Mesothelial cell senescence induced by high glucose in peritoneal dialysis drives fibroblast activation and peritoneal fibrosis through KLF6-mediated activation of TGF-β pathway.

Long-term peritoneal dialysis (PD) frequently leads to peritoneal fibrosis (PF), resulting in ultrafiltration failure. This study aims to elucidate the molecular mechanisms underlying PD-associated PF and provide new insights for its treatment. Single-cell RNA sequencing (scRNA-seq) data from PD patients in the GEO database were analyzed to examine the cellular composition and gene expression profiles of peritoneal tissues. Human peritoneal mesothelial cells (PMCs) were treated with high glucose (2.5%) to simulate the PD microenvironment. Cellular senescence and related signaling pathways were

Peritoneal Neutrophil Extracellular Traps contribute to septic AKI via peritoneal IL-17A and distant organ CXCL-1/ CXCL-2 pathway in abdominal sepsis.

There are no specific treatments for Sepsis-associated acute kidney injury (SAKI). We previously reported that Il-17a-knockout mice had dramatically improved survival after cecal ligation and puncture (CLP). Neutrophil extracellular traps (NETs) induce IL-17A, which causes harm in some diseases, but this pathway is poorly understood in sepsis. We found that knockout of Pad4 (Peptidyl Arginine Deiminase 4), an enzyme essential for NET formation, improved survival and AKI, and suppressed neutrophil infiltration into remote organs, involving a peritoneal IL-17A/distant organ CXCL-1/CXCL-2 pathway

Prospective Randomised Control Study to Evaluate the Effectiveness of Hypochlorous Acid as a Peritoneal Lavage Agent to Prevent Surgical Site Infection After Exploratory Laparotomy for Perforation Peritonitis.

Introduction Surgical site infections (SSIs) are a major concern following emergency gastrointestinal procedures, particularly exploratory laparotomy for perforation peritonitis, where infection risk is amplified by the inherent contamination of the operative field. Despite prophylactic antibiotics, the microbial burden frequently surpasses host defence thresholds. Intraoperative peritoneal lavage with antimicrobial agents such as hypochlorous acid (HOCl) has emerged as a promising strategy, combining physical removal of contaminants with direct microbicidal action. Methods This was a prospect…

Sodium Alginate Microgel-Wrapped Primary Human Peritoneal Mesothelial Cells for the Treatment of Peritoneal Fibrosis Caused by Dialysis via the POSTN/NF-κB/CXCL8 Pathway.

BACKGROUND: Peritoneal fibrosis is a significant complication arising from long-term peritoneal dialysis (PD), primarily due to the loss of peritoneal mesothelial cells (PMCs). Recent studies have implicated periostin (POSTN) in the progression of various fibrotic diseases; however, its specific role in PD-induced peritoneal fibrosis remains unclear. Sodium alginate (SA) microgels have emerged as promising carriers for cell encapsulation in tissue engineering and regenerative medicine. This study investigated the therapeutic potential of PMCs encapsulated in SA microgels (SA/PMC) for reducing

The measured distance between tumor cells and the peritoneal surface predicts the risk of peritoneal metastases and offers an objective means to differentiate between pT3 and pT4a colon cancer.

Substantial variability exists in what pathologists consider as pT4a in colorectal cancer when tumor cells are within 1 mm of the free peritoneal surface. This study aimed to determine if the measured sub-millimeter distance between tumor cells and the free peritoneal surface would offer an objective means of stratifying patients according to the risk of developing peritoneal metastases. Histological slides of patients included in the COLOPEC trial, with resectable primary c/pT4N0-2M0 colon cancer, were centrally reassessed. Specific tumor morphological variables were collected, including dist

Infectious Agents Identified by Real-Time PCR, Serology and Bacteriology in Blood and Peritoneal Exudate Samples of Cows Affected by Parietal Fibrinous Peritonitis after Caesarean Section.

The aim of this study was to identify the pathogens potentially involved in parietal fibrinous peritonitis (PFP). PFP is a complication of laparotomy in cattle, characterized by an accumulation of exudate inside a fibrinous capsule. We have studied 72 cases of PFP in Belgian blue cows, confirmed by a standard diagnostic protocol. Blood was collected to evaluate the presence of antibodies for Mycoplasma bovis(M. bovis), Coxiella burnetii(C. burnetii) and Bovine Herpesvirus 4(BoHV4) by enzyme-linked immunosorbent assays. Peritoneal exudate was obtained from the PFP cavity to perform bacteriologi…

Particle stability and structure on the peritoneal surface in pressurized intra-peritoneal aerosol chemotherapy (PIPAC) analysed by electron microscopy: First evidence of a new physical concept for PIPAC.

Pressurized intra-peritoneal aerosol chemotherapy (PIPAC) has been introduced to the clinical setting as a novel approach for the treatment of peritoneal metastasis. The local interaction of chemoaerosol droplets with the peritoneal surface as well as their distribution pattern is considered the main advantage over conventional liquid intraperitoneal chemotherapy. The aim of the present study was to investigate the behavior of these aerosol particles during PIPAC application via electron microscopy. Solutions of doxycycline, liposomal doxorubicin and macrophage cells were aerosolized using an

Peritoneal Equilibration Test Reference Values Using a 3.86% Glucose Solution During the First Year of Peritoneal Dialysis: Results of a Multicenter Study of a Large Patient Population.

BACKGROUND: The original peritoneal equilibration test (PET) was used to classify peritoneal dialysis (PD) patients using a 2.27% glucose solution. It has since been suggested that a 3.86% glucose solution be used because this provides better information about ultrafiltration (UF) capacity and the sodium (Na) sieving of the peritoneal membrane. OBJECTIVE: The aim of this study was to determine reference values for a PET using a 3.86% glucose solution (PET-3.86%). METHODS: We evaluated the PET-3.86% in a large population of incident PD patients attending 27 Italian dialysis centers. RESULTS:
