From HSV infection to erythema multiforme through autoimmune crossreactivity.

Scientific and clinical data indicate that human herpes simplex virus 1 (HSV1) and, at a lesser extent, human herpes simplex virus 2 (HSV2) are factor(s) implicated in the development of erythema multiforme (EM). With a focus on oral EM, the present structured review of proteomic and epitope databases searched for the molecular basis that might link HSV1 and HSV2 infections to EM. It was found that a high number of peptides are shared between the two HSVs and human proteins related to the oral mucosa. Moreover, a great number of the shared peptides are also present in epitopes that have been e

A novel mutation in fibrillin-1 gene identified in a Chinese family with marfan syndrome.

Marfan syndrome (MFS) is an autosomal dominant inheritary disorder of the connective tissue. We report clinical features of a Chinese family with MFS and identify mutations in fibrillin-1 gene (FBN1). In this study, all three members of this family underwent complete ophthalmologic examinations. Two of the three members were diagnosed with MFS. Molecular genetic analysis was performed on the three members. All coding exons of FBN1 were amplified by polymerase chain reaction (PCR). The amplified products were sequenced and compared with a reference sequence in the database. Possible structural

STEAP1 is overexpressed in cancers: a promising therapeutic target.

The six-transmembrane epithelial antigen of prostate (STEAP) protein was identified in advanced prostate cancer and is up-regulated in multiple cancer cell lines, including prostate, bladder, colon, ovarian, and Ewing sarcoma. STEAP1 was described as a suitable antigen for T-cell-based or antibody-based immunotherapy. We have investigated the expression of STEAP1 in 40 human tumor types - brain, epithelial, lymphoid - and in their normal tissue counterparts using publicly available gene expression data, including the Oncomine Cancer Microarray database. STEAP1 was found significantly overexpre

Gene cloning, expression, and functional characterization of an ornithine decarboxylase protein from Serratia liquefaciens IFI65.

Putrescine has a negative effect on health and is also used as an indicator of quality on meat products. We investigated the genes involved in putrescine production by Serratia liquefaciens IFI65 isolated from a spoiled Spanish dry-cured ham. We report here the genetic organization of its ornithine decarboxylase encoding region. The 5506-bp DNA region showed the presence of three complete and two partial open reading frames. Putative functions have been assigned to several gene products by sequence comparison with proteins included in the databases. The second gene putatively coded for an orni

What is a 'novel' mtDNA mutation--and does 'novelty' really matter?

The hunt for pathogenic mitochondrial DNA (mtDNA) mutations is often fueled by the seeming novelty of mutations that are either nonsynonymous or affect the protein synthesis machinery in patients. In order to determine the novelty of a detected mutation, the working geneticist nearly always consults MITOMAP--often exclusively. By reanalyzing some case studies of refractory anemia with ring sideroblasts, prostate cancer, and hearing impairment, we demonstrate that the practice of solely relying on MITOMAP can be most misleading. A notorious example is the T1243C mutation, which was assessed to

Distinct novel mutations affecting the same base in the NIPA1 gene cause autosomal dominant hereditary spastic paraplegia in two Chinese families.

Hereditary spastic paraplegia (HSP) is a neurodegenerative disease characterized by lower-limb spasticity, hyperreflexia, progressive spastic gait abnormalities, and an extensor-plantar response. It is genetically very heterogeneous, with 28 Human Genome Organisation (HUGO)-approved IDs in the database (last search: August 8, 2004). Following the identification of the SPG6 gene, NIPA1, we have identified two novel mutations, c.316G>C and c.316G>A, in two independent Chinese families linked to the SPG6 locus. These two mutations would cause a p.G106R substitution, and cosegregated with the dise

LipiDecipher: A Structure-Oriented Analytical Framework for Interpretable Clinical Lipidomics.

BACKGROUND: Clinical lipidomics can capture disease-associated molecular alterations at high resolution, yet translating complex lipid species data into interpretable biological insight remains challenging. Existing workflows often emphasize statistical discrimination while underutilizing the structural information embedded in lipid species. To address this gap, we developed LipiDecipher, a structure-oriented analytical framework designed to summarize lipidomic alterations into interpretable structural patterns and to provide database-supported biological contextualization. METHODS: LipiDecip

A bioinformatics and experimental approach identifies glycoprotein-based diagnostic and prognostic biomarkers for colon adenocarcinoma.

Colon adenocarcinoma (COAD) remains a leading cause of cancer-related mortality worldwide, partly due to the lack of robust biomarkers for early diagnosis and accurate prognosis. Glycoproteins play critical roles in tumorigenesis and may serve as promising sources of biomarkers. This study aimed to identify glycoprotein-related candidate diagnostic and prognostic biomarkers for COAD through integrated bioinformatics approaches and experimental validation. Gene expression profiles and clinical information of COAD patients were obtained from The Cancer Genome Atlas database (TCGA). The expressio

MAPK8 and HDAC6: potential biomarkers related to autophagy in diabetic retinopathy based on bioinformatics analysis.

INTRODUCTION: One of the most common vascular diseases of the retina is diabetic retinopathy (DR), a microvascular condition caused by diabetes. The autophagy system transports and degrades cytoplasmic substances to lysosomes as part of the intracellular degradation process. Autophagy appears to be an important regulator in the development and progression of DR, but its mechanism and potential role are unclear. The purpose of this study is to identify autophagy-related genes in DR and find potential biomarkers associated with DR through bioinformatics analysis. METHOD: We retrieved the datase

miR-455-3p has superior diagnostic potential to PSA in peripheral blood for prostate cancer.

BACKGROUND: Prostate-specific antigen (PSA) is commonly used as a biomarker to diagnose and predict the course of prostate cancer (PCa). However, PSA detection is susceptible to changes in the physiologic environment, which may lead to some misdiagnosis. Thus, it is crucial to find a novel diagnostic marker. METHODS: We accessed microRNA (miRNA) expression datasets (GSE206793 and GSE112264) from the GEO database, analyzing peripheral blood samples from PCa patients. Differentially expressed miRNAs (DEmiRNAs) were identified using GEO2R. A specific miRNA, miR-455-3p, was pinpointed through rig

In Silico Identification of Natural SIRT1 Inhibitors through Molecular Docking, Dynamics Simulation, and MM/PBSA.

Huntington’s disease (HD) is a progressive neurodegenerative disorder caused by mutations in the huntingtin (HTT) gene, leading to transcriptional dysregulation, mitochondrial dysfunction, and neuronal loss. Sirtuin 1 (SIRT 1), a NAD+-dependent deacetylase, can be neuroprotective in gene expression. Selective SIRT1 inhibition may restore transcriptional balance, making it a potential therapeutic strategy. To identify natural product-derived SIRT1 inhibitors with potential therapeutic relevance for HD. Selisistat (EX-527), a selective SIRT1 inhibitor, was used as a reference to retrieve 1401 st
