Synergistic regulation of fibroblast phenotype by high-dose estrogen and matrix stiffness: implications for intrauterine adhesion therapy†.

Intrauterine adhesion (IUA) is a leading cause of uterine infertility and often recurs post surgery due to limited treatments. Although estrogen is widely used to prevent recurrence, its efficacy remains unclear. A key characteristic of IUA is the excessive production of extracellular matrix (ECM) by activated myofibroblasts, which disrupts normal endometrial mechanics. Whether estrogen and ECM stiffness synergistically promote fibroblast to myofibroblast transition remains unknown. This study investigates why estrogen shows unsatisfactory efficacy in preventing IUA recurrence. We cultured cel

Endometrial receptivity in polycystic ovary syndrome: altered biomarkers and underlying mechanisms.

Polycystic ovary syndrome (PCOS) is a common endocrine disorder characterized by hyperandrogenism and ovulatory dysfunction. Women with PCOS are at an increased risk for adverse pregnancy outcomes, such as miscarriage and preterm birth. Endometrial receptivity plays a pivotal role in embryo implantation. However, there has been limited review or discussion regarding potential alterations in endometrial receptivity in PCOS patients, the nature of these changes, and their underlying mechanisms. In this review, we aim to summarize the alterations in endometrial receptivity associated with PCOS, h

Noninvasive Imaging Diagnostics for Endometriosis.

Endometriosis is a prevalent gynecological condition affecting approximately 10% of women of reproductive age and up to 50% of those with infertility. It is characterized by the presence of endometrial-like tissue outside the uterine cavity. Despite its high prevalence, diagnosis is often delayed by up to 10 years from symptom onset, partly due to the historical lack of accurate, noninvasive diagnostic tools. Although major reproductive societies continue to define videolaparoscopy as the gold standard for diagnosing endometriosis, it does not reliably predict disease extent. Consequently, onc

Decidualization potential of endometrial mesenchymal stem cells and their role in reproductive health.

The establishment of a successful pregnancy depends on proper embryo development and coordinated endometrial differentiation, particularly through the process of decidualization. Mesenchymal stem/stromal cells (MSCs), especially those derived from endometrial tissue (eMSCs) and menstrual fluid (MenSCs), have emerged as critical players in regulating decidualization, trophoblast invasion, angiogenesis, and immune modulation at the maternal-fetal interface. These cells exhibit potent immunoregulatory functions, underscoring their role in establishing a tolerant yet responsive environment for imp

Advances in the Role of Endometrial Microbiota Alterations in the Pathogenesis of Endometriosis.

The uterine cavity was formerly considered sterile; however, over recent years, researchers have identified the presence of endometrial microbiota. An imbalance in the endometrial microbiota, or dysbiosis, has been shown to be associated with a variety of gynecological diseases. Endometriosis (EM) is a chronic and inflammatory gynecological disease that affects 6-10% of all women of reproductive-age. Previous studies suggested that endometrial dysbiosis can participate in the development of EM by generating an inflammatory state, leading to disorders of immune homeostasis and thus creating an

Vaginal and endometrial microbiota dysbiosis in patients with chronic endometritis: a systematic review and meta-analysis.

BACKGROUND: Chronic endometritis (CE), a persistent inflammatory condition of the endometrial lining, is clinically linked with adverse reproductive outcomes. It is currently hypothesized to be associated with infection, and is often treated with broad-spectrum antibiotics. However, the specific microbial alterations remain poorly defined due to heterogeneous findings. METHODS: PubMed, Web of Science, Medline, Embase, Cochrane Library, and Scopus were searched for studies published up to July 2025. Studies were included if they compared CE patients to non-CE controls and analyzed vaginal or e

Real-Time in Vivo Monitoring of Circulating Endometrial Cells: Uncovering Systemic Dissemination and Temporal Fluctuations in Endometriosis.

Endometriosis is a common gynecological disorder characterized by the ectopic growth of endometrium-like tissue outside the uterine cavity and is associated with pelvic pain, infertility, and reduced quality of life. Although several theories have been proposed, the mechanisms by which endometrial cells disseminate and establish ectopic lesions remain incompletely understood. Here, we developed a dual-color pseudo-menstrual mouse model that enables real-time in vivo tracking of circulating endometrial cells (CECs) while distinguishing their tissue origin as uterine-derived or lesion-derived. U…

Extracellular matrix deficiency contributes to irregular uterine bleeding following progestin therapy for adenomyosis†.

Adenomyosis is a common gynecological disorder that causes heavy menstrual bleeding and chronic pelvic pain. Progestin-based therapy is widely used for long-term management, but irregular uterine bleeding frequently occurs and severely affects patients' quality of life. The mechanisms underlying this side effect remain unclear. In this study, we performed proteomic sequencing on endometrial tissues from adenomyosis patients before progestin treatment. We found that patients who later developed irregular bleeding exhibited significantly reduced extracellular matrix and collagen levels. Cell exp

Functional Analysis of the Histidine N-Methyltransferase SETD3 in Endometriosis.

Endometriosis is a disease associated with pain symptoms and reduced fertility, characterized by the presence of endometrial tissue outside the uterus. SETD3 is an actin-specific histidine N-methyltransferase that regulates actin stability and flexibility. Here, we investigate the effects of altered SETD3 expression on cytoskeletal function and endometriotic cell motility. SETD3 expression in endometriotic lesions was analyzed using EndometDB, and in uteri of the superfertile Dummerstorf mouse line FL1 by RT-qPCR. The functional impact of siRNA-mediated SETD3 depletion on endometriotic 12Z cel

The correlation between maternal expression of miR-146b and repeated failure of frozen-thawed embryo transfer pregnancy.

Frozen-thawed embryo transfer (FET) failure rates still account for a considerable percentage in assisted reproductive technology (ART) treatment, which seriously hinders the reproductive needs of infertile families. This study investigates the potential role of miR-146b level in repeated failures of FET. 175 patients with repeated implantation failure (RIF) after FET (the RIF population [RIFP]) and 155 patients with successful pregnancy after FET (SP) (the SP population [SPP]) were enrolled. The plasma miR-146b level was measured by RT-qPCR. The ROC curve was utilised to evaluate the predicti

BMAL1 Dysregulation as a Contributing Mechanism Linking Obesity to Oocyte and Endometrial Dysfunction in IVF.

Obesity affects nearly one in three women of reproductive age worldwide and consistently reduces success rates in in vitro fertilization, yet the molecular basis for this reduction remains fragmented across separate lines of evidence. Circadian clock genes, particularly BMAL1, orchestrate metabolic and reproductive physiology through transcription-translation feedback loops present in adipose tissue, ovarian granulosa cells, and endometrial stroma. Adiposity-driven metabolic shifts, including altered PPAR-γ signaling and reduced glutamine-methionine uptake, degrade BMAL1 expression and flatten

The Molecular Pathology of Morules in Endometrial Carcinoma: Biological Significance and Clinical Implications.

Morules were ordinally described as mulberry-like clusters of endometrial metaplasia with the potential to differentiate toward true squamous metaplasia (SqM). Although both morular and SqM lesions are categorized under "endometrial carcinoma (Em Ca) with squamous differentiation" in the WHO classification, they represent biologically distinct entities. Notably, morules are strongly associated with CTNNB1 (β-catenin) mutations. This review summarizes the molecular mechanisms driving the differentiation of Em Ca cells toward both morular and SqM phenotypes. Morules are frequently identified in

Novel screening approach for stem cell selective inhibitors and their possible translational therapeutic potential for endometriosis.

Endometriosis is an estrogen-dependent benign disease characterized by growth of the endometrial tissue outside the uterine wall. Several reports suggest the possibility of the pathogenesis and recurrence of endometriosis being related to functions of stem/progenitor cells of the endometrium. The drawback of the widely used method of using Hoechst 33342, a fluorescent dye, to collect stem cell-like populations, is the requirement of an ultraviolet (UV) excitation source not commonly provided on standard flow cytometers. Here, we aimed to overcome this hurdle by establishing a novel method that…

Proteomics Analysis of Porcine Endometrial Cell-Derived Extracellular Vesicles Involved in Embryo Attachment.

Maternal-embryo interactions play a critical role in early mammalian development, with extracellular vesicles (EVs) playing a key role in intercellular communication. Recent studies have focused on the mechanisms by which maternal-derived factors, such as RNA, proteins, and metabolites influence gap junctions, EVs, and direct cell-to-cell interactions, contributing to embryonic development. In this study, using a proteomics approach, we investigated the impact of EVs secreted from porcine endometrial cells (pEECs) and their protein cargoes on embryonic development. We characterized EVs isolate
