Primary yolk sac tumor of the urachus.

INTRODUCTION: Neoplasms originating from the urachus are rare. The most common urachal malignancy is adenocarcinoma, whereas extragonadal germ cell tumors, primarily of the urachus, are an extremely rare finding. OBJECTIVE: To describe a primary yolk sac tumor (YST) of the urachus in an adult. CASE REPORT: A 44-year-old woman presented with 6 months of pelvic pain associated with a sensation of progressive mass growth. At the time of tumor resection, the tumor was found to be attached by a pedicle to the dome of the bladder, with no injury to the adjacent organs. Pathological study showed a

Testicular degeneration in Huntington disease.

Huntington disease (HD) is an adult onset, neurodegenerative disorder that results from CAG expansion in the HD gene. Recent work has demonstrated testicular degeneration in mouse models of HD and alterations in the hypothalamic-pituitary-gonadal (HPG) axis in HD patients. Here, we show that HD patients have specific testicular pathology with reduced numbers of germ cells and abnormal seminiferous tubule morphology. In the YAC128 mouse model, testicular degeneration develops prior to 12 months of age, but at 12 months, there is no evidence for decreased testosterone levels or loss of GnRH neur…

Molecular basis of haemophilia A.

Technologies in molecular biology have greatly advanced the knowledge regarding the origin of haemophilia A and the physiology of the factor VIII (FVIII) protein. A variety of different mutations in the FVIII gene have been identified and their effects on the FVIII protein described. It has been shown that the frequency of haemophilia A is due to a high mutation rate predominantly in male germ cells. A significant proportion is originating de novo in early embryogenesis from somatic mutations, a finding that has implications for genetic counselling. The life-cycle of the FVIII protein and its …

Comparison between two HNK-1-related antibodies immunoreactivity (HNK-1-anti-leu 7 and anti-HNK-1/N-CAM) during rat cephalogenesis.

The fixation sites of two antibodies, HNK-1-anti-leu 7 and anti- HNK-1/N-CAM, were studied during visceral cephalogenesis in the rat. The labelling patterns of both antibodies were different. Anti-leu 7 staining was restricted to trunk neural crest cells, the peripheral nervous system, myotomes and premuscular cells, the eye rudiment and the olfactory vesicle. On the other hand, anti- HNK-1/N-CAM extended to the otocyst, several epithelia (including tooth germs) and precartilages. Anti-HNK-1/N-CAM staining of somites and cranial nerve ganglia was more precocious than anti-leu 7 labelling. We c

Precocious puberty associated with malignant thoracic teratoma and malignant histiocytosis in a child with Klinefelter's syndrome.

A 10-year-old boy with precocious puberty of 2 1/2 years' duration presented with a malignant thoracic teratoma with elevated levels of beta human chorionic gonadotropin (beta-HCG) and alpha fetoprotein (alpha FP). The mediastinal tumor was completely excised and adjuvant chemotherapy commenced. Within 3 weeks of commencing chemotherapy he developed hematologic abnormalities that, with the subsequent clinical illness, led to a diagnosis of malignant histiocytosis. Death occurred a few days after start of therapy for malignant histiocytosis. Cytogenic studies showed the somatic karyotype to be

Regulation of mRNA poly(A) tail length governs mammalian body plan formation in gastruloids.

The establishment of the body plan during gastrulation represents a hallmark of animal life. It emerges from the interplay of gene-regulatory programs and positional cues, yet how these signals are integrated post-transcriptionally remains largely unexplored. Here, we combine the scalability of mouse gastruloids with a single-cell CRISPR screening platform to functionally dissect germ layer specification at single-cell transcriptomic resolution. Focusing on post-transcriptional regulation, we systematically map drivers of mesodermal and endodermal fate and identify the deadenylase Cnot8. Loss

Choriocarcinoma presenting with cystic lung metastasis mimicking diffuse cystic lung disease.

Choriocarcinoma is a highly aggressive malignant germ cell tumour containing syncytiotrophoblasts that secrete beta-human chorionic gonadotropin, and it has a poor prognosis, with a dismal 5-year survival rate of <5%. It generally affects young individuals. Cystic lesions in the lung are uncommon in malignancies that can predispose patients to spontaneous pneumothorax. Multiple cystic lesions are less commonly seen in metastatic disease. Diffuse cystic lung diseases (DCLD) are a group of diseases characterised by cysts in the bilateral lung fields that are not necessarily evenly distributed. T

PIWI-Interacting RNAs in Cardiovascular Disease: From Epigenetic Regulators to Clinical Biomarkers and Therapeutic Targets.

PIWI-interacting RNAs (piRNAs) are a class of non-coding RNAs approximately 24-32 nucleotides (nt) in length. They are well known for silencing transposons and maintaining genomic integrity during germ cell development. Recent studies have detected specific piRNAs in cardiovascular-relevant tissues, including myocardial tissue, circulating samples, and distinct cardiac cell populations, as well as in various disease settings. However, the biogenesis and functions of somatic piRNA/P-element-induced wimpy testis proteins (PIWI) signaling in mammalian cardiovascular tissues remain incompletely un

Generation of two induced pluripotent stem cell lines (HIMRi006-A and HIMRi007-A) from Pompe patients with infantile and late disease onset.

Here we present the generation of HIMRi006-A and HIMRi007-A Pompe disease (PD) patient derived human induced pluripotent stem cell (hiPSC) lines. HIMRi006-A represents an infantile onset disease (IOPD) phenotype caused by a homozygous c.307 T > G mutation in the GAA gene. HIMRi007-A is characterized by heterozygous mutations c.-32-13 T > G/c.1716C > G and is associated with an adult onset of disease symptoms (LOPD). Both lines are generated via lentiviral expression of OCT4, SOX2, KLF4, and c-MYC. The lines display a typical embryonic stem cell morphology, express pluripotency markers, retain

Establishment of pluripotent stem cell line induced by PATL2 heterozygous mutation in patients with oocyte maturation defect-4.

Oocyte maturation defect-4 (OOMD4) is an autosomal recessive disease characterized by oocyte maturation arrest. On chromosome 15q21, the PATL2 gene is mutated, resulting in OOMD4 in either a homozygous or compound heterozygous form. Herein, the peripheral blood mononuclear cells (PBMCs) were obtained from a female patient who was heterozygous for OOMD4 due to a PALT2 gene mutation. Then we obtained the induced pluripotent stem cell (iPSC) by using episomal vectors with transcription factors for reprogramming. The teratoma assay revealed that the iPSC line exhibited pluripotency with the abilit

Mosaicism of a TCOF1 mutation in an individual clinically unaffected with Treacher Collins syndrome.

Treacher Collins syndrome (TCS) or mandibulofacial dysostosis is an autosomal dominant disorder of craniofacial development with 60% of its cases arising de novo. Other modes of inheritance such as autosomal recessive, gonadal mosaicism, and chromosomal rearrangement have also been proposed. This syndrome can result from TCOF1 gene mutations. In this study we identified a TCOF1 1408delAG heterozygous mutation in a patient with the clinical diagnosis of TCS. This same mutation was found in the clinically unaffected mother's leukocytes, hair root bulbs, buccal mucosa, urine, and stool. The mothe

Infundibulocystic basal cell carcinoma: a newly described variant.

A report by Tozawa and Ackerman (Am J Dermatopathol 9(6):474, 1987) upon a distinctive type of basal cell carcinoma with a particular type of follicular differentiation has stimulated lively correspondence in the literature of dermatopathology. The lesion in question presents itself clinically as a basal cell carcinoma on the face and histopathologically as a neoplasm typified by small size, sharp circumscription, and a unique combination of kinds of follicular differentiation, namely, follicular germs and infundibula. The principal point of contention concerning this neoplasm has been whether

Case Report: Ovarian dysgerminoma with pseudo-Meigs syndrome in a child.

Ovarian dysgerminoma is a rare pediatric germ cell tumor, and its association with pseudo-Meigs syndrome (PMS) is exceedingly uncommon. PMS is characterized by massive pleural effusions and ascites accompanying a pelvic mass, which often leads to clinical misdiagnosis. This report describes the case of a 15-year-old female patient who presented with abdominal distension, chest tightness, and significant pleural and ascitic effusions. Following surgical resection of the tumor, the effusions resolved rapidly. Postoperative pathology confirmed the diagnosis of ovarian dysgerminoma. The patient re

Establishment and characterization of induced pluripotent stem cell lines from individuals with Down syndrome and age-matched euploid donors.

Down syndrome (DS) is caused by trisomy 21 and is characterized by cognitive impairment, among other symptoms. Existing mouse models of DS do not fully recapitulate the genetic and phenotypic features of human trisomy 21. Here, we generated two induced pluripotent stem cell (iPSC) lines (ABCRIi003-A and ABCRIi004-A) from pediatric individuals with DS and two iPSC lines (ABCRIi002-A and ABCRIi005-A) from age-matched euploid donors. All generated lines displayed normal morphology, expressed key pluripotency markers, and demonstrated the ability to differentiate into the three germ layers. These

Simultaneous reprogramming and gene correction to generate six iPSC lines and isogenic controls from individuals with neurofibromatosis type 1.

Neurofibromatosis type 1 (NF1) is a neurodevelopmental disorder that affects ∼ 1: 2700 individuals (Lee et al., 2023) however the underlying pathogenic mechanisms are poorly understood. In this study, we performed simultaneous reprogramming and CRISPR-Cas9 genome editing to generate pluripotent stem cell (iPSCs) lines and their respective isogenic controls from six individuals with different pathogenic NF1 variants. All iPSC lines had a normal karyotype, were pluripotent and able to differentiate into the three embryonic germ layers. These iPSC lines are valuable pre-clinical models to investi

Generation of human induced pluripotent stem cell (iPSC) lines from two patients with BAG3 P209L myofibrillar myopathy-6.

As member of the chaperone-assisted selective autophagy (CASA) complex, BAG3 is important for the turnover of muscle proteins. Patients with a point mutation at position 626 in the BAG3 gene (p.P209L, c.626C>T, Chr.10q26) suffer from polyneuropathy and severe myofibrillar myopathy-6 (MFM6). The latter manifests as skeletal muscle dystrophy and restrictive cardiomyopathy. To model MFM6, we generated two non-isogenic human induced pluripotent stem cell (iPSC) lines from patients with this variant. Pluripotency analyses and germ layer differentiations were performed to check the iPSC quality. The

Generation of patient-derived and gene-corrected hiPSC lines from Hereditary Hemorrhagic Telangiectasia type 2 patients with ACVRL1 c.1042delG mutation.

Hereditary Hemorrhagic Telangiectasia type 2 (HHT2) is a vascular disorder caused by mutations in ACVRL1. We generated human induced pluripotent stem cell (hiPSC) lines from two HHT2 patients with a heterozygous 1 bp deletion in exon 7 of ACVRL1 (c.1042delG) by reprogramming skin fibroblasts. Gene-corrected isogenic hiPSCs were created using CRISPR-Cas9. All lines displayed normal karyotypes, expressed markers of the undifferentiated state, differentiated into all germ layers in vitro, and showed no off-target effects.
