Developmental expression of two members of a new class of transcription factors: I. Expression of aryl hydrocarbon receptor in the C57BL/6N mouse embryo.

The aryl hydrocarbon receptor (AhR) is a ligand-activated transcription factor with a basic region/helix-loop-helix (bHLH) motif. AhR has been sequenced and the functional domains defined and there is information on the formation of complexes with other peptides and interactions with DNA, although these areas continue to be investigated. AhR mediates many biological effects such as developmental toxicity, including induction of cleft palate and hydronephrosis. This regulatory protein is expressed in embryonic liver and has been immunohistochemically localized in cells of human and mouse second

Activation and immortalization of leukaemic B cells by Epstein-Barr virus.

We have studied the responses of chronic leukaemic B cells to infection by Epstein-Barr virus (EBV). Our results define one population of B lymphocytes, represented by prolymphocytic leukaemic (PLL) cells, which are highly susceptible to immortalization by EBV. Another B-cell type, represented by chronic lymphocytic leukaemic (CLL) cells, can be readily infected by the virus but is resistant to immortalization. Comparative studies of viral and cellular related events early after infection in these 2 cell types reveal that both express the EB viral nuclear antigen (EBNA) complex, but the immort

A comparison of the prevalence of autoantibodies in individuals with chronic hepatitis C and those with autoimmune hepatitis: the role of interferon in the development of autoimmune diseases.

BACKGROUND AND OBJECTIVES: Viral hepatitis due to hepatitis C virus results in chronic liver disease in more than 70% of individuals infected with the virus. Hepatitis C virus is also thought to be the cause of autoimmune chronic hepatitis, type II. The only treatment for chronic hepatitis C is interferon (IFN). IFN is both an antiviral agent and an up regulator of the cellular immune system. The latter effect is non-specific. Thus, IFN diffusely activates the cellular immune system and can initiate new autoimmune diseases in patients treated with it. To determine the prevalence of autoantibod

The E3 ubiquitin ligase NEDD4 protects against nonesterified fatty acid-induced hepatic inflammatory injury in ketotic cows.

Cows with ketosis frequently exhibit liver injury, primarily driven by increased influx of nonesterified fatty acids (NEFA) into the liver. This excess NEFA induces lipotoxicity, which triggers hepatic inflammatory responses and ultimately results in liver damage, further exacerbating ketosis. Neural precursor cell expressed developmentally downregulated 4 (NEDD4), an E3 ubiquitin ligase, plays a crucial role in regulating anti-inflammatory processes, yet its role in the hepatic injury in ketotic cows remains unclear. This study aimed to investigate the link between NEDD4 and hepatic inflammat

Hepatobiliary organic anion transporters are differentially regulated in acute toxic liver injury induced by carbon tetrachloride.

BACKGROUND/AIMS: Hepatobiliary transporters are down-regulated in cholestasis, but their expression in acute, non-cholestatic, cytokine-mediated liver injury is unknown. Thus we studied the molecular mechanisms, by which sodium taurocholate cotransporting polypeptide (Ntcp), organic anion transporting polypeptide 1 (Oatp1), Oatp2, Oatp4, multidrug-resistance protein 2 (Mrp2) and bile salt export pump (Bsep) are regulated in liver injury induced by carbon tetrachloride (CCl(4)). METHODS: mRNA and protein levels were determined in rats 24 and 72h after CCl(4) injection. Transporter gene transcr

Elevated CERS6/C16:0 Ceramide in taPVAT Promotes Aortic Dissection by Driving Macrophage Inflammation via FPR2/NF-κB Signaling.

BACKGROUND: Perivascular adipose tissue (PVAT) is the endocrine microenvironment closest to the aorta. Using multiomics analysis, we found that Cers6 (ceramide synthase 6) was upregulated and C16:0 ceramide accumulated in thoracic aortic PVAT (taPVAT) from aortic dissection (AD) mice; however, its role and underlying mechanism in AD remain unclear. METHODS: A mouse model of AD was established by combined β-aminopropionitrile and AngII (angiotensin II) administration, followed by proteomic and quantitative lipidomic analyses of taPVAT. Cers6 expression in taPVAT adipocytes was locally manipula

KRAS-ERK signaling drives metastasis in colorectal cancer via phosphorylation-dependent activation of the ZBTB20-TGFBR2 axis.

BACKGROUND: Metastatic colorectal cancer (CRC) harboring KRAS mutations presents a major therapeutic challenge due to its aggressive nature, poor prognosis, and resistance to EGFR-targeted therapies. This study aimed to identify novel drivers of metastasis specifically in KRAS-mutant CRC and to elucidate the underlying molecular mechanisms to undercover new therapeutic vulnerabilities. METHODS: We integrated data from clinical databases (TCGA, CPTAC) with experimental validation using human CRC cell lines, a tissue microarray, and two distinct in vivo metastasis models (liver and lung coloniz

YAP-activated lymph node fibroblasts contribute to extracellular matrix remodeling associated with extranodal extension of oral squamous cell carcinoma.

Desmoplasia, characterized by excessive expansion of cancer-associated fibroblasts (CAFs) and aberrant extracellular matrix (ECM) deposition, is widely present especially in invasive cancers. The extranodal extension (ENE) of nodal metastasis involves the extension of invasive tumor cells through the lymph node capsule into the perinodal adipose tissue, leading to systemic metastasis. Thus, the specific roles of desmoplasia in ENE formation need to be identified. This study aimed to investigate the association between desmoplasia and its role in ENE formation of oral squamous cell carcinoma, w

Mitochondria-targeted metformin analogs activate the ER stress-unfolded protein response pathway to drive apoptosis in pancreatic cancer.

Accumulating evidence indicates that evasion of apoptosis and metabolic reprogramming are necessary for pancreatic cancer growth, early invasion, and chemotherapeutic resistance. Building on our prior work, we investigated the anti-tumor potential of a rationally designed mitochondria-targeted variant of the anti-diabetic drug metformin, Mito-Met10, in cell culture models and orthotopic xenografts. Using MALDI-mass spectrometry imaging, therapeutic concentrations of fluorinated Mito-Met10 were shown to preferentially localize within pancreatic tumors relative to adjacent tissue and liver. Trea

Ferroptosis regulates metabolic inflammation-driven tumorigenesis in obesity-related gastric cancer.

BACKGROUND AND AIMS: The incidence of gastric cardia adenocarcinoma (GCA) has increased with increasing prevalence of metabolic syndrome (MetS). Peroxisome proliferator-activated receptor-delta (PPAR-δ) emerged as a key target in MetS and carcinogenesis. We aimed to elucidate its role in GCA progression. METHODS: Male K19-Wnt1/C2mE mice were fed a control or high-fat diet (HFD) with or without the PPAR-δ agonist GW-501516 or the PPAR-δ antagonist GSK-3787 (GSK). The involvement of nuclear factor erythroid 2-related factor 2 (Nrf2) in the PPAR-δ-regulated, metabolism-driven tumorigenesis was e

A LINE-1 retrotransposon promotes SEMA3C expression as a cis-regulatory enhancer to sustain breast cancer stem cell survival.

Transposable elements (TEs), which are under tight epigenetic control, have been co-opted as cis-regulatory elements to regulate gene expression during development and cancer. Among them, long interspersed element 1 (LINE-1) retrotransposons are the most abundant and exhibit high activity in embryonic stem cells. However, the precise role of LINE-1 in breast cancer stem cells (BCSCs) remains poorly understood. Here, using RNA sequencing, an enhancer dual-luciferase reporter assay, and a nuclease-dead Cas9 (dCas9)-based CRISPR activation (CRISPRa) assay, we show that the LINE-1 retrotransposon

Adipose tissue-specific Nrf2 knockdown inhibits the cGAS-STING pathway to attenuate inflammation in obese mice.

BACKGROUND: Obesity is a chronic, non-infectious inflammatory disease associated with oxidative stress and is triggered by adipose tissue expansion. Nuclear factor erythroid 2-related factor 2 (NRF2) is a core antioxidant defense system transcription factor in adipose tissue and is associated with the cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway. However, the regulatory roles of NRF2 and the cGAS-STING pathway in obesity-related metabolic disorders remain unclear. Therefore, this study aimed to evaluate the effects of adipose tissue-specific Nrf2 knockout (Nrf2 △

The Cyclin C-CDK8/19 Mediator kinase module controls PRCC-TFE3 driven senescence in renal epithelium and tumorigenesis in TFE3-RCC.

TFE3-rearranged renal cell carcinoma (TFE3-RCC) is an aggressive kidney cancer driven by oncogenic TFE3 fusion transcription factors, yet the molecular machinery that enables these fusions to reprogram transcription and drive tumor growth remains poorly defined. Here, we identify the Cyclin C-CDK8/19 Mediator kinase module as an essential co-regulator of TFE3 fusion driven transcriptional programs and tumorigenesis. Inducible expression of PRCC-TFE3 in HK-2 cells, immortalized from normal renal epithelial cells, triggered a robust oncogene-induced senescence (OIS) phenotype. Using OIS as a fun

PKM2 preconditioning protects endothelial cells from pyroptosis and BBB disruption via NRF2/TRX/TXNIP signaling in neonatal hypoxic-ischemic brain injury.

BACKGROUND: Neonatal hypoxic-ischemic brain damage (HIBD) is a leading cause of neurological deficits and death in neonates. In HIBD, the death of endothelial cells and disruption of the blood-brain barrier (BBB) are closely related to the severity of brain damage and long-term clinical outcomes. There is increasing evidence that a glycolytic enzyme, pyruvate kinase M2 (PKM2), is essential for managing metabolic processes in endothelial cells, but its role (and underlying molecular mechanism) in hypoxic-ischemic (HI)-associated endothelial cell metabolism, cell survival, and BBB function remai

GULP1 Fuels Resistance to PD-1 Blockade through Lipid-Mediated Metabolic Reprogramming of Tumor-Associated Macrophages in Gastric Cancer.

Background: Immune checkpoint inhibitors targeting programmed cell death protein 1 (PD-1) have substantially improved the treatment of multiple cancers; however, only a small proportion of gastric cancer (GC) patients achieve durable clinical benefit. The underlying tumor-intrinsic mechanisms driving resistance to anti-PD-1 therapy remain poorly understood. This study aimed to identify key regulators of immunotherapy resistance in GC and to elucidate their functional and mechanistic roles. Methods: Transcriptomic data from anti-PD-1-responsive and anti-PD-1-resistant GC patients were analyzed

The TWEAK-HOIP-HuR Axis: A Novel Mechanism of AMPK Inactivation and Metabolic Reprogramming in Lupus Nephritis Mesangial Cells Hyperproliferation.

Cellular proliferation is intrinsically coupled to metabolic reprogramming, a conserved biological association that modulates the progression of diverse proliferative pathologies. In the present study, we unveil a novel mechanistic link between tumor necrosis factor-like weak inducer of apoptosis (TWEAK) signaling and Human antigen R (HuR)-driven metabolic alterations in lupus nephritis (LN), a progressive renal disorder characterized by mesangial cells (MCs) hyperproliferation and glomerular dysfunction. We demonstrate that TWEAK stimulation elicits linear ubiquitination of AMP-activated prot
