A pilot study to control Lutzomyia umbratilis (Diptera:Psychodidae), the major vector of Leishmania braziliensis guyanensis, in a peri-urban rainforest of Manaus, Amazonas State, Brazil.

In the second half of 1980, 112 (or ca. 16%) of the inhabitants of the new settlement of São José, city of Manaus, contracted cutaneous leishmaniasis whilst clearing their properties of terra firme rainforest. With the aid of SUCAM, the authors carried out a pilot study to investigate the feasibility of reducing populations of Lutzomyia umbratilis, the local silvatic vector of Leishmania braziliensis guyanensis, by spraying insecticide on its favoured diurnal resting sites, the bases of the larger forest trees. Most manvector contact is at these resting sites and, therefore, it was encouraging

Medium for the production of primary powder of Bacillus thuringiensis subsp. israelensis.

Five media, formulated from the seeds of five legume varieties, dried cow blood, and mineral salts, were assessed for the growth and production of insecticidal properties of Bacillus thuringiensis subsp. israelensis. Bacterial powders prepared from the broth cultures were assayed against the larvae of Aedes aegypti, Culex quinquefasciatus, and Anopheles gambiae. A standard primary powder of B. thuringiensis subsp. israelensis (IPS78) was included in the assay for comparison. Good growth was obtained in all the media, and all powders were effective against the three types of mosquito larvae. Th

Metrifonate in schistosomiasis - therapy and prophylaxis.

The introduction and development of the organophosphorous drug metrifonate, first as an insecticide and then for the treatment of urinary schistosomiasis, has followed an unusual course. In its most widely accepted treatment regimen it is safe, effective and apparently without significant side effects. These facts, together with its low cost and ease of dosing, lead to a field trial of its efficacy in the treatment and prevention of infection in children in an area of high S. haematobium and S. mansoni endemicity in Rhodesia (Zimbabwe). Three doses of 7.5 mg/kg b.wt. given at fortnightly inter

Microsomal activation of chlordane isomers to derivatives that irreversibly interact with cellular macromolecules.

Incubation of 14C-labeled cis and trans isomers of chlordane with cofactor-fortified mouse hepatic microsomes resulted in binding of insecticide-derived material to endogenous protein and RNA and to added DNA. The microsomes were prepared from male C57BL/6J mice. Chlordane is known to cause hepatocellular carcinoma in a similar strain. The highest concentrations of radioactive material bound to protein, followed by RNA and DNA. The cis isomer produced greater amounts of bound radioactivity, while binding from trans-chlordane was slight and, in the case of DNA, not detectable. Investigation of

Fine-scale spatio-temporal dynamics of kdr mutation V1016G in Aedes albopictus along a transect encompassing the Rome metropolitan area.

Pyrethroid insecticides are largely exploited in Italy to reduce Aedes albopictus nuisance and to discontinue autochthonous transmission of exotic arboviruses. V1016G mutation in the voltage-gated sodium channel gene is strongly associated with resistance to pyrethroids in this species. Here, we provide an unprecedented high-resolution spatiotemporal analysis of the distribution/frequency of the V1016G mutation over an 11-year period in Ae. albopictus populations sampled across a transect encompassing the eco-geographical variability of Mediterranean Europe, including Rome, the only European m…

The risk posed by exposure to pesticides through beeswax to adult western honey bees (Apis mellifera).

In western honey bee (Apis mellifera) colonies, beeswax can help manage bee exposure to lipophilic compounds via absorption, making the compounds bio-unavailable. However, trapped chemicals may be slowly released into the nest environment, posing risks to bees through chronic exposure. Due to their lipophilic properties (logP > 1), many insecticides used in agriculture or for Varroa destructor control in the honey bee nest can accumulate in beeswax. Via in vitro cage assays, we determined the LC50 values of six pesticides (coumaphos, amitraz, acrinathrin, chlorpyrifos, chlorfenvinphos, and bif…

Oral RNAi reduces survival and grain loss in Rhyzopertha dominica, including phosphine-resistant populations.

BACKGROUND: Phosphine fumigation remains a cornerstone of stored-grain protection, yet widespread resistance in Rhyzopertha dominica threatens its long-term effectiveness. RNA interference (RNAi) through ingestion of double-stranded RNA (dsRNA) offers a sequence-specific mode-of-action (MoA) distinct from conventional insecticides. However, practical development requires identification of orally effective target genes, demonstration of grain-protection benefits, and validation of efficacy in phosphine-resistant populations. RESULTS: Oral delivery of dsRNA produced dose-dependent reductions in…

Pesticides as Hidden Modifiers of Drug Action: Mechanisms and Human Health Implications.

Pesticides have been widely used in agricultural practices, as well as in the prevention of vector-borne and other communicable diseases. Although the ecotoxicology and adverse effects on human health are known, recent studies have shown that some types of pesticides may interact with therapeutic drugs, a fact that should be recognized by public health authorities as a new public health issue. The current review was intended to provide an overview of the present knowledge of different groups of pesticides (insecticides, herbicides, and fungicides), as well as their mechanisms of action, which …

Genome-wide identification of neuropeptides from Plutella xylostella (Lepidoptera: Plutellidae) and elucidating the essential roles of adipokinetic hormone in lipid mobilisation and food intake.

The diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae), is a major pest of cruciferous crops and has developed high levels of resistance to many widely used insecticides. Neuropeptides are involved in regulating essential insect biological processes, which could be considered potential target genes for pest control. In this study, a total of 45 neuropeptide precursor genes were identified in the genome of P. xylostella, including two adipokinetic hormone genes (PxAKH1 and PxAKH2). The qPCR analysis showed that PxAKH genes were highly expressed in third- and fourth-instar lar…

Prolonged Deltamethrin Exposure Induces Dose-Dependent Glycerol Overproduction and Efficient Deltamethrin Removal by Saccharomyces cerevisiae.

Background/Objectives: Pest management strategies rely on insecticides such as deltamethrin (DM), a commonly applied type II pyrethroid. As a natural component of food-associated microflora, Saccharomyces cerevisiae inevitably encounters DM residues in crops used for fermentation processes, including dough leavening and winemaking. However, the prolonged effect of DM exposure on yeast fermentation performance and its capacity to remove DM remained unclear. Methods: In this study, S. cerevisiae was continuously exposed to a non-lethal concentration (10 mg/L) and a low-inhibition toxic concentra…

Pesticide occurrence, transformation, and transport from wastewater treatment plants into stream networks with diverse land uses.

Neonicotinoid insecticides and strobilurin fungicides are detected in many environmental compartments and have been associated with negative environmental and human health implications. Wastewater treatment plants (WWTPs) are often hotspots for introducing such contaminants into the environment. Therefore, the occurrence of strobilurin fungicides, neonicotinoids, and their metabolites at two WWTPs with varying land uses and population sizes was investigated. Polar organic chemical integrative samplers were deployed in WWTP influent and effluent and placed upstream and downstream of the effluen…

A chromosome-level genome assembly and developmental transcriptome profiling reveal stage-specific remodeling of the molecular chaperone system in Helicoverpa armigera.

Helicoverpa armigera is one of the most destructive lepidopteran pests worldwide owing to its remarkable polyphagy, long-distance migration, and rapid adaptation to insecticides. Here, we present a chromosome-level genome assembly of H. armigera generated from a field-collected individual in southwestern China, providing a valuable resource for future population genomic and pangenome studies. Developmental transcriptome analyses of first-instar larvae, fifth-instar larvae, and adults identified 6817, 3519, and 5518 differentially expressed genes, respectively, including 797 shared among all de…

Functional characterization of CaspOR20 involved in repellency of 1-penten-3-ol, sulcatone, and methyl salicylate against the rape stem weevil Ceutorhynchus asper.

The rape stem weevil (Ceutorhynchus asper) is a devastating pest of cruciferous crops, and its management heavily relies on insecticides, leading to resistance development and environmental concerns. CaspOR20 is a highly expressed odorant receptor in the antennae of C. asper, yet its functional role remains unclear. In this study, by using the transgenic Drosophila expression system combined with single sensillum recordings, we identified six volatile compounds (1-penten-3-ol, 2-heptanone, sulcatone, methyl salicylate, benzaldehyde, and ethyl acetate) as ligands for CaspOR20. Subsequent electr…

Pesticides, Environmental Contamination, and Public Health: A One Health Perspective.

Pesticides are chemicals used in the agricultural, industrial, and public health sectors to increase crop productivity, prevent plant diseases, and reduce pest-related hazards. They can be classified as herbicides, insecticides, fungicides, and rodenticides based on the pests they target. Their active ingredients include organochlorines, organophosphorus compounds, carbamates, pyrethrins, and pyrethroids. Although the use of pesticides has provided significant benefits to humans, their widespread application has also led to the contamination of air, soil, water, and food systems. After applica…

Optimized conditions for the long-term growth of primary cell cultures derived from the southern green stink bug, Nezara viridula.

The southern green stink bug, Nezara viridula L., is a highly polyphagous global pest that causes substantial economic losses on a broad range of crops. Current management strategies rely heavily on chemical insecticides, which can promote resistance development and negatively affect non-target beneficial organisms. The lack of a tractable in vitro cell culture system has limited the development of more targeted and sustainable control approaches for this species. In this study, we identified the optimal egg age for culture initiation and evaluated seven insect cell culture media or medium com…
