Cadmium is a nonessential heavy metal and ubiquitous potential environmental pollutant. Although the kidney proximal tubule is an important target for cadmium, the underlying cellular mechanisms of cadmium-induced renal toxicity remain elusive. Cyclophilin B (CypB) is an endoplasmic reticulum (ER)-resident member of the cyclophilin family of proteins that bind cyclosporin A (CsA). We report that as in other cell types, CypB trafficked from the ER and was secreted by keratinocytes into the media in response to CsA. Concentrations as low as 1 pM of CsA induced secretion of CypB. BACKGROUND: Long-term β-adrenergic receptor (β-AR) blockade reduces mortality in patients with heart failure. Chronic sympathetic hyperactivity in heart failure causes sustained β-AR activation, and this can deplete Ca(2+) in endoplasmic reticulum (ER) leading to ER stress and subsequent apoptosis. Legionella pneumophila is a gram-negative pathogen that causes a severe pneumonia known as Legionnaires' disease. Here, we demonstrate for the first time that L. pneumophila infects and grows within cultured human endothelial cells. Endothelial infection may contribute to lung damage observed during Legionnaires' disease and to systemic spread of this organism.

PURPOSE OF REVIEW: Aquaporin-2 is an aquaporin water channel protein present at the apical membrane of kidney collecting duct cells and plays a key role in urine concentrating ability. Like other membrane proteins, aquaporin-2 undergoes dynamic processes within the cells: synthesized, targeted to the subapical region, exocytosed to the apical membrane, endocytosed, recycled and finally degraded. The understanding of the molecular and cellular mechanisms of these events is advancing rapidly, and recent new findings characterizing such processes are reviewed.

Polycystic kidney disease (PKD) is caused by mutations in two genes, PKD1 and PKD2, which encode for the proteins, polycystin-1 (PC1) and polycystin-2 (PC2), respectively. The coronal pulps of four primary canines, free from radicular resorption, and of three primary teeth, with roots entirely resorbed, were prepared for electron microscopy. Mature fibroblasts, similar to those reported in the pulps of permanent teeth, were present in the pulps of the clinically mature teeth. Regression of fibroblasts, evident in the pulps of deciduous teeth at the root resorption stage, was characterized by cytoplasmic changes, modifications in the cytoskeleton and dissociation of cells and fibrils. Deciduous teeth might provide a model for studying pulp fibroblast ageing.

Clinical, electrophysiological, and morphological findings are presented, and possible etiological factors are discussed.
