Spectral-Spatial EPR Imaging as a Single-Window Probe for Both Macro-Distribution and Micro-Architecture of Lithium Deposits.

An ideal diagnostic for metallic lithium anodes should be noninvasive and can concurrently probe from submicrometer dendrites to millimeter-scale plating distribution due to the multiscale differences in deposit architectures. Here we show that spectral-spatial electron paramagnetic resonance imaging (EPRI) fulfils this need by resolving the Dysonian line shape of Li deposits at every pixel of the electrode plane. The spectral parameters of asymmetry ratio A/B, peak-to-peak line width ΔBpp and double integral (DI) are correlated to microstructure thickness, packing compactness and deposit quan…

The ER/Golgi Protein FNDC3B Facilitates Climbing Fibre to Purkinje Cell Synapse Elimination in the Developing Mouse Cerebellum.

Synapse elimination during development is crucial for refining neural circuits by removing excess synapses formed around birth. In the neona … (FNDC3B-cKO) mice, CF synapse elimination from P10 was impaired, and the extension of CFs along PC dendrites was reduced at P21. However, these phenotypes were recovered by P40. In contrast, parallel fiber-mediated excitatory synaptic inputs and inhibitory synaptic inputs to PCs were not affected in FNDC3B-cKO mice. These results suggest that FNDC3B facili …

Review of the Applications of Density Functional Theory Calculations in Lithium-Sulfur Batteries.

Lithium-sulfur (Li-S) batteries, featuring a superior theoretical energy density of 2600 Wh/kg and a high specific capacity of 1675 mAh/g fo … early identified. In addition, DFT calculations also play a vital role in the inhibition of lithium dendrites and the regulation of the solid-electrolyte interphase (SEI) film. This review suggests that DFT calculations could provide more applications in the development of Li-S batteries.

Perineuronal Net and Inhibitory Synapse Remodeling on Striatal Fast-Spiking Interneurons by Chronic Alcohol Exposure.

BACKGROUND: Alcohol use disorder is characterized by persistent drinking in the face of negative consequences. Such inflexible drinking requ … not glutamatergic, synapses onto dorsolateral striatum fast-spiking interneuron somata and proximal dendrites where perineuronal nets (PNNs), a subdivision of the extracellular matrix, are enriched. We found that chronic ethanol exposure degraded these PNNs and that enzymatically degrading PNNs similarly reduced GABAergic transmission onto dorsolateral stri …

The kinetochore proteins Ndc80 and Dsn1 are required for survival of postmitotic neurons.

Dsn1 and Ndc80 are essential proteins of the kinetochore complex and required for chromosome segregation in dividing cells and for regulatin … eted. Similarly, whereas the synaptogenic function involves regulation of microtubules in axons and dendrites, a nucleus-localized Dsn1 is sufficient to rescue the viability of neurons from which Dsn1 has been deleted. Thus, postmitotic neurons retain a nuclear requirement for components of the kinetochore in order to prevent apoptosis.

Population-level geometric filtering generates motion-coherence selectivity in the retina.

Object-motion-sensitive (OMS) circuits must ignore self-motion while remaining sensitive to independently moving objects. We show that the m … ibition and voltage-gated sodium channels allow this bipartite arbor to multiplex signals: Proximal dendrites compute locally, whereas distal neurites integrate over hundreds of micrometers. A population model built from this architecture predicts a daisy-shaped inhibitory surround for OMS ganglion cells (W3/Ultra-High-Definition), with a circular near-surr …

Procognitive and neurotrophic benefits of α5-GABA-A receptor positive allosteric modulation in a β-amyloid deposition mouse model of Alzheimer's disease pathology.

Reduced somatostatin (SST) and SST-expressing GABAergic neurons are well-replicated findings in Alzheimer's disease (AD) and are associated with cognitive deficits. SST cells inhibit pyramidal cell dendrites through α5-GABA-A receptors (α5-GABAA-R). α5-GABAAR positive allosteric modulation (α5-PAM) has procognitive and neurotrophic effects in stress and aging models. We tested whether α5-PAM (GL-II-73) could prevent cognitive deficits and neuronal spine loss in early stages, and reverse them in late stages of β-amyloid deposition in the 5xFAD model (N = 48/study; 50 % female). Acute administra…

Expression of human Ras-related protein Rab39B variant T168K in Caenorhabditis elegans leads to motor dysfunction and dopaminergic neuron degeneration.

Human RAB39B gene is related to familial early-onset Parkinson disease. In early adulthood, men with the RAB39B c.503C > A (Thr168Lys, p. T168K) mutation develop typical tremor, bradykinesia, and alpha-synuclein accumulation. We investigated the pathological mechanism of RAB39B T168K in a Caenorhabditis elegans model. In early adult C. elegans, RAB39B T168K led to dopaminergic neuron degeneration that presented as disrupted dendrites and blunt neuronal cells. Abnormal dopamine secretion was inferred from a decline in motor function and a positive basal slowing phenotype. Dopamine-associated te…

Inhibition of Melanosome Transport by Inducing Exon Skipping in Melanophilin.

Exon skipping is an efficient technique to inhibit specific gene expression induced by a short-sequence peptide nucleic acid (PNA). To date, there has been no study on the effects of PNA on skin pigmentation. In melanocytes, the tripartite complex is responsible for the transport of mature melanosomes from the nucleus to the dendrites. The tripartite complex is composed of Rab27a, Mlph (Melanophilin), and Myosin Va. Defects in the protein Mlph, a melanosome transport-related protein, are known to cause hypopigmentation. Our study shows that Olipass peptide nucleic acid (OPNA), a cell membrane-…

Role of the Osteocyte in Musculoskeletal Disease.

PURPOSE OF THE REVIEW: The purpose of this review is to summarize the role of the osteocyte in muscle atrophy in cancer patients, sarcopenia, spinal cord injury, Duchenne's muscular dystrophy, and other conditions associated with muscle deterioration. RECENT FINDINGS: One type of bone cell, the osteocyte, appears to play a major role in muscle and bone crosstalk, whether physiological or pathological. Osteocytes are cells living within the bone-mineralized matrix. These cells are connected to each other by means of dendrites to create an intricately connected network. The osteocyte network ha…

Defective neurite elongation and branching in Nibp/Trappc9 deficient zebrafish and mice.

Loss of function in transport protein particles (TRAPP) links a new set of emerging genetic disorders called "TRAPPopathies". One such disor … es of neurites and growth cones. This deficiency also impaired elongation and branching of neuronal dendrites and axons, without significant effects on neurite initiation or neural cell number/types in embryonic and adult brains. The positive correlation of TRAPPII stability and neurite elongation/branching suggests a potential role for TRAPPII in regulatin …

Caldendrin represses neurite regeneration and growth in dorsal root ganglion neurons.

Caldendrin is a Ca2+ binding protein that interacts with multiple effectors, such as the Cav1 L-type Ca2+ channel, which play a prominent role in regulating the outgrowth of dendrites and axons (i.e., neurites) during development and in response to injury. Here, we investigated the role of caldendrin in Cav1-dependent pathways that impinge upon neurite growth in dorsal root ganglion neurons (DRGNs). By immunofluorescence, caldendrin was localized in medium- and large- diameter DRGNs. Compared to DRGNs cultured from WT mice, DRGNs of caldendrin knockout (KO) mice exhibited enhanced neurite rege…

Degradation of Perineuronal Nets in the Cerebellar Interpositus Nucleus Ameliorated Social Deficits in Shank3-deficient Mice.

Perineuronal nets (PNNs) are structures that contain extracellular matrix chondroitin sulfate proteoglycan and surround the soma and dendrites of various neuronal cell types. They are involved in synaptic plasticity and undertake important physiological functions. Altered expression of PNNs has been demonstrated in the brains of autism-related animal models. However, the underlying mechanism is still unknown. In this study, we demonstrated that the PNNs in the cerebellum are involved in modulating social and repetitive/inflexible behaviors in Shank3B-/- mice, an established animal model of aut…
