision loss due to neuroretinal and photoreceptor degeneration affects millions of individuals worldwide. In numerous neurodegenerative diseases, including age-related macular degeneration, dysregulated immune response-mediated retinal degeneration has been found to play a critical role in the disease pathogenesis. To better understand the pathogenic mechanisms underlying the retinal degeneration, we used a mouse model of systemic immune activation where we infected mice with lymphocytic choriomeningitis virus (LCMV) clone 13. Here, we evaluated the effects of LCMV infection and p

BACKGROUND: Prader-Willi syndrome (PWS) is a rare genetic neurodevelopmental syndrome with highly increased risk of obesity and cardiovascular disease (CVD). Recent evidence suggests that inflammation is implicated in the pathogenesis. Here we investigated CVD related immune markers to shed light on pathogenetic mechanisms.
METHODS: We performed a cross-sectional study with 22 participants with PWS and 22 healthy controls (HC), and compared levels of 21 inflammatory markers that reflect activity in different aspects of CVD related immune pathways and analyzed their association with clinical C

BACKGROUND: In-situ tumor ablation provides the immune system with the appropriate antigens to induce anti-tumor immunity. Here, we present an innovative technique for generating anti-tumor immunity by delivering exogenous ultra-high concentration (> 10,000 ppm) gaseous nitric oxide (UHCgNO) intratumorally.
METHODS: The capability of UHCgNO to induce apoptosis was tested in vitro in mouse colon (CT26), breast (4T1) and Lewis lung carcinoma (LLC-1) cancer cell lines. In vivo, UHCgNO was studied by treating CT26 tumor-bearing mice in-situ and assessing the immune response using a Challenge assa

The RIG-I pathway can be activated by RNA containing 5' triphosphate, leading to type I interferon release and immune activation. Hence, RIG-I agonists have been used to induce immune responses against cancer as potential immunotherapy. However, delivery of 5' triphosphorylated RNA molecules as RIG-I agonists to tumour cells in vivo is challenging due to the susceptibility of these molecules to degradation. In this study, we demonstrate the use of extracellular vesicles (EVs) from red blood cells (RBCs), which are highly amenable for RNA loading and taken up robustly by cancer cells, for RIG-I
