The existence of viruses as obligate intracellular parasites with high mutation rates sets them apart from other forms of life in fundamental ways. Frequent errors in replication and assembly processes give rise to substantial genetic and genomic diversity within viral populations. The diverse constituents of viral populations often interact within and across infected cells, giving rise to complex emergent phenotypes and genetic interactions. This review examines the causes and consequences of viral population diversity and explores how the complex collective interactions among viral genomes i

Alpha-synuclein (α-syn) is best known as a presynaptic protein that supports synaptic vesicle dynamics and neurotransmission. Conversely, misfolded or aggregated α-syn represents a hallmark of synucleinopathies, including Parkinson's disease. Beyond the nervous system, α-syn has been detected in peripheral compartments, including blood cells and selected epithelial tissues, although the robustness and context dependence of expression outside neuronal and erythroid lineages remain under active investigation. Also, it can be released extracellularly through unconventional secretion or cell damag

The trade-off hypothesis plays a foundational role in the theoretical framework of modern evolutionary epidemiology, proposing that pathogen fitness is maximized at intermediate levels of virulence. Despite its influence on the study of host-pathogen interactions, the hypothesis has been criticized for its limited empirical support and the conceptual ambiguities surrounding the definition of virulence. This paper reassesses the scientific status of the trade-off hypothesis by interpreting the dependencies among epidemiological parameters as constitutive principles. Therefore, the results of th

IgA nephropathy (IgAN) is the most common glomerulonephritis worldwide, characterized by a 4-hit pathogenesis primarily driven by mucosal immune dysregulation in the gut-associated lymphoid tissue (GALT). However, the specific environmental triggers initiating this cascade remain elusive. We propose a novel hypothesis that chronic exposure to ubiquitous micro- and nano-plastics (MNPs) serves as a primary environmental catalyst for IgAN. Through a proposed 5-step cascade, we outline how ingested MNPs directly degrade intestinal tight junctions, precipitating a 'leaky gut' that permits the syner

Uncontrolled hemorrhage persists as a primary cause of death in trauma and arterial injuries. The deep, narrow, and irregular tracks of penetrating wounds prevent external manual compression from transmitting sufficient pressure to deep damaged vessels, limiting the effectiveness of traditional hemostatic materials. Therefore, effective hemostasis requires materials with superior plugging capability. Here, we present hemoadhican-functionalized hydrophobic gauze (HHG), a polysaccharide-based hemostat engineered via Pickering emulsion immobilization and freeze-drying to integrate structural and

Uncontrolled hemorrhage causes 30% of trauma deaths. Conventional gauze has limitations in rapid hemostasis and tissue adhesion. Here, we introduce Gauze@LDH, a hemostatic gauze modified with magnesium/aluminum layered double hydroxide nanosheets via a one-step hydrothermal synthesis. This surface engineering transforms the gauze from hydrophilic to hydrophobic, reducing blood absorption. Gauze@LDH exhibits excellent biocompatibility with a 0.209% hemolysis rate and minimal cytotoxicity. It accelerates clotting to 1 min in recalcification models, three times faster than conventional gauze. In

The cationic molecules and low-molecular-weight amphiphiles commonly used in nonviral gene delivery vehicles often induce adverse nonspecific effects, notably cytotoxicity and detergent-like unfavourable membrane activity. To address these limitations, here we report the development of a nontoxic mRNA-condensing agent based on electrically neutral self-assembling peptides (ENSPs). Driven by cooperative hydrogen bonding and hydrophobic interactions, ENSPs coassemble with mRNA into discrete nanotube-shaped complexes. ENSP-based mRNA vehicles are almost nontoxic to mammalian cells and exhibit neg

All eukaryotes share a single-celled ancestor from ∼1.5 to 1.8 billion years ago, the so-called last eukaryotic common ancestor (LECA). Roughly half of gene families found in modern eukaryotes were already present in LECA, forming molecular systems that continue to influence genetic diseases and traits today. To investigate these systems, we compared genes across 156 organisms to define a core set of protein-coding gene families likely present in LECA, with a quarter remaining uncharacterized. Integrating >26,000 mass spectrometry proteomics analyses from 31 species, we inferred higher-order c

Ankylosing spondylitis (AS) is a common immune inflammatory disease. PANoptosis, as a novel programmed cell death pathway, its mechanism of action in ankylosing spondylitis remains unclear. Therefore, this study aims to clarify the role of a novel programmed cell death pattern - PANoptosis - in the pathogenesis of ankylosing spondylitis (AS), and to screen and verify key genes (APRGS), providing new ideas for the diagnosis and treatment of AS. Based on multiple gene expression datasets (GSE25101, GSE73754, GSE11886, GSE134290) of AS patients and known gene libraries related to panoptosis, by i

Rapid advances in microbiome research are transforming our understanding of human health and disease, with growing focus on the female reproductive tract as a critical but understudied niche. Evidence for a local microbiome largely derives from bacterial nucleic acid detection; however, the biological relevance of these signals remains debated, with whether they reflect viable microbial communities, transient colonisation, or mere microbial remnants with immunomodulatory effects. Bacterial extracellular vesicles (BEVs) have emerged as pivotal mediators of host-microbiota crosstalk. Their small

Receptor-interacting protein kinases (RIPKs) are a family of serine/threonine kinases that regulate innate immunity, inflammation, and several modalities of regulated cell death, and are increasingly implicated in cardiovascular disease. Increasing evidence suggests that dysregulated RIPK signaling contributes to cardiomyocyte injury, fibrosis, maladaptive remodeling, mitochondrial dysfunction, and inflammatory amplification in multiple cardiac pathologies. While the roles of RIPK1 and RIPK3 in necroptosis are well recognized, recent studies indicate broader and more complex role in heart dise

Fungal extracellular vesicles (FEVs) have emerged as sophisticated emissaries in cross-kingdom communication, redefining our understanding of fungal secretome in pathogens like Cryptococcus neoformans and Candida species. Rather than mere cellular debris, these bilayered structures are actively exported across the fungal cell wall via complex biogenesis mechanisms, carrying a potent cargo of virulence factors, genetic material, proteins, lipids, and polysaccharides. In human hosts, FEVs facilitate myco-pathogenesis by interacting directly with immune cells to modulate the immune response, eith

Migraine is one of the most disabling neurological diseases globally; however, a significant proportion of patients do not respond to current treatments, or treatment efficacy diminishes over time, highlighting the need to identify additional molecular candidates for further investigation. Recent studies have increasingly focused on the role of neuro-immune interactions in migraine. This study aimed to identify peripheral immune-related candidate genes associated with NAD+ metabolism-related transcriptional features in migraine through integrated bulk and single-cell transcriptomic analyses. Apicomplexan parasites establish intracellular infections that profoundly alter host cell physiology and elicit complex immune responses. The long-standing coevolution between these parasites and vertebrate hosts has resulted in extensive overlap between parasite and host metabolic pathways, limiting the feasibility of conventional parasite-centered therapeutic approaches. Increasing evidence indicates that host-derived signals generated during infection play a decisive role in shaping parasite survival and dissemination. Among these signals, extracellular nucleotides released in response to c

Klebsiella pneumoniae, a facultative anaerobe and Gram-negative bacterium, has high clinical significance in public health concerns. K. pneumoniae is given its name in recognition of the German-Swiss microbiologist Edwin Klebs. Carl Friedlander first identified K. pneumoniae as a bacterium isolated from the lungs of individuals who had died from pneumonia. K. pneumoniae is a prominent member of the Enterobacteriaceae family, an opportunistic pathogen, and one of the pathogens sweeping the world in the antibiotic resistance pandemic. This organism accounts for approximately one-third of all Gra

Circular RNAs (circRNAs) are a special category of non-coding RNAs. The latter are known for their pivotal regulatory functions over many cellular processes. In biology, circRNAs exert regulatory functions on many physiological events, most likely through the regulation of gene expression. They are also implicated in a variety of health-related issues in medicine. Advances in molecular techniques and bioinformatics are expected to expand our knowledge of circRNAs, better characterizing their nature and functions. Remarkably, parasites elaborate their own repertoire of circRNAs to regulate diff

Leishmaniasis remains a major global health challenge, hampered by an antiquated therapeutic arsenal compromised by toxicity, resistance, and an inability to achieve a sterile cure. The high attrition rate in drug and vaccine development stems from a fundamental disconnect between traditional experimental models and the true biological complexity of the Leishmania life cycle. This review synthesizes recent breakthroughs that have reshaped our understanding of the parasite, arguing that these nonclassical traits are the primary obstacles to effective treatment. We deconstruct the updated life c

Protozoan infections remain a major challenge to human health, owing to the substantial impact these parasites exert on their hosts. In vitro culture systems are fundamental for elucidating the host-parasite interactions underlying these infections; however, traditional two-dimensional models are often limited and fail to recapitulate key aspects of human tissue architecture and function. In this context, advanced in vitro cell culture models have emerged as powerful tools, offering increased morphofunctional resemblance to human tissues. This review critically examines recent studies employin

The reemergence of the mpox virus (MPXV) as a global health threat, with >100 000 cases reported across 115 countries since the WHO's 2022 alert, has intensified research into its pathophysiology. MPXV exists in two main clades with distinct geographic and clinical profiles. Despite progress, the impact of MPXV infection in patients with autoimmune diseases remains underexplored. Emerging data highlight immunomodulatory MPXV proteins that suppress innate immunity and dysregulate adaptive responses, through inhibition of type I interferons, nuclear factor kappa-B, and tumor necrosis factor-α.
