The use of the oral mucosa for drug delivery and the erroneous belief that it is a nonkeratinized tissue have given rise to the suggestion that the oral mucosa is a permeable tissue. Such an assumption is not supported by studies which indicate that permeability differs significantly in different oral regions, depending on the pattern of epithelial differentiation. The pharmacokinetics of sulfadimidine (SDM) and its N4-acetyl metabolite (N4SDM) were investigated after intravenous bolus injection of a single dose (200 mg/kg) of SDM in normal and diseased New Zealand white rabbits. Following topical administration to albino rabbits, bunolol hydrochloride, a beta 1- and beta 2-adrenoceptor antagonist, was rapidly absorbed and distributed in ocular tissues, and transformed intraocularly to a single metabolite, dihydrobunolol. The half-life of bunolol was approximately 1.26 hours in the aqueous humor and 1.63 hours in the ciliary body. There was no significant accumulation of bunolol during a four-day period of twice-daily dosing. Most of the drug was excreted in the urine following ocular or intravenous administration.

The antimycobacterial activity of rifaximin was studied in the guinea pig, after oral administration. The experiment confirmed the scanty absorption of rifaximin in the gastrointestinal tract, as the two dosages used (60 and 30 mg/kg) did not modify the tubercular illness after four months of therapy. Furthermore, sensitivity to rifaximin of the callenging Mycobacterium tuberculosis isolated at the end of the study from the spleen, lung, liver and lymph nodes of the guinea pigs that received rifaximin did not change.
