Heterogeneous cerebral glucose metabolism in normal pressure hydrocephalus.

The regional cerebral metabolic rate for glucose (rCMRglu) has never been investigated in large consecutive groups of patients with normal pressure hydrocephalus (NPH), a potentially treatable form of dementia with an unpredictable outcome after shunt surgery. Using PET and 18F-2-fluorodeoxyglucose, rCMRglu was studied in 18 patients who fulfilled hydrodynamic criteria for NPH and in whom a biopsy of the frontal cortex was obtained. When compared with an age matched group of 11 healthy subjects, the patients with NPH showed a significant rCMRglu reduction in all cortical and subcortical region

Quantitative relation between myocardial viability and improvement in heart failure symptoms after revascularization in patients with ischemic cardiomyopathy.

BACKGROUND: Studies of patients with coronary artery disease and left ventricular dysfunction have shown that preoperative quantification of myocardial viability may be clinically useful to identify those patients who will benefit most from revascularization both functionally and prognostically. However, the relation between preoperative extent of viability and change in heart failure symptoms has not been documented carefully. We assessed the relation between the magnitude of improvement in heart failure symptoms after coronary artery bypass surgery (CABG) and the extent of myocardial viabili

The cerebral metabolism of glucose and oxygen measured with positron tomography in patients with mitochondrial diseases.

Cerebral oxygen and glucose metabolism have been studied in 8 patients with biochemically defined mitochondrial myopathies and 7 normal control subjects using positron emission tomography. Four patients had myopathy alone and 4 had predominantly central nervous system (CNS) disease. Cerebral oxygen and glucose metabolism were measured concurrently in the two groups of patients with 15O, 11C and 18F labelled tracers and positron emission tomography (PET). Patients with major CNS disease showed an uncoupling of glucose and oxygen metabolism when compared with patients without cerebral disease an

[Perifusion of monolayer-cultured B cells of the infant rat: a comparative study of the effects of 2-deoxy-2-fluoroglucose and iodoacetic acid].

Cell culture techniques for monolayer islets of 3-week-old rat pancreases and the responsiveness of B cells are described. In this procedure, whole pancreatic tissues from 3-week-old rats were enzymatically dispersed and then cultured in a medium with 5.5 mM glucose plus 1 mM 2-deoxy-2-fluoroglucose or with 5.5 mM glucose following a 3-day exposure to a medium with 5.5mM glucose plus 5 microM iodoacetic acid. The use of 2-deoxy-2-fluoroglucose or iodoacetic acid allowed a selective deletion of fibroblasts, yielding large clusters that consisted mostly of islet cells. The immunocytochemical eva

Glucose utilization by intracranial meningiomas as an index of tumor aggressivity and probability of recurrence: a PET study.

Seventeen patients with intracranial meningiomas were studied with positron emission tomography and fluorine-18-2-fluorodeoxyglucose (PET-FDG) to assess the glucose utilization of these tumors. Four meningiomas followed for 3-5 years after PET-FDG and surgery showed no evidence of recurrence. These tumors had significantly lower glucose utilization rates (1.9 mg/dl/min +/- 1.0) than 11 recurrent or regrowing meningiomas (4.5 mg/dl/min +/- 1.96) (P less than .01). The glucose metabolic rates of meningiomas correlated with tumor growth, as estimated from changes in tumor size on repeated compute

Cerebellar glucose consumption in normal and pathologic states using fluorine-FDG and PET.

We studied cerebellar metabolism in 118 subjects including young and elderly controls and patients suffering from stroke, supratentorial brain tumor and Alzheimer's disease using fluorine-18 fluorodeoxyglucose ([18F]FDG) and position emission tomography (PET). Alzheimer's disease and normal aging did not alter mean cerebellar metabolism. In stroke and tumor mean cerebellar metabolism was lower in the hemisphere contralateral to the supratentorial lesion. In tumor bilaterally significant reductions in absolute cerebellar metabolism also were noted, unlike stroke. Primary sensory stimulation did

[Perfusion of monolayer-cultured B cells of the neonatal rat pancreas--effect of 2-deoxy-2-fluoroglucose].

The effects of 2-deoxy-2-fluoroglucose in promoting the function of neonatal rat pancreatic B cells in vitro are presented. Functional studies of insulin secretion were carried out by perifusion procedures. At day 0 of culture, B cells showed a minimal monophasic insulin secretion in response to a 16.7 mM single dose of glucose, whereas in the presence of 10 microM forskolin or 1 mM 3-isobutyl-1-methylxanthine, the same dose of glucose stimulated insulin secretion in a biphasic fashion. In contrast, after 7 days of culture in medium supplemented with 1 mM 2-deoxy-2-fluoroglucose, B cells showe

Superior mucoepidermoid carcinoma detection with ⁶⁸Ga-FAPI PET/CT reflects FAP-α expression and MAML2 fusion status.

This study evaluated the diagnostic potential of the FAP-targeted tracer ⁶⁸Ga-FAPI PET/CT in mucoepidermoid carcinoma (MEC), demonstrating its superior sensitivity over ¹⁸F-FDG PET/CT in detecting primary tumors (100% vs. 85.7%) and metastatic lesions (33 lymph nodes and 19 distant metastases), with significantly higher SUVmax and TBR values. High-grade MEC exhibited elevated stromal FAP-α expression, strongly correlating with ⁶⁸Ga-FAPI uptake metrics (r = 0.91–0.96, p < 0.01), while MAML2-negative tumors showed amplified FAP activity, suggesting stromal activation-driven aggressive behavior.

Brown Adipose Tissue and Metabolic Markers Differ Between Greenlanders and Danes With Cold Activation.

CONTEXT: Brown adipose tissue (BAT) is a thermogenic tissue that converts energy into heat in response to cold exposure, and is therefore a promising target for improving cardiometabolic health. Habitually cold-exposed Arctic people represent a unique model of metabolic adaptation to cold. OBJECTIVE: We investigated cold-induced BAT activation in Greenlanders in comparison to Danes. METHODS: A comparative crossover study was conducted of 13 Greenlanders and 11 Danes, with 2 experimental sessions conducted 2 weeks apart: A, a thermoneutral session; and B, a cooling session. Prognostic Value of Fibroblast Activation Protein-Directed PET Imaging in Pleural Mesothelioma.

High expression of fibroblast activation protein (FAP) has been associated with inferior survival in several tumor entities. Novel 68Ga-radiolabeled FAP inhibitors (68Ga-FAPIs) allow noninvasive measurement of FAP, which enables the development of prognostic imaging parameters from 68Ga-FAPI PET/CT. In this study, we compared the prognostic value of 68Ga-FAPI-46 with 18F-FDG PET in a cohort of patients with malignant pleural mesothelioma from the FAPI PET observational trial (NCT04571086). Methods: Between May 2020 and January 2024, 49 patients with suspected or proven malignant mesothelioma w

Standardized FDG-PET interpretation during CAR T-cell therapy in R/R DLBCL: a DESCAR-T study.

PURPOSE: CAR T-cell therapy has changed the management of relapsed/refractory (R/R) Diffuse large B-cell lymphoma (DLBCL). Fluorodeoxyglucose Positron Emission Tomography Computed Tomography (PET/CT) plays a central role in lymphoma response assessment, but its prognostic use remains insufficiently standardized in this setting. METHODS: Patients from the French DESCAR-T registry treated in third line or beyond with commercial anti-CD19 CAR T-cells in real-life, and having centrally reviewed PET/CT before infusion, and at one month (M1) or three months (M3) post-infusion were included. Beyond FDG: Serial 68 Ga-FAPI PET/CT for Staging and Treatment Response Assessment in Metastatic Pleomorphic Lobular Breast Cancer.

68 Ga-FAPI PET/CT has demonstrated higher sensitivity than 18 F-FDG PET/CT in breast cancer, particularly in invasive lobular carcinoma. However, its role in treatment response assessment remains limited. We report a case of recurrent metastatic pleomorphic lobular breast cancer in which baseline 68 Ga-FAPI PET/CT revealed extensive regional and distant disease underestimated by 18 F-FDG PET/CT. Serial follow-up FAPI PET/CT after initiation of olaparib demonstrated marked response with near complete resolution of disease, highlighting the potential utility of FAPI PET/CT for response assessmen

Metabolic Tumor Area: A Clinically Practical Prognostic Marker for Improved Risk Stratification in IPI Non-High-Risk DLBCL.

Metabolic tumor volume (TMTV) is a core 18F-FDG PET/CT prognostic marker for diffuse large B-cell lymphoma (DLBCL), but its complex measurement and limited standardization restrict clinical utility. Metabolic tumor area (MTA) features simple operation and good reproducibility, holding potential as a practical alternative to TMTV. This retrospective study aimed to validate MTA's substitutability and explore its prognostic value in DLBCL risk stratification. A total of 295 newly diagnosed DLBCL patients were enrolled. TMTV and MTA were measured via semi-automatic segmentation (41% SUVmax thresho
