Nonlinear Strain Engineering via Meta-Material Substrates for Flexible Sensors With Simultaneously Enhanced Sensitivity and Wide Sensing Range.

Flexible strain sensors face an unresolved trade-off between sensitivity and sensing range. We present a meta-material-based sensor platform (meta-sensor) that overcomes this by engineering local strain input to the sensing element. A strain-stiffening curved spiral structure and a uniform-stiffness kirigami-patterned structure connected in series produce nonlinear strain partitioning: before mechanical transition, global strain concentrates at the sensor interface to amplify sensitivity; after transition, strain redistributes to extend sensing range. Integrating a kirigami crack-based sensor …

Engineering host‑defense peptides enhanced by artificial intelligence and nano delivery systems to overcome biofilms and antimicrobial resistance.

Infections caused by bacteria pose a risk to humanity as drugs become increasingly ineffective as resistance to bacterial strains emerge along with biofilm and persister formation. This review critically evaluates host defense peptides, rational design strategies that have guided next-generation antimicrobial peptide (AMP) discovery, and their current limitations. We also highlight optimization approaches including sequence engineering and chemical modification, synergistic combinations of antibiotics or adjuvants, and nanoscale delivery platforms that enhance stability, targeted delivery, and…

Outer membrane engineering through lipid A modification allows modulation of Burkholderia pseudomallei membrane permeability and innate immune responsivity.

Burkholderia pseudomallei causes the tropical disease melioidosis. Host mortality primarily results from sepsis-related complications and associated cytokine release. Lipopolysaccharide (LPS), a major virulence factor and mediator of sepsis, is modified by B. pseudomallei in response to external stimuli. LpxO and PagL are lipid A-modifying proteins encoded by the B. pseudomallei genome to decrease recognition by host defenses. The contribution of lpxO and pagL-dependent lipid A modifications to outer membrane permeability and host response has not been clearly demonstrated. Mono-phosphorylated

Spin-State Engineering of Iron Phthalocyanine D-Orbitals via Atomic Fe-N4 Coupling for Enhanced Oxygen Reduction Reaction.

Since the properties of electron transfer and orbital interactions in oxygen electrocatalysts are highly spin-dependent, reaction kinetics and thermodynamics are very sensitive to the spin configuration. However, understanding the spin-related origin of catalytic activity in heterogeneous molecular electrocatalysts still remains challenging. Herein, a molecular-atomic coupled catalyst is constructed by integrating iron phthalocyanine (FePc) molecules with Fe-N4 atomic sites anchored on nitrogen-doped carbon nanotubes (FePc-Fe-NCNT). The strong electronic coupling between FePc and the Fe-N4-con

Aggregation Engineering of Toluene-Processed Acceptor Layer Enables Over 19% Efficiency of Air-Blade-Coated Organic Solar Cells.

Understanding the unique features of photovoltaic materials in high-performance blade-coated organic solar cells (OSCs) is critical to narrow the device performance difference between spin-coating and blade-coating methods. In this work, it is clarified that the molecular packing of acceptor and molecule-solvent interaction plays an essential role in determining the photovoltaic performance of blade-coated layer-by-layer OSCs. It is demonstrated that the unique dimer packing feature of L8-BO-4Cl can lead to lower excited energy (∆ES1) and dominant J-aggregates in the blade-coated film compared

Engineering Spatially Adjacent Sites with Synergistic Spin Polarization Effect to Promote Sulfur Redox Kinetics in Lithium-Sulfur Batteries.

The kinetics of sulfur catalytic reactions serve as a fundamental cornerstone for maximizing both the capacity and stability of lithium-sulfur batteries (LSBs). Recently, investigations into Li-S catalysts predominantly center around the optimization of energy levels to bolster adsorption and catalytic conversion processes. However, a significant oversight persists, as the profound influence of electron spin states on charge transfer dynamics and orbital interactions is frequently neglected. Herein, NH4MnF3 is synthesized as a sulfur host due to the strongly polarized Mn-F bonds, which generat

Semi-rational engineering of Issatchenkia terricola acetaldehyde dehydrogenase for enhanced thermostability and catalytic efficiency in detoxification of aldehydes.

Acetaldehyde dehydrogenase is crucial for detoxifying aldehydes in pharmaceutical, food, and biofuel industries. We enhanced Issatchenkia terricola ALDH (ist-ALDH) thermostability and catalytic activity toward aliphatic and aromatic aldehydes via molecular engineering. The combinatorial mutant H62G/D240P/F429Vshifted the optimal temperature from 35 °C to 55 °C. It had better thermal stability with a half-life (t1/2) of 315.07 ± 4.24 min at 40 °C, 14.41-fold that of the wild-type (WT). Kinetic studies showed higher catalytic efficiency for both aliphatic and aromatic aldehydes. H62G/D240P/F429V…

Engineering virus-like particles for safe and versatile modeling of SARS-CoV-2 host interaction and immune escape.

The continuous evolution of SARS-CoV-2 poses global health challenges. A safe, rapid, and versatile method for assessing functions of Spike protein mutations in ACE2 receptor binding and immune evasion would be highly valuable. To address this, we engineered a transcription- and replication-competent virus-like particle (trVLP) derived from the Sindbis virus, pseudotyped with the SARS-CoV-2 receptor binding domain (RBD). This trVLP exclusively propagates in BHK-21 cells engineered to express both RNA replicase and human ACE2, providing a controllable, safe model of SARS-CoV-2 RBD-ACE2 interact

A Reverse Engineering Approach to Optimize Chemical Synergy Between Target and Phenotype: Bridging the Cancer and Malaria Indications.

Linking a drug's mechanism of action to a disease-related phenotype is the greatest challenge in pharmaceutical research. Target-based and phenotype-based screening are the two basic tools to identifying drug candidates which display efficiency in triggering disease-related phenotype through a known Mechanism of Action (MoA) via a predefined primary target. In this paper, we present a reverse engineering approach which identify drug candidates from active compounds in phenotypic-based assay while bypassing the biochemical assay on the primary target. This important information is deduced impli…
