Antigenic and enzymatic changes in infected and transformed human diploid cells.

Human diploid fibroblasts, WI-38, were infected with various agents and the levels of lysosomal enzymes determined by immunochemical quantitation. Esterase levels were raised by mumps virus and Toxoplasma gondii infection. The concentration of beta-D-glucuronidase was reduced by these same agents. Beta-D-N-acetyl glucosaminidase was greatly increased in cultures infected with T. gondii and decreased by mycoplasma infection. Two cultures transformed by SV40 showed reduced levels of esterase compared with WI-38, as did one of two transformed amnion cultures. A second amnion culture and a culture

Type-specific polysaccharide antigens of group B streptococci. II. The chemical basis for serological specificity of the type II HCl antigen.

The type-specific antigen of Group B Type II streptococci has been prepared free of other known cell components. This antigen is a capsular polysaccharide containing D-galactose, D-glucose, N-acetyl-D-glucosamine, and a labile component which has not been chemically characterized. Extraction of Type II streptococci with cold TCA yielded this antigen which contained two serological determinants. A partial antigen with only one of the determinants was obtained by extraction with dilute HCl at 100 degrees C. By means of the quantitative precipitin inhibition technique, a (beta-D-galactopyranoside

Hydrogen sulfide engages catalytic zinc in HDAC6 to modulate an alternative deacetylation pathway.

Hydrogen sulfide (H₂S) is a gasotransmitter whose non-covalent effects on protein function remain incompletely understood. Here we show that H₂S-derived sulfur species engage the catalytic Zn²⁺ center of histone deacetylase 6 (HDAC6) and enhance deacetylase activity under defined conditions. Molecular dynamics simulations support access of HS⁻ to the CD2 active-site tunnel, whereas surface plasmon resonance and zinc-depletion experiments support reversible, zinc-dependent engagement. Quantum chemical calculations support a chemically feasible sulfur-associated deacetylation pathway that genera…

CCER1 condensates participate in histone-to-protamine transition by recruiting the TIP60/EPC1/NuA4 acetyltransferase complex.

Cases of idiopathic male infertility may be caused by as-yet unrecognized genetic factors. Here, we report that the testis-specific protein … airs the recruitment and interaction of TIP60 and EPC1 subunits, leading to reduced histone H4 hyperacetylation in nucleosomes, defective DNA strand breakage and insufficient histone-to-protamine (HTP) replacement during spermiogenesis. In conclusion, our data indicate that the TIP60/EPC1/NuA4-CCER1 complex is involved in the epigenetic regulation of HTP repl …

SIRT2 deacylase modulators control B cell metabolic reprogramming in EBV infection and mitogenic activation.

Sirtuin-2 (SIRT2) is a cellular deacylase, regulating cell cycle progression and metabolic homeostasis. Recently, SIRT2 has emerged as a tar … l swelling, and inducing nutrient stress and autophagy. Treatment with SIRT2 modulators drives hyperacetylation of targets involved in lipid metabolism, central carbon metabolism, and oxidative phosphorylation. EBV-positive and EBV-negative B cell lymphomas rely on glycolysis to avoid cell death after SIRT2 modulation, revealing a metabolic vulnerability that …

Running rich: how excess fatty acid oxidation drains the cardiac engine.

Fatty acid oxidation (FAO) provides the healthy heart with 60%-90% of its ATP, with the remainder coming from metabolism of glucose. Metabolic flexibility is key to heart function, ensuring an uninterrupted source of fuel. In heart failure, a shift from FAO to glucose-dependent metabolism occurs as disease progresses, supporting the widely held notion that fat is the optimal substrate in the heart. In this issue of the JCI, Kim et al. challenge this assumption. In studies of acetyl-CoA carboxylase-deficient (ACC-deficient) mice, they found that unregulated use of fat as a substrate led to card

Mechanism of the synergistic action of oxythiamine and ketoconazole against the yeast Malassezia pachydermatis.

This article explains the biochemical basis of the synergistic effect of oxythiamine (OT) and ketoconazole (KTC) against Malassezia pachydermatis yeast, which was isolated from dogs exhibiting clinical signs of otitis externa. All strains were incubated on MLNA medium supplemented with OT, KTC, or a mixture of both compounds. We found that the ergosterol content was reduced by the compounds tested, both separately (20%-50%) and in combination (80%). OT alone and in combination with KTC reduced NADPH levels. However, we found no differences in acetyl-CoA levels under the influence of the compou

Application of chitosan as a novel antimicrobial dental material -Effects of form, molecular weight, and deacetylation degree on its antimicrobial activity against Candida albicans.

Oral candidiasis is a significant health concern, especially for elderly individuals. The present study examined chitosan as a potential antimicrobial dental material for Candida albicans treatment. Four types of chitosan products purified from Chionoecetes opilio and Decapodiformes, with varying weight average molecular weight and degree of deacetylation (DAC) were used. Antimicrobial activity was assessed with fungal viability assays, culture turbidity measurements, and live/dead cell staining, while mechanism of action was determined by examining extracellular adenosine triphosphate (ATP) l…

Deletion of pyruvate dehydrogenase kinases reduces susceptibility to deep vein thrombosis in mice.

Neutrophils contribute to deep vein thrombosis (DVT) by releasing prothrombotic neutrophil extracellular traps (NETs). NET formation (known as NETosis) is an energy-intensive process that requires an increased rate of aerobic glycolysis. The metabolic enzymes pyruvate dehydrogenase kinases (PDKs) inhibit the pyruvate dehydrogenase complex to divert the pyruvate flux from oxidative phosphorylation toward aerobic glycolysis. Herein, we identified that the combined deletion of PDK2 and PDK4 (PDK2/4-/-) renders mice less susceptible to DVT (measured by thrombus incidence, weight, and length) in th

Identification of programmed cell death-related genes and diagnostic biomarkers in endometriosis using a machine learning and Mendelian randomization approach.

BACKGROUND: Endometriosis (EM) is a prevalent gynecological disorder frequently associated with irregular menstruation and infertility. Programmed cell death (PCD) is pivotal in the pathophysiological mechanisms underlying EM. Despite this, the precise pathogenesis of EM remains poorly understood, leading to diagnostic delays. Consequently, identifying biomarkers associated with PCD is critical for advancing the diagnosis and treatment of EM. METHODS: This study used datasets from the Gene Expression Omnibus (GEO) to identify differentially expressed genes (DEGs) following preprocessing. By c

Blood meal drives de novo lipogenesis in the fat body of Rhodnius prolixus.

In insects, lipids are stored in the fat body mainly as triacylglycerol. Lipids can be directly provided by digestion and incorporated from the hemolymph, or synthesized de novo from other substrates such as carbohydrates and amino acids. The first step in de novo lipid synthesis is catalyzed by acetyl-CoA carboxylase (ACC), which carboxylates acetyl-CoA to form malonyl-CoA. Rhodnius prolixus is a hematophagous insect vector of Chagas disease and feeds exclusively on large and infrequent blood meals. Adult females slowly digest the blood and concomitantly accumulate lipids in the fat body. In

[3-hydroxy-3-methylglutaryl-CoA lyase deficiency: a case report and literature review].

INTRODUCTION: 3-hydroxy-3-methylglutaryl-CoA (HMG-CoA) lyase deficiency is an autosomal recessive disorder that usually presents in the neonatal period with vomiting, metabolic acidosis, hypoglycemia and absent ketonuria. Few cases are reported in the literature, and optimal dietary management and long term outcome are not fully understood. CASE REPORT: We report a 2 year old girl with HMG-CoA-lyase deficiency who had limited fasting tolerance on a low protein diet, with several recurrent hospital admissions with severe hypoketotic hypoglycaemia and metabolic acidosis. We also review the diet

Novel roles of Pkd2 in male reproductive system development.

Autosomal dominant polycystic kidney disease (ADPKD) is one of the most common inherited genetic diseases, caused by mutations in PKD1 and/ or PKD2. Infertility and reproductive tract abnormalities in male ADPKD patients are very common and have higher incidence than in the general population. In this work, we reveal novel roles of Pkd2 for male reproductive system development. Disruption of Pkd2 caused dilation of mesonephric tubules/efferent ducts, failure of epididymal coiling, and defective testicular development. Deletion of Pkd2 in the epithelia alone was sufficient to cause reproductive

Peptide: N- glycanase is expressed in prestalk cells and plays a role in the differentiation of prespore cells during development of Dictyostelium discoideum.

Peptide: N-glycanase (PNGase) enzyme is found throughout eukaryotes and plays an important role in the misfolded glycoprotein degradation pathway. This communication reports the expression patterns of the pngase transcript (as studied by the analysis of beta-galactosidase reporter driven by the putative pngase promoter) and protein (as studied by the analysis of beta-galactosidase reporter expressed under the putative pngase promoter as a fusion with the pngase ORF) during development and further elucidated the developmental defects of the cells lacking PNGase (png(-)). The many roles of histone deacetylases in development and physiology: implications for disease and therapy.

Histone deacetylases (HDACs) are part of a vast family of enzymes that have crucial roles in numerous biological processes, largely through their repressive influence on transcription. The expression of many HDAC isoforms in eukaryotic cells raises questions about their possible specificity or redundancy, and whether they control global or specific programmes of gene expression. Recent analyses of HDAC knockout mice have revealed highly specific functions of individual HDACs in development and disease. Mutant mice lacking individual HDACs are a powerful tool for defining the functions of HDACs

Effects of ethanol and phenobarbital on hemoglobin adducts formation in rats exposed to benzidine and Direct Black 38.

The objective of this study was to evaluate the effects of pretreatment of ethanol (EtOH) and phenobarbital (PB), which are known to affect … te that EtOH not only increases the adduct formation by inducing N-hydroxylation but also induces N-acetylation. PB induced N-hydroxylation and increased the adduct formation in BZ group, but decreased the adduct formation in DB38 group due to decreasing azo reduction. These results suggest that the effects of EtOH or PB should be considered in biochemical mo …
