MLL3 adaptor function, not methyltransferase catalytic activity, is essential for breast tumor suppression.

MLL3 (Mixed-Lineage Leukemia 3), also known as KMT2C, is one of the most frequently altered epigenetic regulators in breast cancer. MLL3 loss-of-function leads to accelerated tumor onset and growth and increased metastasis. As a large multi-domain protein, MLL3 functions as a histone methyltransferase and a nuclear protein adaptor interacting with other epigenetic proteins. Since breast cancer MLL3 mutations are often truncating mutations that lead to protein degradation, whether the MLL3 tumor suppressor activity depends on its catalytic activity or non-catalytic chromatin adaptor function re

Ex vivo bone marrow subniches influence the fate of human antibody-secreting cells.

Long-term humoral immunity relies on long-lived plasma cells in the bone marrow (BM). However, the processes governing plasma cell transport, positioning, and longevity within the BM niche remain poorly understood, especially in humans. Most existing knowledge comes from mouse studies or limited human-based models, which makes translating findings to human biology challenging. Here, we introduce a physiologically relevant human bone marrow-on-a-chip (hBMOC) model to investigate the behavior and interactions of human immune organoid-derived antibody-secreting cells (ASCs) within the human BM mi

Molecular Diagnosis to Individualized Therapies in Rare Genetic Diseases: New Approach Methodologies, RNA Therapeutics, and the Case for a Human-First Filter.

Rare genetic diseases are heterogeneous across mechanisms, trajectories, and treatment responses. To date, approved therapies remain available for only a small proportion of rare genetic diseases. Oligonucleotide-based RNA therapeutics, particularly antisense oligonucleotides (ASOs) and small interfering RNAs (siRNAs), offer a promising therapeutic avenue for rare genetic diseases with sequence-level precision. However, traditional preclinical paths may mis-predict human outcomes when disease biology diverges from animal models. New approach methodologies (NAMs), including patient-derived indu

Extracellular vesicles derived from cancer-associated fibroblasts facilitate NSCLC progression and metastasis via miR-664b-5p-mediated activation of the CXCL12/CXCR4 chemokine pathway.

Exosomal miRNAs mediate intracellular communication between the tumor microenvironment and cancer cells in non-small cell lung cancer (NSCLC). However, effects of exosomal miRNAs on NSCLC and its mechanisms have not been completely clarified. Here, exosome miRNA profiling and patient serum analysis revealed that the expression level of exosomal miR-664b-5p is significantly elevated in NSCLC, with particularly high levels in cancer-associated fibroblasts (CAFs) and their secreted exosomes. Functionally, miR-664b-5p promotes cell proliferation, migration, and invasion and inhibits apoptosis, the

Interleukin-18 Constrains the Intestinal Tuft Cell-Driven Responses During Helminth Infection.

Type 2 immunity at mucosal surfaces is essential for helminth clearance, tissue repair, and barrier maintenance, processes governed by a robust feed-forward circuit between chemosensory tuft cells and group 2 innate lymphoid cells (ILC2s). However, the regulatory mechanisms that calibrate the magnitude and duration of this epithelial immune circuit remain incompletely defined. Here, we identify interleukin-18 (IL-18), a cytokine traditionally associated with type 1 immunity, as a key regulator of tuft cell-mediated type 2 responses during Nippostrongylus brasiliensis (N. brasiliensis) infectio

Macrophage polarization in Sjögren's syndrome: from mechanisms to therapeutic translation.

Sjögren's syndrome (SS) is a chronic systemic autoimmune disease characterized by damage to exocrine glands, and no effective treatment is currently available to reverse glandular fibrosis. As core effector cells of innate immunity, macrophages play a pivotal role in the dynamic imbalance of polarization states, which critically influences inflammation initiation, tissue injury, and irreversible fibrosis in SS. This review systematically delineates the heterogeneity of M1/M2 macrophage subsets, their spatiotemporal dynamics, and their functional transitions across different pathological stages

The insulin-like growth factor 2 mRNA-binding protein 2 affects tumor cell metabolism via mitochondrial transporter activity and lipid alterations.

The insulin-like growth factor 2 mRNA-binding protein (IGF2BP) family is overexpressed in cancer and associated with poor prognosis. IGF2BP2 has been linked to single metabolic alterations by acting on its RNA targets. Here, we used a comprehensive approach to elucidate the effects of IGF2BP2 on primary and lipid metabolism. 13C-metabolic flux analysis (MFA) combined with RNA-Seq data revealed that IGF2BP2 affects mitochondrial fluxes by regulating the expression of several mitochondrial transporters, such as mitochondrial pyruvate carrier 1 (MPC1) and uncoupling protein 2 (UCP2). Methyl pyruv

Recent Progress in 2D, 3D, and On-a-Chip Models of the Placenta.

BACKGROUND: The placenta is a temporary organ that develops throughout pregnancy, connecting a developing fetus to the maternal uterine wall. The placenta's structure is species-specific and complex, necessitating the advancement of in vitro models to study this dynamic organ. The main cell types composing the placenta, trophoblast cells, serve several roles and have been incorporated within biomaterials and devices to recapitulate the placental microenvironment. SUMMARY: This review highlights in vitro 2D, 3D, and on-a-chip models of two placental interfaces: the interface between the endome

CSTF2 Supports Hypoxia Tolerance in Hepatocellular Carcinoma by Enabling m6A Modification Evasion of PGK1 to Enhance Glycolysis.

Cleavage stimulation factor subunit 2 (CSTF2) is a fundamental factor in the regulation of 3'-end cleavage and alternative polyadenylation of pre-mRNAs. Previous work has identified a tumor-promoting role of CSTF2, suggesting that it may represent a potential therapeutic target. In this study, we aimed to elucidate the mechanistic function of CSTF2 in hepatocellular carcinoma (HCC). CSTF2 upregulation was frequent in HCC, and elevated levels of CSTF2 correlated with poor patient prognosis. Although CSTF2 inhibition did not suppress HCC growth under nonstress conditions, it supported tolerance

In Vivo Efficacy of a Macrocyclic Peptoid-Peptide Hybrid That Selectively Modulates the Beta-Catenin/TCF Interaction to Inhibit Prostate Cancer.

BACKGROUND: Prostate cancer is the most common form of male cancer and can initially be treated as a localized disease. Although the 5-year survival rate at diagnosis approaches 100 percent, a subset of patients will subsequently develop resistance to treatment. This may ultimately lead to metastatic castration resistant prostate cancer (mCRPC), for which the prognosis is much less favorable. The importance of the Wnt/β-catenin pathway in treatment-resistant prostate cancer has inspired efforts to exploit the interaction of β-catenin with its transcription binding partners as a therapeutic str

Human CD4 T cells are a functional target for lipid nanoparticle-based mRNA vaccines.

Billions of mRNA-based severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) lipid nanoparticle (LNP) vaccine doses have been administered globally and shown to be both safe and effective. However, the mechanisms by which mRNA vaccines facilitate protection remain unclear. Peripheral blood analyses from vaccinated individuals show B and T cell responses are elicited, but it remains unknown which cells contribute to particle uptake and protein expression to elicit protective immunity. Using mRNA LNP fluorescent reporters, we show that CD4 T cells are an unexpected target for LNP-based pr
