The genetics of multiple sclerosis: an updated perspective.

PURPOSE OF REVIEW: This review highlights the latest advances in the genetics of multiple sclerosis (MS). While earlier research defined the polygenic architecture of disease susceptibility, recent efforts have focused on unraveling the genetics of disease progression and dissecting the interplay between host genetics and environmental triggers. RECENT FINDINGS: The discovery of the first locus associated with MS severity ( DYSF-ZNF638 ) revealed central nervous system (CNS)-intrinsic mechanisms of progression distinct from immune-mediated susceptibility. Recently, a multi-ancestry genome-wid

Synthetically engineered probiotic suppresses colorectal cancer via inhibition of Wnt/β-catenin signaling.

Colorectal cancer (CRC) is one of the most prevalent malignancies globally. Recent breakthrough of synthetic biology stimulates the utilization of engineered bacteria for cancer diagnosis and treatment. We perform a comprehensive fecal metabolomic analysis in CRC mouse models and patients and identify ornithine as a specific metabolic biomarker for tumor development. We subsequently engineer an E. coli Nissle 1917 (EcN)-based bioluminescent reporter via ornithine-responsive tumor detection. For therapeutic intervention, we construct a probiotic system that locally releases Dickkopf 3 (DKK3), a

Synaptic vesicle-omics in mice captures signatures of aging and synucleinopathy.

Neurotransmitter release occurs through exocytosis of synaptic vesicles. α-Synuclein's function and dysfunction in Parkinson's disease and other synucleinopathies is thought to be tightly linked to synaptic vesicle binding. Age is the biggest risk factor for synucleinopathy, and ~15% of synaptic vesicle proteins have been linked to central nervous system diseases. Yet, age- and disease-induced changes in synaptic vesicles remain unexplored. Via systematic analysis of synaptic vesicles at the ultrastructural, protein, and lipid levels, we reveal specific changes in synaptic vesicle populations,

Potential therapeutic targeting of BKCa channels in glioblastoma treatment.

Potassium channels in brain tissue orchestrate essential cellular processes, including the regulation of membrane potential and neuronal excitability. Among them, large-conductance calcium-activated potassium (BKCa) channels play a pivotal role in both normal brain physiology and the pathogenesis of glioblastoma multiforme, a highly aggressive primary brain tumor. Within the central nervous system, BKCa channels are widely expressed in neurons, astrocytes, and oligodendrocytes, contributing to ion homeostasis and synaptic transmission. In glioblastoma cells, overexpression of BKCa channels, pa

The human mycobiome: a critical yet understudied component of health and disease.

The human body hosts a complex and dynamic microbial community that is crucial for maintaining health. While bacteria dominate this system, fungal communities, collectively called the mycobiome, are increasingly recognized as vital contributors. However, fungi remain understudied due to challenges in culturing many species, limiting our understanding of their roles, interactions and effects on human biology. Advances in next-generation sequencing have transformed mycobiome research, revealing fungal diversity and its impact on health and disease. This review examines the mycobiome's compositio

Molecular landscape, subtypes, and therapeutic vulnerabilities of central nervous system solitary fibrous tumors.

Solitary fibrous tumors (SFTs) of the central nervous system (CNS) are rare, aggressive mesenchymal neoplasms with high recurrence and metastasis rates. Here, we perform a comprehensive multi-omics analysis of 189 cases of CNS SFTs integrating 94 whole genome sequencing, 88 transcriptomics, 7 single-nucleus RNA sequencing and 3 spatial transcriptome sequencing. We find that receptor tyrosine kinase mutations are significantly more prevalent besides the widespread NAB2-STAT6 fusion and correlate with tumor grade. Transcriptomic analysis reveals four molecular subtypes-classical, neural-like, in

Cyanobacterial Peptides in Anticancer Therapy: A Comprehensive Review of Mechanisms, Clinical Advances, and Biotechnological Innovation.

Cyanobacteria-derived peptides represent a promising class of anticancer agents due to their structural diversity and potent bioactivity. They exert cytotoxic effects through mechanisms including microtubule disruption, histone deacetylase inhibition, and apoptosis induction. Several peptides-most notably the dolastatin-derived auristatins-have achieved clinical success as cytotoxic payloads in antibody-drug conjugates (ADCs). However, challenges such as limited tumor selectivity, systemic toxicity, and production scalability remain barriers to broader application. Recent advances in targeted

Hemophagocytic lymphohistiocytosis as the initial manifestation of bone marrow failure in a child with a TERC variant telomere biology disorder.

Hemophagocytic lymphohistiocytosis (HLH) is a life-threatening systemic hyperinflammatory syndrome, rarely associated with bone marrow failure (BMF). Telomere biology disorders (TBD) are caused by inherited defects in telomerase processes and can have heterogeneous presentations including idiopathic pulmonary fibrosis, cirrhosis, and BMF. We report a case of a 10-year-old male from Lima, Peru, who presented with HLH as the initial manifestation of a TBD. He experienced fever, gastrointestinal symptoms, and mucocutaneous involvement. Initial laboratory analyses revealed pancytopenia and elevate

Animal models in leukotriene research: Current insights into complex pathways and therapeutic intervention.

Oxylipins are oxygenated bioactive lipid mediators formed from different polyunsaturated fatty acids through the bioactions of cyclooxygenases, lipoxygenases and cytochrome P450 enzymes, and participate in many physiological and pathophysiological processes. For example, oxylipins regulate vascular tone, the onset of labor, pain, inflammation, fever and contribute to diseases such as asthma, cancer, arthritis, diabetes, obesity and neurodegenerative disorders. Understanding the mechanisms and cellular components involved in oxylipin biosynthesis is crucial for developing effective therapies th

Lysine-reactive tag system for cell-surface protein labeling via N-acyl-N-alkyl sulfonamide chemistry.

Target-selective and site-specific incorporation of a desired functionality into a membrane protein endogenously expressed in cells is a great challenge in chemical biology. Here, we describe a versatile strategy for covalent chemical modification of short peptide tag-fused membrane proteins expressed in cells. This method is based on the recognition-driven reaction of a lysine-containing short histidine tag (KH6 or H6K sequence) and a binuclear nickel (II)-nitrilotriacetic acid (BisNi2+-NTA) complex probe containing a lysine-reactive N-acyl-N-alkyl sulfonamide (NASA). Our reactive peptide tag

Mapping the complexity of ME/CFS: Evidence for abnormal energy metabolism, altered immune profile, and vascular dysfunction.

Myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) is a complex disorder with undefined mechanisms, no diagnostic tools and treatments. To investigate concurrent system dysfunctions, we recruited age- and sex-matched ME/CFS patients and healthy controls for a multimodal analysis of energy metabolism, immune profiles, and plasma proteomics. Immune cells from ME/CFS patients show elevated adenosine monophosphate (AMP) and adenosine diphosphate (ADP) with a reduced ATP/ADP ratio, indicating decreased ATP generation and cellular energy stress. Immune profiling reveals skewing toward less

A human iPSC-Derived myelination model for investigating fetal brain injuries.

Cerebral white matter injuries, such as periventricular leukomalacia, are major contributors to neurodevelopmental impairments in preterm infants. Despite the clinical significance of these conditions, human-relevant models for studying fetal brain development and injury mechanisms remain limited. This study introduces a human iPSC-derived myelination model developed using a microfluidic device. The platform combines spinal cord-patterned neuronal and oligodendrocyte spheroids to recapitulate axon-glia interactions and myelination processes in vitro. The model successfully achieved axonal fasc

Evolution and trajectory of B-cell targeted therapies in rheumatic diseases.

Aberrant B-cell function, which could arise from various underlying causes, is central to the pathogenesis of diverse autoimmune rheumatic diseases. B cells remain the only cell type to be specifically therapeutically targeted through depletion and have the only therapy with a routinely available flow cytometric biomarker of treatment. Since first use and subsequent licensing for rheumatoid arthritis, rituximab has had a transformative impact on patients globally and across the rheumatic diseases. Further insights from B-cell-activating factor (BAFF) blockade with belimumab in systemic lupus e

Reversible and reusable compartmentalized thermoplastic chip for coculture of dorsal root ganglion neurons.

Compartmentalized microfluidic chips play an important role in understanding the cellular mechanisms involved in neurodegenerative disorders. Dorsal root ganglia are a well-established model for modelling the peripheral nervous system (PNS), but their development on a chip remains limited. Furthermore, it would be beneficial for the devices to be openable in order to access the biological material inside for analyses. Easy to prototype and biocompatible, styrenic block copolymers (SBC) are an alternative to polydimethylsiloxane (PDMS) that offer both reversible and permanent bonding properties

Mitochondrial Measures in Primary Cells Isolated from Patients with ME/CFS.

Fibroblasts and peripheral blood mononuclear cells (PBMCs) are commonly utilized cell types for the analysis of mitochondrial function. Fibroblasts, derived from connective tissue, provide a reliable model for studying mitochondrial metabolism due to their active role in energy production and their accessibility for experimental manipulations. PBMCs, on the other hand, are a heterogeneous population of immune cells that include lymphocytes and monocytes. They offer the advantage of reflecting mitochondrial function in circulating cells and providing insights into systemic aspects of mitochondr

Unraveling the Intricacies of Xenophagy: Lessons from Legionella pneumophila.

Autophagy, a conserved eukaryotic process for maintaining cellular homeostasis, plays a crucial role in innate immunity by targeting intracellular pathogens through selective autophagy, known as xenophagy. While xenophagy is vital for pathogen clearance, many intracellular bacteria have evolved sophisticated strategies to avoid or subvert this process. Legionella pneumophila, a Gram-negative intracellular pathogen, manipulates host pathways by an extensive repertoire of effector proteins delivered through its type IV secretion system. This review summarizes the current understanding of xenopha

New insights on an old friend: AroA linked to iron-dependent outer membrane stability.

Salmonella is a common causative agent of infectious intestinal and systemic disease and has been extensively studied for several decades. Yet, much of Salmonella pathogenicity remains a mystery due in part to the highly complex virulence and adaptation strategies at the pathogen's disposal. One of the more influential tools within the field, an attenuated aroA-deficient Salmonella strain, has been used for many years to probe the host immune response that would otherwise be impossible with a fully virulent strain. Now, new work by Rooke et al. (J. L. Rooke, E. C. A. Goodall, K. Pullela, R. Da

Exploring the Immunoresponse in Bladder Cancer Immunotherapy.

Bladder cancer (BC) represents a wide spectrum of diseases, ranging from recurrent non-invasive tumors to advanced stages that require intensive treatments. BC accounts for an estimated 500,000 new cases and 200,000 deaths worldwide every year. Understanding the biology of BC has changed how this disease is diagnosed and treated. Bladder cancer is highly immunogenic, involving innate and adaptive components of the immune system. Although little is still known of how immune cells respond to BC, immunotherapy with bacillus Calmette-Guérin (BCG) remains the gold standard in high-risk non-muscle i
