Effects of virus dose and extrinsic incubation temperature on vector competence of Culex nigripalpus (Diptera: Culicidae) for St. Louis encephalitis virus.

Culex nigripalpus Theobald is a primary vector of St. Louis encephalitis virus in the southeastern United States. Cx. nigripalpus females were fed blood containing a low (4.0 +/- 0.01 log10 plaque-forming unit equivalents (PFUeq) /ml) or high (4.7 +/- 0.1 log10 PFUeq/ml) St. Louis encephalitis virus dose and maintained at extrinsic incubation temperatures (EIT) of 25 or 28 degrees C for 12 d. Vector competence was measured via quantitative real-time reverse transcriptase polymerase chain reaction to estimate PFUeq using rates of infection, dissemination, and transmission. There were no differe

Prolonged culture for mycobacteria in mediastinal lymph nodes from patients with pulmonary sarcoidosis. A negative study.

BACKGROUND: Histological similarity between sarcoidosis and tuberculosis, with epithelioid cell granulomas as the typical finding, suggests that sarcoidosis could be a specific manifestation of infection with mycobacteria. However, convincing evidence for a causal relationship between mycobacterial infection and sarcoidosis has not been produced. OBJECTIVE: To examine whether mycobacteria could be cultured from mediastinal lymph nodes from patients with newly diagnosed pulmonary sarcoidosis using a prolonged culture technique. METHODS: The series comprised 15 patients (10 men) with a median

Involvement of CD4+ T cells and macrophages in acquired protection against infection with Sporothrix schenckii in mice.

The cell-mediated immune response against infection with Sporothrix schenckii was investigated in vivo and in vitro. Following primary infection, mice were protected against a secondary subcutaneous inoculation with S. schenckii as determined by the local growth of fungi, and subcutaneously immunized mice were able to survive a lethal intravenous infection. Protection could be transferred to naive congenitally athymic nude mice using lymph node cells taken from immune mice. When immune lymph node cells had CD4+ cells depleted by antibody plus complement, the ability to transfer protection was

Chemical and pathogen-induced inflammation disrupt the murine intestinal microbiome.

BACKGROUND: Salmonella is one of the most significant food-borne pathogens to affect humans and agriculture. While it is well documented that Salmonella infection triggers host inflammation, the impacts on the gut environment are largely unknown. A CBA/J mouse model was used to evaluate intestinal responses to Salmonella-induced inflammation. In parallel, we evaluated chemically induced inflammation by dextran sodium sulfate (DSS) and a non-inflammation control. We profiled gut microbial diversity by sequencing 16S ribosomal ribonucleic acid (rRNA) genes from fecal and cecal samples. These dat

Analysis of HBV cccDNA Minichromosome Accessibility by MNase-qPCR and High-Throughput Sequencing.

The covalently closed circular DNA (cccDNA) of the hepatitis B virus (HBV) is organized as a minichromosome structure in the nucleus of infected hepatocytes and considered the major obstacle to the discovery of a cure for HBV. Until now, no strategies directly targeting cccDNA have been advanced to clinical stages as much is unknown about the accessibility and activity regulation of the cccDNA minichromosome. We have described the method for evaluation of the cccDNA minichromosome accessibility using micrococcal nuclease-quantitative polymerase chain reaction and high-throughput sequencing, wh

Molecular Typing of Syphilis-Causing Strains Among Human Immunodeficiency Virus-Positive Patients in Antwerp, Belgium.

Centers for Disease Control and Prevention and sequencing-based treponeme typing was used to analyze 72 blood samples, collected from human immunodeficiency virus and syphilis co-infected patients during 2014 to 2015 in Antwerp, Belgium. Twenty-nine (40.3%) isolates were polymerase chain reaction positive for Treponema pallidum, and all tested were macrolide-resistant. Four genotypes were identified by sequencing-based typing including two new genotypes, U4NR8 and SU9R8, whereas enhanced Centers for Disease Control and Prevention typing revealed 7 subtypes.

Opisthorchis viverrini: detection by polymerase chain reaction (PCR) in human stool samples.

A polymerase chain reaction (PCR) assay was evaluated for detection of Opisthorchis viverrini eggs in the stool specimens of light and heavily infected individuals in Khon Kaen province of Thailand. A total of 75 fecal specimens were analyzed by PCR following DNA extraction. All the microscopically positive samples were positive by PCR, while 23 of 30 (76.6%) microscopically negative samples were also PCR positive. The sensitivity of the assay was 5 eggs/g of stool. This method is potentially useful in the diagnosis of human opisthorchiasis in endemic areas for treatment and in epidemiological

Detection of Japanese encephalitis virus by reverse transcription/polymerase chain reaction.

A simple and rapid reverse transcription/polymerase chain reaction (RT-PCR) assay for detection of Japanese encephalitis virus (JEV) envelope (E) gene sequences in various biological samples is described. The assay successfully amplified JEV E gene sequences from infected cell cultures, Aedes aegypti larvae, mosquitoes and mouse blood. The sensitivity of the assay was currently 1 ng of JEV RNA and could be increased up to 1 pg on the background of 1 microgram of cellular RNA by biotinylation of the PCR product, Southern blot analysis and streptavidin/alkaline phosphatase detection.

Direct isolation of DNA from patient stools for polymerase chain reaction detection of Cryptosporidium parvum.

Although polymerase chain reaction (PCR) can sensitively detect parasitic or other infections, its use with fecal samples is extremely limited, primarily because of the presence of substances that inhibit DNA extension. Here an improved protocol is reported for directly isolating DNA from aged or fresh formalin-fixed stools, which can then be used to detect Cryptosporidium parvum by nested PCR. This method is highly reproducible, sensitive, and specific. It detects <1 pg of C. parvum DNA in human stool, and there are no cross-reactions with other parasites commonly found there.

[Genotyping of Shigella flexneri 2a strains isolated from patients with acute dysentery in St. Petersburg].

Polymerase chain reaction with universal primers (UP-PCR) was used for the genotyping of 76 S.flexneri 2a cultures isolated from patients with acute dysentery in infectious and psychoneurological hospitals of St. Petersburg. 9 types were determined, and each of them included cultures with identical UP-PCR patterns. The population of the infective agent was more heterogeneous in psychoneurological hospitals where the change of types was registered in May-September 1995. Some of these types were probably epidemic strains. UP-PCR was found to be a promising method increasing the efficiency of tra

Targeted interventions suppress epidemic outbreaks in spatial higher-order activity-driven networks.

Traditional pairwise epidemic models fail to capture group-based infection risks in dense urban environments, where higher-order interactions in shared spaces drive explosive contagion. We develop a spatial activity-driven higher-order framework integrating agent-based mobility modeling with the microscopic Markov chain approach to simulate these complex contagion processes. Our results demonstrate that higher-order interactions significantly advance outbreak peaks by intensifying localized infection pressure, triggering explosive spreading patterns driven by cumulative infection pressure. Profound Hemolytic Anemia in Babesiosis With Lyme Disease Coinfection and Occult Strongyloidiasis.

Babesiosis is a tick-borne infection caused by intraerythrocytic Babesia parasites and may result in severe hemolytic anemia and systemic complications. Coinfection with other Ixodes-borne pathogens may further complicate diagnosis and management. We report a 45-year-old immunocompetent man working as a landscaper in Connecticut who presented with syncope following two weeks of fever, fatigue, diaphoresis, arthralgias, jaundice, and dark urine. Laboratory evaluation revealed profound hemolytic anemia with a hemoglobin of 6.8 g/dL, elevated lactate dehydrogenase (LDH), undetectable haptoglobin,

Superior Mesenteric Vein and Portal Vein Thrombosis in an Immunocompetent Patient With Acute Cytomegalovirus Infection.

Cytomegalovirus (CMV) infections are common worldwide, with clinical manifestations varying from asymptomatic infections in healthy people to life-threatening end-organ dysfunction in immunocompromised patients. Acute CMV infection has been associated with venous thrombosis in immunocompetent patients. We present a rare case of acute superior mesenteric vein (SMV) and portal vein thrombosis in an immunocompetent patient with acute CMV infection. A 28-year-old patient with a past medical history of obesity presented with two days of sharp, 9/10 left upper quadrant (LUQ) pain that radiated to th

Outcome of hematopoietic stem cell transplantation in severe combined immune deficiency with central nervous system viral infection.

BACKGROUND: Patients with severe combined immunodeficiency and preexisting viral pneumonitis formally had a poor outcome from hematopoietic stem cell transplantation. With inhaled steroid and antitumor necrosis factor alpha antibody treatment, results improved. The poor outcome of patients with viral central nervous system infection prompted this retrospective single center review. RESULTS: Eight of 71 patients with severe combined immunodeficiency transplanted since 1987 were identified with viral central nervous system infection (adenovirus [1], cytomegalovirus [2], Epstein-Barr virus [2],

Helicobacter pylori and cagA gene detected by polymerase chain reaction in gastric biopsies: correlation with histological findings, proliferation and apoptosis.

CONTEXT AND OBJECTIVE: The virulence of Helicobacter pylori (HP) in gastroduodenal disease is related to pathogenicity islands (cagPAI) present in some strains. Infection with cagPAI induces IL-8 secretion, increases epithelial cell proliferation and may be important in carcinogenesis. Our objective was to detect HP and the cagA gene (cagPAI marker) by polymerase chain reaction (PCR) and to correlate these results to histological findings, epithelial cell proliferation and apoptosis. DESIGN AND SETTING: Retrospective, at the Surgical and Molecular Pathology Laboratory, Hospital Sírio-Libanês.

Co-infection dynamics of Aeromonas jandaei and Edwardsiella tarda in Oscar fish (Astronotus ocellatus): Effects on pathogenicity, oxidative stress and immune responses.

Co-infections involving multiple bacterial pathogens represent an emerging challenge in aquaculture, yet their effects on host physiology and immune responses remain poorly understood. The present study investigated the pathogenicity and host responses associated with experimental co-infection of Aeromonas jandaei and Edwardsiella tarda in Oscar fish (Astronotus ocellatus). Following determination of the median lethal dose (LD50) for each pathogen, fish were challenged with single infections or a co-infection and evaluated for mortality, clinical signs, biochemical responses, and immune-relate

Diagnostic advantage of droplet digital PCR over blood culture in sepsis: a retrospective study of 115 patients.

BACKGROUND: Sepsis is a life-threatening condition that requires rapid and accurate identification of causative pathogens to guide timely antimicrobial therapy. Blood culture is the current diagnostic standard, but it suffers from low sensitivity and long turnaround times. Droplet digital polymerase chain reaction (ddPCR) has emerged as a promising molecular tool that may overcome these limitations. This study aimed to evaluate the diagnostic utility of ddPCR compared with conventional blood culture in patients with sepsis. METHODS: We performed a retrospective study of 115 patients admitted

From dysbiosis to disease: the role of gut microbial communities in Toxoplasma gondii pathogenesis, zoonotic transmission, diagnostic innovation, and therapeutic outcomes.

Toxoplasma gondii, an obligate intracellular protozoan infecting approximately one-third of the global human population, causes substantial morbidity in immunocompromised individuals, congenital complications, neuropsychiatric sequelae, and considerable economic losses in livestock production. Gut microbial communities critically modulate T. gondii infection susceptibility, disease progression, and clinical outcomes, positioning the microbiome as a central axis in toxoplasmosis pathogenesis. This review examines the bidirectional relationship between gut microbiota and T. gondii, wherein dysbi
