Markerless On-Device Detection of Compensatory Movement Patterns in Upper-Limb Rehabilitation Exercises from Monocular RGB Video: A Validation Study in Healthy Adults.

Neurological disorders drive demand for prolonged upper-limb rehabilitation, yet specialist access is uneven and assessment stays subjective. Marker-based and inertial measurement unit (IMU) systems are accurate but costly and impractical at home, while pose estimation pipelines mostly stop at keypoints, and many process video server-side, raising privacy concerns. We present a markerless pipeline that analyzes monocular RGB video entirely on-device in the browser, so it never leaves the machine. From 33 BlazePose keypoints, it derives five geometric metrics designed to limit body-size depende

Automated assessment of coronal lower extremity alignment on long-leg radiographs using a deep-learning model: validation, efficiency gains, and superiority of an edge-based tibial joint-line definition.

PURPOSE: To develop, validate, and benchmark a fully automated deep learning (DL) system that simultaneously measures 15 coronal lower-limb alignment parameters on standing long-leg radiographs (LLRs) and localizes deformity, and to compare two tibial joint-line definitions for suitability in DL-based measurement. METHODS: A retrospective set of 309 anteroposterior standing LLRs was split into training/validation/testing (60/20/20). External generalizability was assessed using 75 independent LLRs from a different scanner and patient cohort. YOLOv5 was used to detect multiple bony landmarks, f

Utilizing intraindividual cognitive variability to predict progression in stage 2 neuronal synuclein disease.

OBJECTIVE: Neuronal synuclein disease (NSD) involves pathological α-synuclein presence and often dopaminergic dysfunction, initially preceding overt clinical symptoms. NSD-Integrated Staging System identifies Stage 2A (no dopaminergic dysfunction) and 2B (dopaminergic dysfunction) as prodromal phases marked by subtle clinical signs without functional impairment. Intraindividual variability/dispersion (IIV-D), reflecting the degree of spread in a person's performance across a battery of cognitive tasks, has emerged as a potential marker of early neurodegenerative changes. This study examined wh

White Matter Microstructural Correlates of Auditory Brainstem Responses in Patients With Charcot-Marie-Tooth Disease.

INTRODUCTION: Some patients with Charcot-Marie-Tooth disease (CMT) exhibit prolonged auditory brainstem response (ABR) latencies or abnormal waveforms, suggesting potential damage to the peripheral auditory nerve or central auditory pathways. Diffusion tensor imaging (DTI), a non-invasive neuroimaging technique, can detect the integrity and functional properties of white matter structures with high sensitivity. However, research on the association between DTI characteristics and ABR changes in patients with CMT remains relatively limited, and whether both modalities reflect synergistic damage

OTOLens: Development and validation of a mobile application for assessment of tympanic membrane and middle ear pathology.

BACKGROUND/OBJECTIVE: Diagnosing tympanic membrane and middle ear pathology in primary care is challenging due to anatomical complexity, overlapping presentations, and limited access to specialized equipment. We developed and externally validated OTOLens, a smartphone-based mobile application classifying otoscopic images using deep learning. METHODS: In this prospective development and validation study, OTOLens combined three ResNet-based image classifiers (OME, TMP, and MYR) with a parallel gradient-boosted tabular classifier operating on PCA-reduced image features, integrated by a per-condi

Implementation and evaluation of a pragmatic community streptococcal treatment programme to improve rheumatic heart disease primary prevention in Uganda.

INTRODUCTION: Detection and treatment of streptococcal pharyngitis in children reduces acute rheumatic fever by 70-80%. However, investment in primary prevention is limited in rheumatic heart disease (RHD) endemic countries due to lack of diagnostic capacity. We conducted a pragmatic public health trial (Rheumatic Heart Disease Community Streptococcal Treatment Program (RESET)) to determine the effect of an integrated group A streptococcal (GAS) education and treatment programme on the echocardiographic burden of RHD among children aged 5-15 years in Uganda. METHODS: We implemented our interv

Machine learning reveals temperature as a key predictor of dengue risk across Thailand's provinces: A 20-year analysis.

BACKGROUND: Dengue fever remains a critical public health challenge in Thailand, with transmission dynamics driven by complex interactions between environmental and socioeconomic factors. Understanding these predictive factors is essential for developing robust forecasting systems. METHODS: We developed a machine learning framework to classify spatiotemporal dengue risk and identify key predictive factors across Thailand. We analyzed 20 years of monthly dengue hemorrhagic fever surveillance data (2003-2022) from 77 provinces, integrating 54 environmental, climatic, and socioeconomic features.

Combinatorial Treatment with Chlorogenic Acid and Cinnamaldehyde Disrupts Intracellular pH and Metabolic Transport in Breast Cancer Cells.

Breast cancer cells exhibit a reversed pH gradient and metabolic plasticity that promote proliferation, invasion, and resistance to therapy. Natural products such as chlorogenic acid (CGA) and cinnamaldehyde (CA) have shown emerging anticancer potential. However, their effects on intracellular pH and metabolic transport systems remain undefined. Therefore, the aim of this study was to characterize these parameters in breast cancer and non-tumorigenic breast cells. This study evaluated the physiochemical properties of CGA and CA using LC-MS, under pH conditions (pH 1.2, 7.4, and 9.0) mimicking

Robust neural network controller for a lower limb exoskeleton with minimal sensor configuration: a deep reinforcement learning approach with policy distillation.

BACKGROUND: Lower limb rehabilitation exoskeletons (LLREs) are increasingly used in clinical settings to improve mobility in individuals with neuromuscular impairments. Most LLREs employ controllers focused on trajectory tracking, which lack adaptability to user-specific variations or voluntary effort. Autonomous LLREs enable hands-free gait but often require a multitude of sensors such as IMU, encoders, and foot force sensors to enable user intent prediction, gait phase detection, and dynamic balance. This often introduces substantial challenges related to cost, complexity, reliability, and s

Accurate classification of benign and malignant breast tumors in ultrasound imaging with an enhanced deep learning model.

BACKGROUND: Breast cancer is the most common malignant tumor in women worldwide, and early detection is crucial to improving patient prognosis. However, traditional ultrasound examinations rely heavily on physician judgment, and diagnostic results are easily influenced by individual experience, leading to frequent misdiagnosis or missed diagnosis. Therefore, there is a pressing need for an automated, highly accurate diagnostic method to support the detection and classification of breast cancer. This study aims to build a reliable breast ultrasound image benign and malignant classification mode

Diagnostic value of deep learning of multimodal imaging of thyroid for TI-RADS category 3-5 classification.

BACKGROUND: Thyroid nodules classified within the Thyroid Imaging Reporting and Data Systems (TI-RADS) category 3-5 are typically regarded as having varying degrees of malignancy risk, with the risk increasing from TI-RADS 3 to TI-RADS 5. While some of these nodules may undergo fine-needle aspiration (FNA) biopsy to assess their nature, this procedure carries a risk of false negatives and inherent complications. To avoid the need for unnecessary biopsy examination, we explored a method for distinguishing the benign and malignant characteristics of thyroid TI-RADS 3-5 nodules based on deep-lear

Evaluation of sampling methods for genomic surveillance of SARS-CoV-2 variants in aircraft wastewater: advancing global early-warning systems for future pandemics.

BACKGROUND: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is an ongoing threat to global health. Wastewater-based surveillance (WBS) has proven to be an important tool for tracking the dissemination of SARS-CoV-2 variants of concern (VOCs) in the community. In Canada, metagenomic analysis of aircraft wastewater was adopted at an early stage of the pandemic to track importation of emerging variants into the country. However, the acute need to determine the presence of emerging SARS-CoV-2 sublineages meant that the sampling methods utilized were not adequately validated. Here, we

Development and Validation of a Cross-Device Platform for Anhedonia Trend Visualization by Using Ecological Momentary Assessment and Moving Averages (Part I): Protocol for a Methodological Pilot Study.

BACKGROUND: Anhedonia is a core symptom of depressive disorders. Although group-level cutoff points for the Snaith-Hamilton Pleasure Scale (SHAPS) are established, real-time, individual-level monitoring remains limited. Ecological momentary assessment (EMA) enables high-frequency sampling, and simple moving averages (SMA; 7/14/30 days) offer an interpretable way to smooth daily signals and surface early-warning trends. OBJECTIVE: The aim of this study was to develop and validate a cross-device platform that collects EMA-based SHAPS responses and visualizes SMA trends for individual anhedonia
