Network Destabilization in Aging: Mitochondrial Dysfunction, Nutrient Sensing, and Chronic Inflammation as Interconnected Drivers.

Aging is the dominant risk factor for most chronic diseases, yet the mechanisms driving this relationship remain poorly integrated across biological scales. Existing frameworks have catalogued key hallmarks of aging but do not explain how these processes converge to produce organism-level decline and multimorbidity. A systems-level framework is introduced in which aging is conceptualized as progressive destabilization of interacting regulatory networks. Mitochondrial quality control, nutrient-sensing pathways, and chronic inflammatory signaling form a putative high-centrality network core: mit

Gut microbiota in type 2 diabetes mellitus: mechanistic links between dysbiosis, insulin resistance, and chronic low-grade inflammation.

It is becoming more well acknowledged that type 2 diabetes mellitus (T2DM) is a metabolic and inflammatory condition linked to microbiota that involves interrelated disruptions in intestinal integrity, immune control, and insulin signalling. Butyrate-producing bacteria, such as Faecalibacterium prausnitzii and Roseburia spp., are reduced in gut dysbiosis in type 2 diabetes, whereas opportunistic Gram-negative pathobionts that cause endotoxemia and mucosal inflammation proliferate. Increased intestinal permeability makes it easier for lipopolysaccharide (LPS) to translocate and activate the TLR

Recent advances in piezoelectric hydrogels for osteoarthritis therapy: material design, signal transduction, and clinical translation.

With no disease-modifying therapy available, osteoarthritis (OA) remains a primary cause of disability worldwide. However, between 2023 and 2026, rapid advances have been made in the development of piezoelectric hydrogels, a technology that transduces joint motion into endogenous electrical signals, thereby restoring the electromechanical microenvironment of degenerated cartilage. This short review provides a critical synthesis of these developments through a four-dimensional framework encompassing material design, signal transduction, therapeutic efficacy, and translational validation. Enhanced treatment of ischemic stroke by scutellarin loaded Angiopep-2-modified milk exosomes via multiple pathological pathways regulation.

Ischemic stroke is a major cerebrovascular disease with high morbidity and mortality. However, effective treatments remain limited due to the narrow thrombolytic window, ischemia-reperfusion injury, and restricted drug delivery across the blood-brain barrier (BBB). Due to multiple pathological processes including oxidative stress, inflammation, mitochondrial dysfunction, and microglial dysregulation involved in ischemic stroke, it is urgent to develop drug delivery systems capable of crossing the BBB and targeting multiple pathological pathways. Scutellarin (SCU) exhibits neuroprotective effec

Dopamine Receptor D1 Activation Alleviates Repeated Ketamine Exposure-Induced Cognitive Dysfunction in Mice by Inhibiting NLRP3/Caspase-1-Dependent Pyroptosis.

BACKGROUND AND HYPOTHESIS: Ketamine triggers cellular pyroptosis through an NOD-like receptor protein 3 (NLRP3)/caspase-1 pathway, leading to neurotoxicity and cognitive deficits. Dopamine receptor D1 (DRD1) activation, however, suppresses NLRP3 inflammasome activation, thereby curbing inflammation. We hypothesized that ketamine induces pyroptosis through the NLRP3/caspase-1 pathway, resulting in neurotoxicity and cognitive dysfunction, and that DRD1 activation could counteract these effects. STUDY DESIGN: To investigate the relationship between DRD1 and ketamine-induced pyroptosis and behavi

Modulation of immune responses by opioids through toll-like and P2X receptor signaling in COVID-19.

The pathogenesis of COVID-19 involves complex interactions between viral replication and host immune dysregulation, mediated by toll-like receptors (TLRs) and P2X receptors (P2XRs). These receptors detect pathogen- and damage-associated molecular patterns (PAMPs and DAMPs), triggering inflammatory cascades. Opioids, beyond their analgesic role, modulate innate and adaptive immune responses via opioid receptors and indirectly through TLR and P2X signaling. This narrative review integrates experimental, clinical, and bioinformatic evidence to explore the mechanistic crosstalk between opioid sign

A Computational Strategy for Identifying Self-Assembling Food-Derived Molecules for Antiparasitic Nanotherapy.

Carrier-free nanodrugs remain difficult to design, and the molecular basis of their self-assembly is still poorly understood. Here, an integrated workflow combining 2D/3D molecular screening with SHAP-assisted analysis was used to identify self-assembling pairs from food-derived compounds. Ursolic acid and 18β-glycyrrhetinic acid were thereby identified and found to self-assemble into stable nanoparticles (UA-18βGA NPs). Compared with the individual components, nanoparticles showed reduced cytotoxicity and enhanced antiparasitic activity under the tested conditions. Spectroscopic characterizat

Neuroendocrine and neuroinflammatory mechanisms in postpartum depression: From hormonal withdrawal to central immune dysregulation and emerging therapeutic targets.

Postpartum depression (PPD) affects approximately 10-15% of new mothers worldwide with profound consequences extending to offspring and partners. While the classical "hormonal withdrawal hypothesis" has long emphasized the abrupt decline in estradiol, progesterone, and cortisol following childbirth, emerging evidence demonstrates that these endocrine perturbations converge with dysregulated neuroimmune pathways as central drivers of PPD pathophysiology. This narrative review integrates mechanistic evidence linking reproductive hormone withdrawal, HPA axis dysregulation, neurotransmitter imbala

[Molecular mechanisms of chronic gastric mucosal inflammation induced by Helicobacter pylori via immune evasion].

Helicobacter pylori (H. pylori) persistently colonizes the gastric mucosa and induces sustained inflammation, making it a major driver of the development and progression of chronic gastritis, peptic ulcer disease, and gastric cancer. Its pathogenicity does not rely on direct tissue damage inflicted by a single virulence factor, but rather on the coordinated actions of multiple factors, including cytotoxin-associated gene A (CagA), vacuolating cytotoxin A (VacA), lipopolysaccharide, and urease that disrupt the gastric epithelial barrier and perturb mucosal immune homeostasis. Concurrently, H. p

LRP1 in Ischemic Stroke: From CNS Homeostasis to Ischemic Injury and Repair.

Ischemic stroke is a leading cause of mortality and long-term disability worldwide, with complex pathophysiological mechanisms and limited therapeutic options. The identification of key molecular targets that can effectively modulate the multifaceted pathological and reparative processes underlying ischemic stroke is urgently needed. Low-density lipoprotein receptor-related protein 1 (LRP1) is a multifunctional receptor widely expressed in the central nervous system (CNS), where it regulates lipid and bioenergetic metabolism, preserves blood‒brain barrier (BBB) function, modulates synaptic sig

Immunometabolic control of cytokine production by micronutrients in health, aging, and inflammation.

Micronutrients serve as critical metabolic sensors and epigenetic regulators that orchestrate cytokine production through multiple overlapping signalling cascades, transcriptional networks, and cellular metabolic states. This comprehensive review synthesizes recent research demonstrating that micronutrient status regulates cytokine biology at five hierarchical levels: (i) nutrient sensing via mTORC1/GCN2 and amino acid sensor networks; (ii) transcriptional control through VDR/RARα-mediated epigenetic remodelling and histone deacetylase inhibition; (iii) redox signalling via SELENOK/selenoprote

Hepatoprotective Effect of Floccularia luteovirens (Agaricomycetes) Polysaccharides on Alcohol-Induced Acute Liver Injury in Mice.

Floccularia luteovirens mushroom polysaccharides (FLPs) have anti-inflammatory and antioxidant effects in many inflammatory diseases. However, its protective effect on alcoholic liver injury has not been studied. This study investigated the protective effects of FLPs on acute alcoholic liver injury in mice. After administering FLPs at doses of 200, 400, and 800 mg/kg for 14 days, it was found that FLPs could inhibit the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), and γ-glutamyl transpeptidase (γ-GT) in the serum. FLPs also significant

The Role of Flavonoids in Alleviating Mammary Gland Inflammation: A Review.

Mammary gland inflammation, clinically expressed as mastitis in dairy ruminants and lactating women, arises from a self-amplifying loop of pathogen sensing, oxidative stress and innate immune activation, and remains a leading driver of antibiotic use, milk-quality loss and morbidity. Mounting pressure to reduce antimicrobial dependence has refocused attention on flavonoids-structurally diverse plant polyphenols with multi-target bioactivity, derived mainly from in vitro and rodent mastitis models. This review integrates contemporary evidence on the six principal flavonoid subclasses within a u

Calcineurin inhibition mitigates activation of enteric glia in vitro and affects their crosstalk with macrophages.

Multiple factors contribute to the physiopathology of inflammatory bowel diseases (IBD) and the enteric nervous system is emerging as a key player in this context. In particular, enteric glial cells (EGCs) share very similar properties with central astrocytes, play a homeostatic function under basal conditions, but can be activated by inflammatory stimuli further contributing to mucosal damage. Calcineurin (CN) is an important mediator of astrocyte inflammation and has been described also in EGCs. The aim of this study was to establish an in vitro model of EGCs challenged with an inflammatory

Targeting the osteoimmune microenvironment to prevent regulated chondrocyte death in osteoarthritis: therapeutic potential of natural products.

Osteoarthritis (OA) is increasingly recognized as a whole-joint disease driven by a dysregulated osteoimmune microenvironment, rather than mere mechanical wear. Despite the high prevalence of OA, current pharmacotherapies are largely palliative, failing to halt cartilage degeneration or reverse chondrocyte death. Consequently, the development of Disease-Modifying Osteoarthritis Drugs (DMOADs) targeting the underlying pathology remains an urgent unmet need. Natural products, distinguished by their pleiotropic nature, have emerged as promising candidates capable of functioning as "network regula

HDAC6 inhibition reprograms endothelial and macrophage responses to protect the lung in endotoxemia.

BACKGROUND: Endotoxemia causes pulmonary barrier disruption, endothelial dysfunction, and myeloid-mediated inflammatory responses leading to sepsis-associated acute lung injury. Histone deacetylase 6 (HDAC6) regulates cytoskeletal dynamics and inflammatory signaling; however, its cell-type-specific effects in inflammatory lung injury remain undetermined. METHODS: We conducted single-cell RNA sequencing in a mouse endotoxemia model to investigate the effects of HDAC6 inhibition on endothelial and macrophage responses. Key findings were confirmed using histology and bulk RNA sequencing studies.
