KLF13 restrains Dll4-muscular Notch2 axis to improve the muscle atrophy.

BACKGROUND: Muscle atrophy can cause muscle dysfunction and weakness. Krüppel-like factor 13 (KLF13), a central regulator of cellular energy metabolism, is highly expressed in skeletal muscles and implicated in the pathogenesis of several diseases. This study investigated the role of KLF13 in muscle atrophy, which could be a novel therapeutic target. METHODS: The effects of gene knockdown and pharmacological targeting of KLF13 on skeletal muscle atrophy were investigated using cell-based and animal models. Clofoctol, an antibiotic and KLF13 agonist, was also investigated as a candidate for re

Electrofluidic fiber muscles.

Actuators are to robots what muscles are to humans. They enable motion and determine strength and dexterity. The fiber form factor makes skeletal muscles modular, scalable, and densely integrated (50% of human body weight). In contrast, servo motors that drive today's robots lack the flexibility and modularity of muscle fibers, limiting integration and dexterity. Here, we report electrofluidic fiber muscles, soft artificial muscles for robotic applications with power density comparable to skeletal muscles (50 watts per kilogram), contraction strains of 20%, and response time of 0.3 second. The

Older mice show decreased regeneration of neuromuscular junctions following lengthening contraction-induced injury.

Progressive muscle atrophy and loss of muscle strength associated with old age have been well documented. Although age-associated impairments in skeletal muscle regeneration following injury have been demonstrated, less is known about whether aging impacts the regenerative response of neuromuscular junctions (NMJ) following contraction-induced injury. Reduced ability of NMJs to regenerate could lead to increased numbers of denervated muscle fibers and therefore play a contributing role to age-related sarcopenia. To investigate the relationship between age and NMJ regeneration following injury,

Research progress on the correlation between estrogen and estrogen receptor on postmenopausal sarcopenia.

Estrogen is a necessary sex steroid and potent neuroprotective hormone. It plays a multifaceted role beyond the reproductive system, extending its influence to the brain, skeletal muscle, and other organs. Estrogen's role in cognition, mood, autonomic regulation, and neuroprotection involves interactions with neurotransmitters, neuromodulators in a distributed manner. Notably, the impact of estrogen on mitochondrial metabolism in skeletal muscle is particularly significant due to a unique modulated bioenergetic profiles, synaptic plasticity, and neuronal health. The deficiency of estrogen in m

miR-196-5p regulates myogenesis and induces slow-switch fibers formation by targeting PBX1.

Skeletal muscle, which is crucial for meat production, color, and quality is regulated by complex genetic mechanisms. MicroRNAs (miRNA), serve a crucial part in regulating skeletal muscle myogenesis together with the switching of muscle fiber types, but the identification of key miRNAs and their underlying molecular mechanism has been hindered. In the present study, miRNA sequencing was utilized to identify the differentially expressed miRNAs (DEMs) in different skeletal muscles, among which miR-196-5p was found notably upregulated in chicken soleus (SOL) muscles, suggesting the potential role

MuRF1/TRIM63, Master Regulator of Muscle Mass.

The E3 ubiquitin ligase MuRF1/TRIM63 was identified 20 years ago and suspected to play important roles during skeletal muscle atrophy. Since then, numerous studies have been conducted to decipher the roles, molecular mechanisms and regulation of this enzyme. This revealed that MuRF1 is an important player in the skeletal muscle atrophy process occurring during catabolic states, making MuRF1 a prime candidate for pharmacological treatments against muscle wasting. Indeed, muscle wasting is an associated event of several diseases (e.g., cancer, sepsis, diabetes, renal failure, etc.) and negativel

Inferring position of motor units from high-density surface EMG.

The spatial distribution of muscle fibre activity is of interest in guiding therapy and assessing recovery of motor function following injuries of the peripheral or central nervous system. This paper presents a new method for stable estimation of motor unit territory centres from high-density surface electromyography (HDsEMG). This completely automatic process applies principal component compression and a rotatable Gaussian surface fit to motor unit action potential (MUAP) distributions to map the spatial distribution of motor unit activity. Each estimated position corresponds to the signal ce

Muscle-specific atrophy of the lower limb musculature in response to simulated microgravity exposure in women.

Despite the increased involvement of women in spaceflight missions, the efforts to understand changes in skeletal muscle health in women with microgravity exposure have been limited. The goal of the current investigation was to expand the limited knowledge on skeletal muscle atrophy responses to microgravity in women. Eight women (34 ± 4 yr) underwent 60 days of simulated microgravity (6° head-down tilt bedrest), and changes in MRI-determined skeletal muscle volume of 17 lower limb muscles were determined after 1 and 2 mo. Muscle volume decreased (P < 0.05) in all 17 muscles of the women after

Gut bacteria are critical for optimal muscle function: a potential link with glucose homeostasis.

Gut microbiota is involved in the development of several chronic diseases, including diabetes, obesity, and cancer, through its interactions with the host organs. It has been suggested that the cross talk between gut microbiota and skeletal muscle plays a role in different pathological conditions, such as intestinal chronic inflammation and cachexia. However, it remains unclear whether gut microbiota directly influences skeletal muscle function. In this work, we studied the impact of gut microbiota modulation on mice skeletal muscle function and investigated the underlying mechanisms. We deter

Physiological determinants of mechanical efficiency during advanced ageing and disuse.

This study aimed to determine which physiological factors impact net efficiency (ηnet) in oldest-old individuals at different stages of skeletal muscle disuse. To this aim, we examined ηnet, central haemodynamics, peripheral circulation, and peripheral factors (skeletal muscle fibre type, capillarization and concentration of mitochondrial DNA [mtDNA]). Twelve young (YG; 25 ± 2 years), 12 oldest-old mobile (OM; 87 ± 3 years), and 12 oldest-old immobile (OI; 88 ± 4 years) subjects performed dynamic knee extensor (KE) and elbow flexors (EF) exercise. Denervation alters the secretome of myofibers and thereby affects muscle stem cell lineage progression and functionality.

Skeletal muscle function crucially depends on innervation while repair of skeletal muscle relies on resident muscle stem cells (MuSCs). However, it is poorly understood how innervation affects MuSC properties and thereby regeneration of skeletal muscle. Here, we report that loss of innervation causes precocious activation of MuSCs concomitant with the expression of markers of myogenic differentiation. This aberrant activation of MuSCs after loss of innervation is accompanied by profound alterations on the mRNA and protein level. Combination of muscle injury with loss of innervation results in

Muscle segmentation in axial computed tomography (CT) images at the lumbar (L3) and thoracic (T4) levels for body composition analysis.

In diseases such as cancer, patients suffer from degenerative loss of skeletal muscle (cachexia). Muscle wasting and loss of muscle function/performance (sarcopenia) can also occur during advanced aging. Assessing skeletal muscle mass in sarcopenia and cachexia is therefore of clinical interest for risk stratification. In comparison with fat, body fluids and bone, quantifying the skeletal muscle mass is more challenging. Computed tomography (CT) is one of the gold standard techniques for cancer diagnostics and analysis of progression, and therefore a valuable source of imaging for in vivo quan

[Effects of ascorbic acid deficiency on lactate dehydrogenase. Quantitative and isoenzymatic study].

Light, acute vitamin C deficiency or repletion had no uniform effect on lactiodeshydrogenase, each organ reaching specifically (skeletal muscle, heart, kidney, spleen, liver adrenals, testes). There was no correlation with the age of the animal except in the case of testicular lacticodeshydrogenase isoenzymes. Reduction of food intake caracteristic of the late state of scurvy had no effect on the distribution of isoenzymes which was also independent of quantitative variations of enzyme activity. Vitamin C repletion restored the normal distribution of isoenzymes in spleen and liver but not in s

Exercise as an anti-inflammatory therapy for cancer cachexia: a focus on interleukin-6 regulation.

Cancer cachexia is a complicated disorder of extreme, progressive skeletal muscle wasting. It is directed by metabolic alterations and systemic inflammation dysregulation. Numerous studies have demonstrated that increased systemic inflammation promotes this type of cachexia and have suggested that cytokines are implicated in the skeletal muscle loss. Exercise is firmly established as an anti-inflammatory therapy that can attenuate or even reverse the process of muscle wasting in cancer cachexia. The interleukin IL-6 is generally considered to be a key player in the development of the microenvi

End-Stage Kidney Disease Following Reflux Nephropathy Since the 2011 AAP Guidelines.

Age-associated sarcopenia decreases mobility and is promoted by cell senescence, inflammation, and fibrosis. The mitochondrial enzyme arginase-II (Arg-II) plays a causal role in aging and age-associated diseases. Therefore, we aim to explore the role of Arg-II in age-associated decline of physical activity and skeletal muscle aging in a mouse model. Young (4-6 months) and old (20-24 months) wild-type (wt) mice and mice deficient in arg-ii (arg-ii-/-) of both sexes are investigated. We demonstrate a decreased physical performance of old wt mice, which is partially prevented in arg-ii-/- animals

Integrative single-cell RNA-seq and ATAC-seq analysis of myogenic differentiation in pig.

BACKGROUND: Skeletal muscle development is a multistep process whose understanding is central in a broad range of fields and applications, from the potential medical value to human society, to its economic value associated with improvement of agricultural animals. Skeletal muscle initiates in the somites, with muscle precursor cells generated in the dermomyotome and dermomyotome-derived myotome before muscle differentiation ensues, a developmentally regulated process that is well characterized in model organisms. However, the regulation of skeletal muscle ontogeny during embryonic development

Single-fiber morphometry and spatial transcriptomics reveal selective oxidative muscle fiber atrophy in non-metastatic breast cancer.

Cancer-related fatigue is the most common and persistent symptom in breast cancer, with fatigue reported up to 10 years post-diagnosis. Unlike many cancers, fatigue in breast cancer often arises during early-stage disease in the absence of cachexia. While many factors contribute to fatigue, the direct contribution of cancer-associated skeletal muscle pathology remains poorly understood. Here we analyzed pectoralis major muscle biopsies from individuals with non-metastatic breast cancer and non-cancer controls using single-fiber morphometry and spatial transcriptomics. We identified fiber-type-

Complement pathway activation mediates pancreatic cancer-induced muscle wasting and pathological remodeling.

Cancer cachexia is a multifactorial condition characterized by skeletal muscle wasting that impairs quality of life and longevity for many cancer patients. A greater understanding of the molecular etiology of this condition is needed for effective therapies to be developed. We performed a quantitative proteomic analysis of skeletal muscle from cachectic pancreatic ductal adenocarcinoma (PDAC) patients and non-cancer controls, followed by immunohistochemical analyses of muscle cross sections. These data provide evidence of a local inflammatory response in muscles of cachectic PDAC patients, inc
