In vitro and in silico evaluation of Turbinaria ornata-derived compounds: elucidating mechanisms of anticandidal and anticancer activities.

Candida genera play a crucial role as infectious pathogens in humans and have the ability to form biofilms, which impede the efficacy of antifungal medications, thereby elevating resistance levels. In this present study, the extracts of Turbinaria ornata were used for the treatment of clinical isolates of Candida species, and UPLC-ESI-Q-TOF MSE was employed for the chemical profiling of macroalgae extracts. The methanol extract of T. ornata has excellent with MIC (125 mg/mL), particularly against C. krusei by reduction of adhesion compared with acetone and ethyl acetate extracts. The toxicity

Chemical constituents of Rhazya stricta as potential Aurora kinase A inhibitors for targeting HER2-negative breast cancer: an integrated in-silico and in-vitro study.

Human Epidermal Growth Factor Receptor 2 (HER2) negative breast cancer is a type of breast cancer that does not overexpress the HER2 protein. In some breast cancers, overexpression of HER2 makes the cancer more aggressive. Computational-aided drug discovery (CADD) was taken as an efficient alternative to conventional methods of drug design and development. This study has also been carried out using the same drug design and development methods from Rhazya stricta; whose extracts are traditionally approved to possess anti-cancer activity while choosing Aurora Kinase A (AURKA) as our target of in

A Systematic Review On ERK/MAPK Signaling Circuit As The Critical Regulator In Chronic Wound Healing: Phytochemicals As Possible Modulators?

INTRODUCTION: Wound healing is an important process in tissue regeneration and repair, during which events such as cell movement, proliferation, and tissue remodeling are keyfactor. Several pathological conditions, such as diabetes, hinder wound-healing processes. Mechanistically, healing processes are governed by many signaling pathways, which also influence scar formation; the ERK/MAPK pathway is a principal regulator of both wound healing and scar formation. Given the prevalence of chronic wounds worldwide, understanding this pathway is crucial for developing better treatment strategies. M

Multitarget Therapeutic Strategies for Chagas Disease: Natural Compounds, Antimicrobial Peptides, and Cell-Based Immunomodulation.

Chagas disease, caused by Trypanosoma cruzi, remains a major public health problem in Latin America and an emerging global health concern due to population mobility. Although benznidazole and nifurtimox remain the only approved antiparasitic drugs, their limited efficacy in chronic infection, prolonged treatment regimens, frequent adverse effects, and variable activity across parasite strains highlight the need for new therapeutic strategies. In addition, the pathogenesis of chronic Chagas disease is driven not only by parasite persistence but also by immune-mediated tissue damage, particularl

Tissue Mosaicism in Xenogenic Common Carp (Cyprinus carpio) Injected with Unsorted Gonadal Cells from Blue Catfish (Ictalurus furcatus).

Xenogenic individuals are sterilized hosts that are colonized by germ cells transplanted from a donor species and can subsequently produce donor-derived gametes. In our case, unsorted gonadal cells from a donor species injected near the genital ridge of a sterile surrogate should colonize only that region, eventually becoming donor gametes. Unsorted gonadal cells from blue catfish (Ictalurus furcatus) injected into triploid common carp (Cyprinus carpio) embryos and fry were detected by PCR assays predominantly in recipient gonads and, in a subset of individuals, occasionally in DNA extracts fr

GAD1 alleviates injury-induced optic neurodegeneration by inhibiting retinal ganglion cell apoptosis.

The degeneration of the optic nerve narrows the visual field, eventually causing overall vision loss. This study aimed to identify global protein changes in the retina of optic nerve crushing (ONC) mice and to identify key regulators and pathways involved in injury-induced cell death during the progression of optic neurodegeneration. Label-free quantitative proteomics combined with bioinformatic analysis was performed on retinal protein extracts from ONC and sham-operated mice. Among the 1433 proteins detected, 121 proteins were differentially expressed in the retina of ONC mice. Further bioin

Perinatal treatment of parents with the broad-spectrum antibiotic enrofloxacin aggravates contact sensitivity in adult offspring mice.

BACKGROUND: Antibiotics, while eliminating pathogens, also partially deplete commensal bacteria. Antibiotic-induced dysbiosis may contribute to the observed rise in "immune-mediated" diseases, including autoimmunity and allergy. The aim of this study is to investigate the impact of perinatal antibiotic treatment on T cell-mediated immune response in adult mice. METHODS: Oral treatment with broad-spectrum antibiotic enrofloxacin during gestation and breastfeeding or breastfeeding or gestation alone was used to evaluate whether antibiotic exposure early in life could modulate contact sensitivit

Diverse Localization Patterns of an R-Type Lectin in Marine Annelids.

Lectins facilitate cell-cell contact and are critical in many cellular processes. Studying lectins may help us understand the mechanisms underlying tissue regeneration. We investigated the localization of an R-type lectin in a marine annelid (Perinereis sp.) with remarkable tissue regeneration abilities. Perinereis nuntia lectin (PnL), a galactose-binding lectin with repeating Gln-X-Trp motifs, is derived from the ricin B-chain. An antiserum was raised against PnL to specifically detect a 32-kDa lectin in the crude extracts from homogenized lugworms. The antiserum detected PnL in the epidermis
