BACKGROUND: Mycobacterium bovis Bacille Calmette-Guérin (BCG) and other mycobacterial infections suppress airway hyperresponsiveness and eosinophilic inflammation in asthma.γδT cells are important modulators of airway function and allergic inflammation. Vγ1+γδT cells increase eosinophilic airway inflammation and airway hyperresponsiveness, while Vγ4+γδT reduce airway hyperresponsiveness. The objective of this investigation was to determine the role of γδT cells and its subsets in the treatment of inhaled inactivated-Mycobacterium phlei for asthma.
METHODS: OVA-sensitised mice were treated wit…

RNA chaperones play a crucial role in the biogenesis and function of various RNAs in bacteria. They facilitate the interaction of small regu … een found in their genomes. We determined the crystal structure of the cold shock protein CspB from Mycobacterium tuberculosis. It forms a dimer due to its elongated C-terminal region, which is a hairpin composed of two α-helices. It was also demonstrated that CspB from M. tuberculosis exhibits high affinity for MTS0997 sRNA and MTS1338 sRNA from the same organ …

Silymarin is a natural polyphenol known for its broad range of biological effects, and research conducted in macrophages and mice infected with Mycobacterium tuberculosis have highlighted its potential as a complementary treatment for tuberculosis. Silymarin modulates multiple cellular signaling pathways associated with various aspects of the immune response. However, its effect on the control of M. tuberculosis infection in pulmonary epithelial cells is still not well understood. In this study, we evaluated the effect of silymarin on the infection control and immune response of A549 pulmonary…

Tuberculosis (TB) remains a major global threat, demanding new therapeutic strategies. Dihydrofolate reductase (DHFR), essential for cell survival and a known drug target, has emerged as a promising enzyme for anti-TB drug development. Using crystallographic structures of Mycobacterium tuberculosis DHFR (MtDHFR) complexed with classical and potential inhibitors, a quantum-chemical analysis was performed through the molecular fractionation with conjugate caps (MFCC) scheme within the density functional theory (DFT) framework to quantify individual amino acid contributions to ligand-protein inte…

The rise in drug-resistant Mycobacterium tuberculosis (M.tb) necessitates new therapeutic strategies. Hypothesizing that a single compound targeting multiple M.tb proteins would make resistance less likely to evolve, we focused on identifying a single compound targeting multiple M.tb OmpR family two-component response regulators (RRs). We screened ∼8000 FDA-approved DrugBank compounds using computational tools (active-site prediction, receptor-ligand docking, and molecular dynamics simulations) to identify one compound acting on M.tb RRs: MtrA, PrrA, MprA, RegX3, and PhoP. Cefpiramide showed s…

Mycobacterial cell-wall assembly depends on multiple glycosyltransferases among which the processive galactofuranosyl transferase 2 (GlfT2) executes alternating β(1 → 5) and β(1 → 6) transfers of galactofuranose (Galf) within a single catalytic site through an oxocarbenium-like transition state. Despite the well characterized SN2-like chemistry of GlfT2, the role of loop-1 dynamics in regulating substrate binding and processivity remains poorly understood. Here, we modeled the ternary complex of GlfT2 with UDP-Galf and a galactan acceptor (n = 3) to show that loop-1 undergoes conformational ga

UNLABELLED: Riboflavin biosynthesis is required for in vitro survival of the human pathogen Mycobacterium tuberculosis ( Mtb ). However, despite the lack of a known transporter, growth can be rescued by exogenous riboflavin. The riboflavin biosynthesis pathway is also predicted essential in vivo , but whether riboflavin levels available in the host can support survival has not been directly tested. Here we constructed a set of inducible CRISPR interference (CRISPRi) knockdown and targeted gene deletion strains for known riboflavin biosynthesis genes ( ribA2, ribG, ribH, ribC ) as tools to char…

Mycobacterium avium is an important zoonotic nontuberculous mycobacteria (NTM), and its continuously rising infection rate has posed a severe public health challenge. However, due to the incomplete annotation of protein-coding sequences in the reference genome and the absence of proteomic data for the type strain, the expression profiles of the proteins encoded by this bacterium remain poorly characterized. To address the gap between genome prediction annotation and protein expression, it is crucial to acquire systematic and reliable experimental evidence of proteins at the type strain level. …

Herein, we report the case of an 87-year-old woman who presented with acute chest pain, fever, and cough. Imaging revealed a right pneumothorax, pleural effusion, and bronchiectasis. Pleural fluid was exudative with elevated adenosine deaminase levels, and Mycobacterium avium complex (MAC) was confirmed. Thoracoscopic plication and empyema cavity curettage were performed. Postoperatively, clarithromycin, rifampicin, ethambutol, and amikacin were initiated, leading to clinical improvement, smear and culture negativity, and discharge. Pleuritis caused by nontuberculous mycobacteria (NTM) is rare…

PURPOSE: Whole Genome Sequencing (WGS) comprehensively detects all drug-resistant mutants of Mycobacterium tuberculosis, which can help in the early initiation of specific treatment for the patient. But there is a need to evaluate the performance of WGS in comparison with Line Probe Assays (LPA) and Phenotypic Drug Susceptibility Tests (pDST).
METHODS: Consecutive sputum samples (58) found positive for Mycobacterium tuberculosis (MTB) by GeneXpert were tested for first-and second-line LPA, pDST and WGS for anti-tubercular drugs.
RESULTS: Of 58, 34 (58.6%) culture isolates were resistant to o…

The connection between genetic mutations linked to delamanid resistance and phenotypic resistance remains unclear. We assessed the phenotypic effects of delamanid-resistant mutations in the Mycobacterium tuberculosis H37Rv strain through gene disruption using homologous recombination and complementation tests. Delamanid resistance was assessed by determining the minimum inhibitory concentration (MIC) via the 7H9 microdilution method. Sanger sequencing identified mutations, and conservation of the mutated residues was predicted through multiple sequence alignments of orthologs. A total of 116 i…

Tuberculosis (TB) is a recurrent and progressive bacterial disease caused by Mycobacterium tuberculosis (Mtb), posing a significant challenge globally due to its drug resistance. This study focuses on identifying natural phytocompounds from the plant Datura innoxia (leaves), which is well known for its biologically active metabolites. Initially, the current study employed in vitro analysis of 20 phytocompounds, revealing that the natural compound 9, o-vanillin, exhibited the best minimal inhibitory concentration (MIC) which was 12.5 µg/mL, and minimal bactericidal concentration (MBC) was 50 µg…

The diagnosis of pleuritis caused by mycobacteria is not always easy. Ziehl-Nielsen staining utilizing cell blocks (CBs) of lower respirator … ce analysis of the 16S rRNA gene using the remaining PE-CB showed high homology (99.79%) to that of Mycobacterium goodii. This case suggests that analysis of PE-CBs may be useful for diagnosing suspected cases of mycobacteria-induced pleuritis with negative acid-fast bacilli smears for PE. J. Med. Invest. 72 : 202-206, February, 2025.

No currently licensed vaccine reliably prevents pulmonary tuberculosis (TB), a leading cause of infectious disease mortality. Developing effective new vaccines requires identifying which Mycobacterium tuberculosis (Mtb) proteins are presented on major histocompatibility complex class II (MHC-II) by infected human phagocytes (target cells) and defining their capacity for recognition by CD4+ T cells. Vaccine designs must elicit T cell responses recognizing the same peptide-MHC complexes presented by infected cells. Although many human CD4+ T cell Mtb epitopes have been described, presentation on…

Drug resistance in Mycobacterium tuberculosis presents a major challenge in tuberculosis treatment, highlighting the need to understand the underlying mechanisms. DNA replication plays an important role in the acquisition of drug resistance, and the expression of the DNA polymerase DnaE2 during adverse conditions has been associated with increased mutation rates. Here, we investigate the functional differences between the high-fidelity replicative DNA polymerase DnaE1 and the predicted error-prone DNA polymerase DnaE2, focusing on which amino acid changes affect polymerase fidelity. For this, …

Mycobacterium tuberculosis (Mtb) is the leading cause of human deaths by an infectious agent. To survive in the human host, Mtb must acquire essential iron nutrients from the host and evade the immune response. In diderm bacteria like Mtb, outer membrane channel transporter proteins are fundamental for nutrient acquisition and immune evasion. Recently, we demonstrated that the Mtb outer mycomembrane PPE64 is a channel protein, providing the first direct evidence of channel-forming capability by a protein of the PPE (proline-proline-glutamate motif) family, which are found exclusively in mycoba…
