Evaluation of a direct ELISA for the serodiagnosis of Oestrus ovis infections in sheep.

Oestrosis is a parasitic disease of sheep and goats caused by the nasal bot fly Oestrus ovis. In the United Kingdom the economic losses as a result of infestation can be considered negligible, but the differentiation of O ovis cases from more serious diseases such as listeriosis, gid and sheep scab is of considerable importance. Currently, diagnosis of oestrosis relies on the subjective observation of clinical signs or the demonstration of larvae postmortem. This paper assesses the effectiveness of a direct enzyme-linked immunosorbent assay (ELISA) using a crude somatic antigen from first-stag…

Experimental infection of Culex (Culex) quinquefasciatus and Aedes (Stegomyia) aegypti with Wuchereria bancrofti.

A study was conducted to determine the susceptibility of local strains of Culex quinquefasciatus and Aedes aegypti to infection with the str … uals. W. bancrofti parasites underwent normal development in Cx. quinquefasciatus, with third stage larvae first being recorded on the 11th day post infection, and their numbers increasing thereafter. Development of filariae in Ae. aegypti did not proceed beyond the first larval stage, and there was a progressively increasing number of non-viable larvae …

Bacteriological investigations on wound myiasis of sheep caused by Wohlfahrtia magnifica (Diptera: Sarcophagidae).

The aim of the present investigation was to get further information about obligate aerobic and facultatively anaerobic bacterial communities … bacteria was higher than 80% in the sample taken from a lesion containing third instar Wohlfahrtia larvae and in the wound discharge collected from a vulval wound free of maggots. It is suggested that there is a shift in the composition of the bacterial communities of vulva as staphylococci disappear from the wounds due to the presence of Wohlfahrtia l …

Field evaluation of biodegradable controlled release formulation of fenthion against Mansonia mosquitos.

Controlled release monolithic formulation of fenthion, an organophosphorus mosquito larvicide, prepared by the physical entrapment of the active agent into biodegradable polymer matrices was evaluated against Mansonia mosquito larvae breeding in hydrophytes infested coconut husk retting ponds. Field evaluation was carried out at two application rates viz, 2.5 and 1.0 ppm. Absolute control of larval breeding was observed for 25 and 17 weeks and over 50% reduction in breeding density was observed for another 3 and 2 weeks for the respective application rates of 2.5 and 1.0 ppm. The bioassay carr…

Polymerase chain reaction-identification of Dirofilaria repens and Dirofilaria immitis.

On the basis of known DNA sequences of Dirofilaria repens and D. immitis we designed specific primers for the amplification by Polymerase Chain Reaction (PCR) of the DNA from the two species. The PCR-based identification was found to be unambiguous and allowed specific diagnosis of microfilariae in blood samples, of developing larvae in the mosquito vector and of immature adults in bioptic material, overcoming the serious constraints of the morphological separation of these filarial parasites at the pre-adult stages. The technique was found to be very sensitive and applicable to samples stored…

Parasitic females of Strongyloides papillosus as a pathogenetic stage for sudden cardiac death in infected lambs.

The present study was aimed at elucidating the responsibility of parasitic females for sudden cardiac death following Strongyloides papillosus infection in calves. A preliminary experiment demonstrated a percutaneous infection with S. papillosus infective larvae to cause sudden cardiac death in lambs as in calves, indicating lambs could serve as a model to study fatal stronglyloidiasis in calves. Parasitic females of S. papillosus were inoculated into the duodenum of lambs. Lambs given live worms developed continuous sinus tachycardia immediately after inoculation, and died of sudden cardiac a…

[An estimate of the potentials for eliminating aqueous biocenoses relative to free-living life forms of Opisthorchis felineus].

Different methods of estimating the elimination potential of invertebrate hydrobionts, members of water biocenoses, in relation to freeliving instars such as egg (miracidium) and cercarium of O. felineus are considered. Calculatons and conclusions are based on field and laboratory investigation carried out in the period 1985-1992. The elimination process in biocenoses does not practically influence on the intensivity of infection, but influences significantly on the extensivity of the infection of the molluscs Codiella with O. felineus larvae. This process is also the cause of low extensivity …

The dynamics of trickle infections with Ancylostoma ceylanicum in inbred hamsters.

Trickle infections with a hamster-adapted strain of the hookworm Ancylostoma ceylanicum were studied by administering doses of 5-30 larvae twice weekly to inbred DSN hamsters. The worm burdens were regulated at very low levels, and this was found to be independent of the size of the infective dose. It is likely that there is some turnover of adult worms during trickle infection, as larvae of all stages were recovered from most time-points. This regulation is immunologically mediated and both the worm burdens and fecundity were increased in hamsters that were immunosuppressed by cortisone treat…

Morphological demonstration of essential functional changes after in vitro and in vivo transition of infective Onchocerca volvulus to the post-infective stage.

Transmission electron microscopic investigation of the morphogenesis of Onchocerca volvulus through the third moult to the post-infective stage revealed essential alterations in various larval organs. Complete rebuilding of surface structures, the reduction of secretory granules in the glandular oesophagus, the unfolding of the intestine, an increase in the number of nerve fibres in the nerve ring and novel sensory papillae were significant findings. Transition from third- to fourth-stage larvae (L4) started as early as 48 h after transfer to vertebrate conditions in vivo in surrogate hosts an…

Characterization of a noncyst-forming isolate of Trichinella from a wild boar in Yugoslavia.

An isolate of Trichinella obtained from a wild boar in Yugoslavia did not form cysts in the musculature of its natural host. Subsequent inoculation into experimental hosts demonstrated that some larvae became encysted only after extended time periods, whereas others remained unencapsulated. Histological staining of larvae in the musculature demonstrated no deposition of collagen typically seen for Trichinella spiralis spiralis, Trichinella spiralis nativa, or Trichinella spiralis nelsoni. The Yugoslavian isolate, given the name of Zagreb isolate after the University where it was first studied,…

Kinetics of the pulmonary phase of Schistosoma mansoni in mice treated with dexamethasone.

In the experimental schistosomiasis mansoni glucocorticoids cause a reduction in the worm burden when administered in the week of infection … recovery. Analysis of the results showed that the non-treated mice presented larger numbers of lung larvae than the treated ones, and this difference was also found later in the worm burden in the portal system. This difference may reflect the early death of larvae in treated animals, before or after reaching the lungs.

Destructive ocular myiasis in a noncompromised host.

A case of destructive ocular myiasis resulting in complete loss of the globe in two days time is documented. To the best of our knowledge this is the first report of such a severe involvement in a healthy and non-compromised host. Mechanical removal and good local hygiene helped heal the wound. The larvae were isolated to be that of Chrysomyia bezziana (screwworm fly). This is possibly the first report of destructive ocular myiasis caused by Chrysomyia bezziana from the Indian subcontinent and the second in world literature. Infestation of human eyes with larvae of flies (myiasis) has been rep…

Survival of adults and development stages of Sarcoptes scabiei var. canis when off the host.

All life-stages of Sarcoptes scabiei var. canis survive in the hosts' environment for several days to several weeks depending on r.h. and temperature. Survival of larvae was comparable to males; survival of nymphs was comparable to females. Females and nymphs generally survived longer than larvae and males. Low temperature (10-15 degrees) and high r.h. prolonged survival of all life stages. At 10-15 degrees C, females and nymphs survived 1-3 weeks at 97% r.h., 1-2 weeks at 75% r.h. and 5-8 days at 45% r.h. At 20-25 degrees C, survival was significantly reduced but all life-stages survived at l…

Lectin binding to larval stages of Brugia patei.

Surface properties of microfilariae (mf) and infective larvae of Brugia patei were investigated to compare them to previous studies with the other brugian species. Of all the lectins tested, only wheat germ agglutinin (WGA) binds to the sheath surface of mf indicating the presence of N-acetyl-D-glucosamine as a major surface carbohydrate. However, cuticle of infective larvae failed to show binding of these lectins. Enzyme treatment of mf with N-acetyl-D-glucosaminidase and L-fucosidase has exposed D-mannose, N-acetyl-D-galactosamine and L-fucose on the sheath surface. The binding of lectins to…

A genetic assay to detect chromosome gain and/or loss in somatic cells of Drosophila melanogaster.

A genetic short-term test is described that allows (i) detection and (ii) quantitative evaluation of aneuploidy induced in somatic cells of Drosophila melanogaster. In this somatic aneuploidy test (SAT) larvae of the genotype z w-/w+J Y are exposed to the test compound. Gain and/or loss of the w+J Y chromosome leads to the formation of aneuploid daughter cells: z w-/w/J Y/w+J Y and z w-/O, respectively. These cells are fully viable, proliferate and, when they are part of an eye primordium, form a yellow//white twin spot on the otherwise red background after metamorphosis. The number of eyes sc…

After the development of the larvae has been made into the capsule, this one ends in breaking, generally when the bark beetle has attacked a new host-tree. Then, the larvae of the nematode leave the insect and invade the galleries. The parasitic development of C. diplogaster always t …
