Cancellous bone remodeling in type I (postmenopausal) osteoporosis: quantitative assessment of rates of formation, resorption, and bone loss at tissue and cellular levels.

The cellular mechanisms for bone loss in type I (postmenopausal) osteoporosis are highly controversial. We attempted to resolve this by assessing rates of formation and resorption of iliac cancellous bone by a new histomorphometric method in 89 women with osteoporosis (mean age +/- SD, 66 +/- 6 years) and in 32 carefully selected normal postmenopausal women (64 +/- 6 years). In the osteoporotic women, bone resorption rate was increased by 39% (P less than 0.05) at the cellular level and by 67% (P less than 0.05) at the tissue level, whereas bone formation was unchanged at the tissue level but

Attainment of therapeutic fluoride levels in serum without major side effects using a slow-release preparation of sodium fluoride in postmenopausal osteoporosis.

The bioavailability, biochemical effects, and safety of a slow-release preparation of sodium fluoride were examined. In 8 normal volunteers, a single administration of slow-release sodium fluoride (25 mg) caused a slow rise and gradual decline in serum fluoride concentration, thus avoiding sharp peaks produced by a rapid-release preparation. In 37 patients with postmenopausal osteoporosis, serum fluoride concentration was kept within the "therapeutic window" (95-100 ng/ml) during long-term intermittent sodium fluoride (slow-release) therapy (25 mg twice/day, given for 3 months in each 5-month

Study of 16,16'-dimethyl-trans-delta 2 prostaglandin E1 methyl ester vaginal suppository for cervical dilatation in premenopausal and postmenopausal women.

In a double-blind placebo-controlled comparative study, 180 female patients were randomly assigned to groups and treated approximately 3 h before dilatation and curettage with either a single 1 mg vaginal suppository of Gemeprost or a matching placebo. The patient population included premenopausal non-pregnant or postmenopausal patients presenting at four participating centers in West Germany. Patients were monitored from the time of drug administration to approximately 24 h after the operation. A marked response to treatment with Gemeprost was noted at the general cervical assessment performe

Anger, aging, and reproductive aging: observations from the Seattle Midlife Women's Health Study.

OBJECTIVES: Although investigators have devoted significant effort to understanding women's experiences of depressed mood and clinical depression during the menopausal transition and early postmenopause, they have focused less on women's perimenopausal experiences of emotional arousal, including anger. The purpose of these analyses was to examine the influence of aging and reproductive aging stages on women's reports of anger. METHODS: Participants aged 35-55 years from the Seattle Midlife Women's Health Study population-based cohort (N=501 at baseline) completed an annual health report. Part…

[Effect of menopause on carbohydrate metabolism].

Endocrine alterations in menopause produce disadvantageous changes in the carbohydrate and lipid metabolism. The character and tendency of these alterations are very similar to those found in type II diabetes mellitus. The abdominal visceral fat content as well as the blood level of cholesterol, triglyceride, glucose and insulin increase in postmenopause. These changes can be advantageously influenced by hormone replacement therapy.

[Teriparatide--new value in osteoporosis treatment: treatment guidelines].

Osteoporosis has become global health issue due to the longer life-cycle and increased percentage of older people in population. The great improvement was done in the treatment of postmenopausal osteoporosis. By the mechanism of action, drugs for osteoporosis treatment are antiresorptives and osteoanabolics. Teriparatide is an osteoanabolic drug that stimulates bone turnover and building of a new bone, indicated for treatment of women in postmenopause with multiple osteoporotic fractures.

Antioxidant status in women with uterine leiomyoma: relation with sex hormones.

Uterine leiomyomas are benign soft-tissues tumors that arise from uterine smooth muscle tissue. Etiopathogenesis of leiomyomas is not well understood. We aimed to examine whether antioxidant enzyme activities and lipid hydroperoxides level in patients with leiomyoma are influenced by changes in sex hormones and gonadotropins (estradiol (E2), progesterone, FSH, and LH) during menstrual cycle and in postmenopause. The material consisted of blood and uterine tissue specimens. Hormone concentrations were determined and assays for superoxide dismutase, catalase, glutathione peroxidase and glutathio…

Menopausal hormone therapy combined with antiresorptive therapy for skeletal outcomes in postmenopausal women: a systematic review and network meta-analysis of randomized controlled trials.

IMPORTANCE: Postmenopausal bone loss is a major contributor to osteoporosis, fracture-related morbidity, loss of independence, and mortality. Although systemic menopausal hormone therapy (MHT), antiresorptive therapy, and combination regimens improve bone mineral density (BMD), their comparative skeletal benefits and safety profiles remain incompletely defined. OBJECTIVE: The study aims to compare the therapeutic and preventive skeletal effects of systemic MHT, antiresorptive therapy, and combined MHT plus antiresorptive therapy on site-specific BMD, fracture outcomes, and selected safety out

Combined baseline P1NP and TRAcP5b classification predicts differential bone mineral density response to romosozumab: A retrospective cohort study of 287 postmenopausal women.

UNLABELLED: Introduction Individual bone turnover markers predict bone mineral density response to romosozumab; however, their combined assessment has not been evaluated. We investigated whether a simple classification based on baseline procollagen type I N-terminal propeptide and tartrate-resistant acid phosphatase 5b levels predicts differential lumbar spine bone mineral density response and fracture risk during romosozumab treatment. METHODS: This retrospective study analyzed 287 female patients (mean age, 76.9 years) treated with 210 mg monthly romosozumab for 12 months at a single center

A case-control study of the association between per- and polyfluoroalkyl substances (PFAS) and risk of postmenopausal breast cancer in the Danish diet cancer and health cohort.

BACKGROUND: Exposure to per- and polyfluoroalkyl substances (PFAS) has been associated with breast cancer, however findings have been conflicting and few studies have examined cancer subtypes or risk among subgroups of women. METHODS: We used an individually matched case-control design, nested within the Danish Diet Cancer and Health cohort. We identified 500 incident breast cancer cases with available plasma samples among postmenopausal women aged 50 to 64 years in the Danish Cancer Registry and for each case, we randomly selected from the study base one control woman enrolled within 180 day

Elinzanetant: Efficacy and safety in the management of vasomotor symptoms in postmenopausal women, a systematic review and meta-analysis with grading of recommendations assessment, development, and evaluation assessment.

BACKGROUND: Menopause is frequently associated with different symptoms and signs, such as vasomotor symptoms (VMSs), which can affect women's quality of life. While hormonal therapy is considered the traditional and most used medication for controlling these VMSs associated with menopause, neurokinin (NK) receptor antagonists resemble a novel, promising generation of medications that provide fewer adverse events compared to hormonal therapy. Elinzanetant is the first drug that can act on both NK-1 and 3 receptors by selectively antagonizing them. It has also been developed for the management o
