Lipid-coated microbubbles are widely used as ultrasound contrast agents (UCAs) and are being developed as carriers for drug and gene delivery. These microbubbles typically consist of an inert gas core and a stabilizing monolayer shell of phospholipid and a PEGylated emulsifier. In practice, a 9:1 M ratio of DSPC (a saturated phospholipid) to PEG-40-stearate (PEG40St) is conventionally used, under a long-standing assumption that the final composition of the microbubble shell is identical to the initial mixture composition. In this study, we tested that assumption over a wide range of DSPC/PEG40

BACKGROUND AND AIMS: Sympathetic overactivation plays a critical role in the pathophysiology of various conditions, such as arterial hypertension, chronic kidney disease, coronary artery disease (CAD), diabetes, metabolic syndrome, and dyslipidemia. Initially developed for hypertension management, renal denervation (RDN) has also been associated with metabolic improvements. Preclinical studies in rodent models suggest that RDN may improve lipid profiles by reducing sympathetic activity. This study analyses the effect of RDN on lipid profiles in hypertensive patients with or without CAD.

During fertilization, sperm and egg membranes signal and fuse to form a zygote and begin embryonic development. As lipids participate in signaling and membrane fusion, we investigated the role of lipid asymmetry in gametogenesis, fertilization, and embryogenesis. We show that the lipid flippase TAT-5, an essential P4-ATPase that maintains phosphatidylethanolamine asymmetry, is required for both oocyte formation and sperm activation, albeit at different levels of flippase activity. Loss of TAT-5 significantly decreases fertility in both males and hermaphrodites and decreases sperm activation. T…

Traumatic brain injury (TBI) represents a major cause of mortality and disability worldwide, particularly affecting young adults and elderly populations. This study investigates the neuroprotective potential of orlistat (ORL), a gastrointestinal lipase inhibitor, in a murine TBI model. Behavioral, histological, and molecular analyses demonstrated that ORL significantly attenuated TBI-induced neurological damage. Microglial depletion experiments revealed that ORL's neuroprotective effects were largely mediated through microglial modulation. In vitro and in vivo studies showed that ORL suppresse

Neurodegenerative and cardiovascular disorders share multifactorial origins, including oxidative stress, (neuro)inflammation, and lipid dysregulation-factors often addressed independently by single-target therapies. In this study, we report a rational multitarget approach through the design and synthesis of novel (benzo)thiazine derivatives that integrate antioxidant, anti-inflammatory, and antihyperlipidemic functionalities within a single molecular framework. The compounds were obtained in good yields via 3-7 step synthetic routes and evaluated through complementary in vitro and in vivo assa

This study aimed to elucidate the effect of extraction solvents on the lipid profiles and volatile compounds in large yellow croaker roe phospholipids (LYPLs) with chloroform/methanol (CM), ethanol/ethanol (EE), and ethanol/hexane (EH). The results demonstrated that CM-extracted LYPLs exhibited the lowest oxidation levels, while EE-extracted LYPLs contained the highest polyunsaturated fatty acids (PUFAs) content. EH extraction was more effective for monounsaturated fatty acids (MUFAs) in LYPLs but was less effective for polar lipids. Across solvents, 410 lipids were identified, with phosphatid…

BACKGROUND: Atherosclerosis (AS), characterized by lipid accumulation, contributes significantly to global cardiovascular morbidity. Ferroptosis, an iron-dependent form of cell death triggered by lipid peroxidation, is emerging as a critical player in AS progression. Therefore, our study seeks to elucidate the intricate mechanisms of ferroptosis within the lipid metabolism pathway in AS.
METHODS: Differentially expressed genes were identified from the GSE100927 dataset, subsequently isolating AS lipid metabolism-related ferroptosis genes (ASLMRFeGs). Unsupervised cluster analysis was performe

The health impacts of ozone (O3) remain vastly underestimated, particularly regarding the transgenerational effects of prenatal O3 exposure on offspring hepatic lipid metabolism, which has rarely been investigated. This study established a dose-dependent prenatal O3 exposure mouse model (0, 0.5, and 1.0 ppm) combined with transcriptomic sequencing and lipidomic profiling to systematically evaluate hepatic metabolic dysregulation in adult offspring. Results revealed that high-dose O3 exposure significantly elevated hepatic total cholesterol (TC) levels in female offspring, with no analogous eff

BACKGROUND: Atypical antipsychotics (AAs) are commonly used in the treatment of schizophrenia and are often preferred as first-line therapy over typical antipsychotics (TAs) due to their lower risk of extrapyramidal side effects. Both groups are efficacious in treating symptoms of schizophrenia, but increasing research has highlighted AAs as being associated with a risk of developing dyslipidaemia. Existing research has pointed to the need for more data focusing on the effects of individual AAs on dyslipidaemia in this population.
METHODS: The scoping review was conducted using the Preferred

Fenofibrate, a peroxisome proliferator-activated receptor α (PPARα) agonist, is widely prescribed to treat hyperlipidemia and has therapeutic potential in liver and kidney diseases. However, fenofibrate is also associated with adverse effects, including elevated creatinine and liver and kidney toxicity, although the underlying mechanisms remain unclear. In addition, how fenofibrate regulates lipid metabolism differently in the liver and kidney is not well understood. Therefore, in this study, we investigated the dose-dependent effects of fenofibrate on liver and kidney metabolism in rats, with

BACKGROUND: Felodipine (FLD) is an L-type calcium channel blocker with pronounced neuroprotection against Alzheimer's disease (AD). Unfortunately, the efficacy of FLD has been impeded by limited solubility, poor bioavailability, and sub-optimal accumulation. Thus, the current study unfolds the potential of nanostructured lipid carriers based on in-situ gel of FLD (FLD-NLCs gel) to ameliorate dementia.
METHODS: The FLD-contained NLCs were prepared using the microemulsion-sonication method and further integrated into thermosensitive gel comprised poloxamer 407 and HPMC K4M. The formulation was

Bridge-like lipid transfer proteins (BLTPs) constitute a superfamily of proteins localized at various intracellular membrane contact sites (MCSs). Members of this family have been implicated in human neurological disorders. Among them, the BLTP Atg2 is essential for the expansion of isolation membranes in macroautophagy. In this study, we demonstrate that another BLTP, Vps13, is involved in microautophagy in budding yeast. Nucleophagy-the autophagic degradation of nuclear components-is crucial for maintaining nuclear proteostasis, and its dysfunction has been linked to neurodegenerative diseas

BACKGROUND: New research points to a complicated relationship between gallstone disease (GSD), thyroid function, and lipid profiles. To better understand how to prevent and treat GSD, this study will look at how common thyroid abnormalities are among people with the diagnosis, as well as any dyslipidemias that may be present.
MATERIAL AND METHODS: This is a cross-sectional observational study worked out at a tertiary care hospital in southern India's Department of General Surgery. We enrolled 60 persons with a GSD diagnosis. We divided patients with only gallstones and gallstones with common

Ferroptosis, a form of cell death instigated by iron-dependent lipid peroxidation reactions (LPO), is emerging as a promising therapeutic target for cancer. While the mechanisms governing LPO induction and suppression have gradually been unveiled, questions persist regarding the specific cellular location of LPO and the utilization of iron in driving cell death. A comprehensive understanding of these aspects holds significant potential for advancing therapeutic applications in disease management. Here, we show lysosomal LPO in the initiation of ferroptosis, leveraging the hidden abilities of f

Intravenous lipid emulsions (ILE) were first proposed in 1998 as a treatment for bupivacaine-induced cardiac arrest. Since then, their use has expanded to include poisonings by various lipophilic drugs such as tricyclic antidepressants, calcium channel blockers, and antipsychotics. This 2025 narrative review explores the evolving pathophysiological mechanisms of ILE therapy, including the lipid sink and lipid shuttle theories, as well as non-scavenging cardiotonic effects such as membrane stabilization, mitochondrial support, and modulation of vascular tone. It summarizes recent findings from

The development of reusable biosensors based on tethered bilayer lipid membranes (tBLMs) is crucial to achieve cost-effective and sustainable pathogen detection. In this study, we investigated the regeneration of tBLMs assembled on fluorine-doped tin oxide (FTO) substrates using organic silane-based molecular anchors and a lipid mixture of dioleoylphosphatidylcholine and cholesterol. The sensors were exposed to α-hemolysin (αHL), a pore-forming toxin from Staphylococcus aureus, and regenerated by a two-step bilayer removal protocol. Electrochemical impedance spectroscopy (EIS) was used to asse
