Radiation-Induced Optical Coherence Tomography Angiography Retinal Alterations in Patients With Nasopharyngeal Carcinoma.

Aim: The aim of the study was to investigate the early neurovascular alterations of the retina in radiation encephalopathy (RE) patients with normal-ranged visual acuity after radiotherapy for nasopharyngeal carcinoma. Methods: Fifty-five RE patients and 54 healthy age-matched subjects were enrolled in this retrospective cross-sectional case-control study. The best corrected visual acuity (LogMAR) of the included eye should not be more than 0. The vessel density and thickness of different locations in the retina were acquired automatically using optical coherence tomography angiography (OCTA).…

Epigenetics in age-related macular degeneration: new discoveries and future perspectives.

The study of epigenetics has explained some of the 'missing heritability' of age-related macular degeneration (AMD). The epigenome also provides a substantial contribution to the organisation of the functional retina. There is emerging evidence of specific epigenetic mechanisms associated with AMD. This 'AMD epigenome' may offer the chance to develop novel AMD treatments.

[Optogenetic reactivation of dormant cone photoreceptors].

Optogenetics enable the restoration of light sensitivity in various retinal cell types. In degenerated retinas, dormant cones whose cell bodies are intact but no longer respond to light can be optogenetically reactivated. This restores all cone-dependent signalling pathways. In RP mouse models the hyperpolarizing opsins eNpHR and Jaws led to the optogenetic reactivation of dormant photoreceptors, with measurable light responses. This was accompanied by improvements in behavioral tests. The transplantation of optogenetically modified photoreceptors also shows functional integration into the ret…

A novel cyp26c1-driven reporter line to study boundaries of retinoic acid signalling during zebrafish development.

Retinoic acid (RA) signalling is essential for vertebrate development, but the spatiotemporal dynamics of its regulation remain largely unch … otic vesicles, pharyngeal arches, and other ectodermal derivatives, as well as in subdomains of the retina. Furthermore, we demonstrate that this line responds to both exogenous and genetic manipulations of RA signalling, particularly within the retina. Our reporter provides a valuable resource for investigating the intricate biology of RA signalling in …

Modulation of glutamate metabolic reprogramming via Ras-Raf-MEK/ERK signaling alleviates immune inflammation of astrocytes in glaucomatous neurodegeneration.

Normal-tension glaucoma (NTG) is characterized by chronic progressive retinal ganglion cell (RGCs) death with progressive optic nerve damage and a diminished visual field that is not accompanied by elevated intraocular pressure (IOP). Recently, the immunological mechanisms underlying nerve injury in glaucoma have been increasingly discussed. Astrocytes are important immune components of the retina that play a crucial role in regulating retinal plasticity and protecting neurons. Studies have revealed that astrocyte dysfunction triggers optic neuropathy without affecting IOP and could be a novel…

Gene augmentation therapy successfully treats mice with complete congenital stationary night blindness (cCSNB), improving retinal function and visual acuity.

Recombinant adeno-associated virus (rAAV) mediated gene therapy is an effective approach for targeting therapeutic genes to retinal photoreceptors. Complete congenital stationary night blindness (cCSNB) is a genetically heterogeneous inherited retinal disease caused by mutations in one of several genes that are part of a large, interdependent depolarizing bipolar cell (DBC) signalplex required for normal synaptic signaling with photoreceptors. These genes include NYX, GRM6, TRPM1, GPR179, and LRIT3, and the resulting cCSNB phenotype is characterized by abnormally low-light vision, myopia, and …

Role of the C-terminal domain in modifying pH-dependent regulation of Cav1.4 Ca2+ channels.

In the retina, Ca2+ influx through Cav1.4 Ca2+ channels triggers neurotransmitter release from rod and cone photoreceptors. Changes in extracellular pH modify channel opening, enabling a feedback regulation of photoreceptor output that contributes to the encoding of color and contrast. However, the mechanisms underlying pH-dependent modulation of Cav1.4 are poorly understood. Here, we investigated the role of the C-terminal domain (CTD) of Cav1.4 in pH-dependent modulation of Ba2+ currents (IBa) in HEK293T cells transfected with the full length CaV1.4 (FL) or variants lacking portions of the C…

Conformal Retinal Image Sensor Based on Electrochemically Exfoliated MoS2 Nanosheets.

Retina-like photoimaging devices with features such as a wide-field-of-view and high spatial resolution have wide application prospects in retinal prosthetics and remote sensing. However, the fabrication of flexible and conformal surfaces is hindered by the incompatible microfabrication processes of traditional rigid, silicon-based substrates. A kirigami strategy for hemispherical surface assembly is proposed to construct a MoS2-based retina-like photodetector array. The device is first fabricated on a flat polyimide (PI) substrate and then tailored using a laser. By approximating the spherica…

Substrate stiffness and pressure alter retinal Müller glia response and extracellular matrix production.

BACKGROUND: The retina is highly influenced by its mechanical environment, with Müller glia (MG) acting as key mechanosensors and extracellular matrix (ECM) producers. This study examined MG responses to substrate stiffness and high pressure (HP), and whether TGF-β1 modulation could mitigate these effects. METHODS: Primary MG from adult rat retinas were cultured on glass (Young's modulus E'=∼1 gigapascal (GPa)) and polyacrylamide gels (10 kPa and 100 kPa). MG were exposed to atmospheric and 70 mmHg (HP) conditions, with TGF-β1 pharmacologically blocked. RESULTS: On glass and 100 kPa gels, MG…

Diabetic retinal disease.

Diabetic retinopathy is a complication of diabetes mellitus that is clinically characterized by changes in retinal microvasculature. Diabetic retinopathy is now better defined as diabetic retinal disease (DRD), as diabetes mellitus affects not only the retinal microvasculature but the whole retina, including neurons and glia. A global concerted effort to study preclinical and clinical signs of DRD and their association with visual acuity and patient-reported vision-related quality of life, and the integration of these features with systemic health and biochemical milieu in people with diabetes…

Gene therapy rescues cone function in opn1mw -/- /opn1sw -/- /Nrl -/- mice, an all-cone model of blue cone monochromacy.

Blue cone monochromacy (BCM) is a congenital vision disorder caused by the loss of L- and M-cone function, leading to severe loss of visual acuity and color perception deficits. To test gene therapy for BCM in a model that mimics the densely packed foveal cones of human retinas, we generated a triple knockout (TKO) mouse model (Opn1mw -/- /Opn1sw -/- /Nrl -/-) with an all-cone retina lacking cone opsins. These mice exhibit abolished cone function, disrupted phototransduction, and altered metabolite profiles resulting from loss of opsin. We then administered AAV-mediated human L-opsin gene supp…

Pancreatic Involvement by Mixed Serous Neuroendocrine Neoplasm Detected by Endosonography With Fine-Needle Tissue Acquisition in Von Hippel-Lindau Disease.

The Von Hippel-Lindau disease (VHL) is an autosomal dominant condition characterized by multiple cystic tumors in several organs, including the pancreas. The symptoms are variable, and suspicion must be raised with typical lesions, such as a hemangioblastoma of the central nervous system (CNS) or retina, associated with a renal cell carcinoma, a pheochromocytoma or multiple pancreatic cysts, besides neuroendocrine tumors (NET). The diagnosis in a patient without a family history should be suspected in case of a hemangioblastoma of the CNS and/or retina, which could also be associated with othe…

Beyond Eye Changes in Ocular Toxicity Studies: Practical Considerations for Sampling and Evaluation of Extraocular Visual Pathways, and Common Findings.

Ocular toxicity studies may encompass a variety of ocular routes of administration. Because the retina and optic nerve are an extension of the central nervous system, safety assessment of ocular compounds must include thorough examination of extraocular visual pathways, as ocular administration may result in primary toxicities in the nervous system due to its exposure to therapeutics via the retina and optic nerve, or changes that are secondary to primary ocular effects. Here we provide a practical guide to sampling extraocular visual pathway structures for commonly utilized ocular toxicology …

Retinal Vessel Analysis and Microvascular Abnormalities in Patients with Philadelphia-Negative Chronic Myeloproliferative Neoplasms.

Background: Philadelphia-negative chronic myeloproliferative neoplasms are a group of clonal hematopoietic disorders including polycythemia vera, essential thrombocythemia, and primary myelofi-brosis. These neoplasms are characterized by an increased risk of thrombotic complications. Several studies have highlighted that the study of vessels of the retina offers the opportunity to visualize, in vivo, the damage to microcirculation that is common in various systemic pathologies. Methods: in our study, forty patients underwent an ophthalmological examination, using non-invasive imaging tech-niqu…
