Hepatic IFRD1 suppresses metabolic dysfunction-associated fatty liver disease via GLUD1/α-KG axis.

The mechanisms underlying metabolic remodeling in metabolic dysfunction-associated steatotic liver disease (MASLD) remain unclear. Targeting the process of de novo lipogenesis (DNL) in the liver has the potential to mitigate MASLD. Here we show that interferon-related developmental regulator 1 (IFRD1) expression negatively correlates with MASLD/metabolic-associated steatohepatitis (MASH) progression in human liver tissues. In multiple mouse models, Ifrd1-/- mice exhibit an exacerbated MASLD phenotype, while hepatocyte-specific IFRD1 expression suppresses MASH progression. Mechanistically, IFRD

The transcriptional cortical response to daily torpor in Phodopus sungorus.

Torpor is an essential evolutionary strategy that allows mammals to conserve resources under extreme environmental conditions. Although the hypothalamus is a key regulator of systemic acclimatisation to energy scarcity and reduced temperature, it remains unclear how the cortex, with its high metabolic demand, endures this state. Here, we performed RNA sequencing on neuronal and non-neuronal nuclei isolated from the cortex of Djungarian hamsters (Phodopus sungorus) across different seasonal and metabolic states. We found that non-neuronal cells primarily adjust during the seasonal transition fr

cnnm-5 knockdown improves proteostasis of mutant Huntingtin protein in C. elegans.

Huntington's disease (HD) is an age-related neurodegenerative disease associated with the aggregation of mutant Huntingtin protein (mHTT). It is theorized that prevention or clearance of these aggregates through autophagy and the ubiquitin proteasome system (UPS) protects neurons from degeneration. Using a C. elegans model of HD, a small reverse genetic screen of 100 random genes on Chromosome 3 identified cnnm-5 as a genetic modifier of mHTT accumulation. During development, loss of cnnm-5 by RNAi ( cnnm-5 i) protects against mHTT accumulation, implicating cnnm-5 as a negative regulator of pr

Nanodelivery Improves Insecticidal Activity of Hexaflumuron against Apolygus lucorum by Inhibiting the Synthesis of Insect Cuticle Protein.

Given the limitations of conventional pesticides, sustainable pest control alternatives are urgently needed. Hexaflumuron (HE), an insect growth regulator, is a potential candidate, yet its efficacy requires enhancement. This study developed a star polycation (SPc)-based nanocarrier to improve HE delivery. The HE/SPc complex was formed via electrostatic and van der Waals interactions, reducing the particle size from 751.21 to 174.62 nm. Compared to HE alone, HE/SPc complex enhanced retention and coverage for effective foliar application, and significantly enhanced insecticidal activity against

MRPL37 promotes hepatocellular carcinoma progression through modulating mitochondrial energy metabolism.

Mitochondrial ribosomal proteins of the large subunit (MRPLs) are critical for mitochondrial function and cellular energy metabolism. However, the role of the MRPL family in hepatocellular carcinoma (HCC) remains poorly understood. Here, we leveraged The Cancer Genome Atlas (TCGA) liver cancer data to develop a subtype classification and prognostic model based on the MRPL family genes, identifying MRPL37 as a key gene associated with HCC progression. Clinically, MRPL37 upregulation is associated with HCC progression and poor prognosis. Functionally, MRPL37 knockdown significantly inhibits HCC

miR-932 Suppresses the Expression of Germline-Specific vasa in Somatic Drosophila Testis Hub Cells.

Germline cells are specified early in embryogenesis and are encapsulated by somatic cells to form the gonads (testis or ovary). This development requires genes with expression restricted to germline cells, such as the DEAD-box RNA helicase Vasa, an evolutionarily conserved protein exclusively expressed in the germline of the testis. However, the mechanisms underlying germline-specific expression remain poorly understood. To identify microRNAs that function in the somatic cells of the testis, we employed the binary Gal4/UAS expression system, which enables the expression of UAS-microRNA sponges

The cell cycle regulator PLK1 promotes murine melanoma progression by regulating the transcription factor BACH1.

Polo-like kinase 1 (PLK1), a critical cell cycle regulator, is associated with cancer progression and negatively correlates with patient survival in cutaneous melanoma based on clinical database analysis. In a melanoma mouse model induced by BRafCA mutation and Pten-deficiency, we observed that PLK1 overexpression mediated metabolic reprogramming to markedly accelerate tumor growth, promote metastasis, and shortened mice survival. Mechanistically, PLK1 stabilizes BTB domain and CNC homolog 1 (BACH1), which serves as a crucial transcription factor for genes involved in cancer metabolism and met

SETDB1 is critically required for uveal melanoma growth and represents a promising therapeutic target.

Metastatic uveal melanomas are highly resistant to all existing treatments. To identify actionable vulnerabilities, we conducted a CRISPR-Cas9 knockout screen using a library composed of chromatin regulators. We revealed that the lysine methyltransferase, SETDB1, plays a critical role in metastatic uveal melanoma cell proliferation and survival. Functionally, SETDB1 deficiency induces a DNA damage response, senescence-like state and growth arrest. Knockdown of SETDB1 is associated with a decreased expression of genes related to replication and cell cycle. Moreover, deficiency in CDC6, an essen

Double heterozygous RhAG mutations causing regulator-type Rhnull phenotype.

BACKGROUND: Rhnull is the rarest blood type worldwide; its scarcity makes compatible blood almost unobtainable once alloantibodies arise. OBJECTIVE: To define the genetic basis of regulator-type Rhnull in a healthy blood donor and translate the findings into transfusion guidance. METHODS: Serology, Sanger and long-read sequencing of RhD, RhCE and RhAG were performed in the donor and his family. RESULTS: The donor displayed Rhnull with normal RhD/CE genes but compound-heterozygous RhAG mutations: c.419 A > C (p.Gln140Arg, novel) and c.1108G > A (p.Gly370Arg). Father and son carry only c.419

A Hybrid compound H93 treats prostate cancer by directly binding UHRF1 and promoting protein dimerization.

UHRF1 is a pivotal epigenetic regulator bridging DNA methylation and histone modifications, frequently overexpressed in prostate cancer (PCa). However, no UHRF1-targeted therapeutics have advanced to clinical trials. Here, we report the development of H93, a hybrid small molecule integrating functional groups derived from NSC232003 and vorinostat, respectively. H93 exhibits potent anticancer activity in vitro, correlating with UHRF1 protein abundance, and co-crystallization studies confirm that H93 directly binding to the SRA domain of UHRF1. Intriguingly, H93 promotes UHRF1 dimerization, gene

The role and mechanism of TSC in kidney diseases: a literature review.

BACKGROUND: Tuberous sclerosis complex (TSC) is an autosomal dominant genetic disorder characterized by multisystem involvement, primarily caused by loss-of-function mutations in the TSC1 or TSC2 genes. TSC is a key integrator of metabolic signaling and cellular stress and has become an important regulator in several kidney diseases. TSC1 and TSC2 can be used not only as genetic markers for disease diagnosis, but also as potential immunotherapeutic targets for kidney disease. Recent studies on the pathogenesis of TSC may provide guidance for developing new treatment strategies for kidney disea

Autoimmunity-associated DIORA1 binds the MRCK family of serine/threonine kinases and controls cell motility.

Genetic association links disordered autoimmunity 1 (DIORA1) to numerous autoimmune rheumatic diseases, including systemic lupus erythematosus, Sjögren's disease, rheumatoid arthritis, polymyositis, and systemic sclerosis. However, its cellular function has remained unknown. Here, we identify the Myotonic Dystrophy Kinase-Related Cdc42-Binding Kinases (MRCK kinases) family of serine/threonine kinases-key regulators of actomyosin contractility and cell motility-as direct interactors of DIORA1. Through interaction mapping, we show that DIORA1 binds three distinct modules of MRCK kinases, includi

TYMS-ENOSF1 Dyskeratosis Congenita in a Patient With Ring Chromosome 18: A Case Report.

Dyskeratosis Congenita (DC) is a rare genetic syndrome due to variants in genes involved in telomeric regulation and maintenance, impacting multiple organ systems. We report a case of DC secondary to TYMS gene deletion in a patient with ring chromosome 18 and related partial monosomy 18p and 18q. TYMS encodes thymidylate synthase, and compound heterozygosity for loss of function variants in TYMS and a specific haplotype of its antisense regulator ENOSFI (enolase super family 1) causes digenic DC. The patient had physical and developmental features of 18p monosomy, including poor growth, feedin

Impaired Aire-dependent IFN signaling in the thymus precedes the protective autoantibodies to IFNα.

Recent studies have highlighted the role of the thymus in maintaining immune tolerance to type 1 interferons (T1 IFNs). Individuals with thymic abnormalities, such as autoimmune regulator (AIRE) gene mutations, frequently develop neutralizing autoantibodies to interferon-alpha (IFNα). Unlike mice, Aire-deficient rats develop robust autoantibodies to IFNα. Using this rat model, we show that Aire regulates the thymic expression of interferon-stimulated genes (ISGs), which occurs before developing anti-IFNα autoantibodies. In the periphery, we observed a widespread downregulation of ISGs across i

Viral proteins resolve the virus-vector conundrum during hemipteran-mediated transmission by subverting salicylic acid signaling pathway.

Hemipteran insects transmit viruses when infesting plants, during which vectors activate salicylic acid (SA)-regulated antiviral defenses. How vector-borne plant viruses circumvent these antiviral defenses is largely unexplored. During co-infections of begomoviruses and betasatellites in plants, betasatellite-encoded βC1 proteins interfere with SA signaling and reduce the activation of antiviral resistance. βC1 inhibits SA-induced degradation of NbNPR3 (Nicotiana benthamiana nonexpressor of pathogenesis-related genes 3), a negative regulator of SA signaling. βC1 does not bind directly to NbNPR

SMYD5 is a regulator of the mild hypothermia response.

The mild hypothermia response (MHR) maintains organismal homeostasis during cold exposure and is thought to be critical for the neuroprotection documented with therapeutic hypothermia. To date, little is known about the transcriptional regulation of the MHR. We utilize a forward CRISPR-Cas9 mutagenesis screen to identify the histone lysine methyltransferase SMYD5 as a regulator of the MHR. SMYD5 represses the key MHR gene SP1 at euthermia. This repression correlates with temperature-dependent levels of H3K36me3 at the SP1-locus and globally, indicating that the mammalian MHR is regulated at th

Mechanosensitive channel Piezo1 is an essential regulator in cell cycle progression of optic nerve head astrocytes.

Optic nerve head (ONH) astrocytes provide structural and metabolic support to neuronal axons in developmental, physiological, and pathological progression. Mechanosensitive properties of astrocytes allow them to sense and respond to mechanical cues from the local environment. We confirmed that ONH astrocytes express the mechanosensitive ion channel Piezo1 in vivo. By manipulating Piezo1 knockdown or overexpression in vitro, we found that Piezo1 is necessary but insufficient for ONH astrocyte proliferation. Loss of Piezo1 can lead to cell cycle arrest at G0/G1 phase, a possible mechanism involv

Oxycodone withdrawal induces HDAC1/HDAC2-dependent transcriptional maladaptations in the reward pathway in a mouse model of peripheral nerve injury.

The development of physical dependence and addiction disorders due to misuse of opioid analgesics is a major concern with pain therapeutics. We developed a mouse model of oxycodone exposure and subsequent withdrawal in the presence or absence of chronic neuropathic pain. Oxycodone withdrawal alone triggered robust gene expression adaptations in the nucleus accumbens, medial prefrontal cortex and ventral tegmental area, with numerous genes and pathways selectively affected by oxycodone withdrawal in mice with peripheral nerve injury. Pathway analysis predicted that histone deacetylase (HDAC) 1

MRSA lineage USA300 isolated from bloodstream infections exhibit altered virulence regulation.

The epidemic community-associated methicillin-resistant Staphylococcus aureus (CA-MRSA) USA300 lineage has recently become a leading cause of hospital-associated bloodstream infections (BSIs). Here, we leveraged this recent introduction into hospitals and the limited genetic variation across USA300 isolates to identify mutations that contribute to its success in a new environment. We found that USA300 BSI isolates exhibit altered virulence regulation. Using comparative genomics to delineate the genes involved in this phenotype, we discovered repeated and independent mutations in the transcript
