Similarities in the histological mouse model of early herpes simplex retinopathy with punctate inner choroidopathy and Ebola virus disease retinopathy.

ABSTRACT PURPOSE: To highlight similarities between the cross-sectional retinal lesion appearance in a patient with punctate inner choroidopathy (PIC), Ebola virus disease (EVD) retinopathy and the von Szily mouse model of herpes simplex virus type 1 (HSV) retinopathy. METHODS: Case report and cross-sectional retinal lesion comparison. RESULTS: Whilst phenotypically different on colour imaging, a near-identical lesion appearance on optical coherence tomography, characterised by a focal photoreceptor loss and distinctive V-shaped collapse of the overlying retinal layers, was observed in both

An imbalance in autophagy contributes to retinal damage in a rat model of oxygen-induced retinopathy.

In retinopathy of prematurity (ROP), the abnormal retinal neovascularization is often accompanied by retinal neuronal dysfunction. Here, a rat model of oxygen-induced retinopathy (OIR), which mimics the ROP disease, was used to investigate changes in the expression of key mediators of autophagy and markers of cell death in the rat retina. In addition, rats were treated from birth to postnatal day 14 and 18 with 3-methyladenine (3-MA), an inhibitor of autophagy. Immunoblot and immunofluorescence analysis demonstrated that autophagic mechanisms are dysregulated in the retina of OIR rats and indi

Endothelial activation of caspase-9 promotes neurovascular injury in retinal vein occlusion.

Central nervous system ischemic injury features neuronal dysfunction, inflammation and breakdown of vascular integrity. Here we show that activation of endothelial caspase-9 after hypoxia-ischemia is a critical event in subsequent dysfunction of the blood-retina barrier, using a panel of interrelated ophthalmic in vivo imaging measures in a mouse model of retinal vein occlusion (RVO). Rapid nonapoptotic activation of caspase-9 and its downstream effector caspase-7 in endothelial cells promotes capillary ischemia and retinal neurodegeneration. Topical eye-drop delivery of a highly selective cas

PRPF31 reduction causes mis-splicing of the phototransduction genes in human organotypic retinal culture.

PRPF31 is ubiquitously expressed splicing factor and has an essential role in the pre-mRNA splicing in all tissues. However, it is not clear how reduced expression of this general splicing factor leads to retinal restricted disease, retinitis pigmentosa (RP). In this study, we used RNA interference and RNA-sequencing to mimic the PRPF31 haploinsufficiency in human organotypic retinal cultures (HORCs). We examined the effects of PRPF31 deficiency on splicing by analyzing the differential exon usages (DEUs) and intron retentions of the retinal transcriptome. Our results revealed that the PRPF31

Retinal nerve fiber layer thickening in ARSACS carriers.

PURPOSE: Two Caucasian Belgian families were diagnosed with autosomal recessive spastic ataxia of Charlevoix-Saguenay (ARSACS). The ophthalmological findings in both ARSACS disease and carriers are described. METHODS: In addition to a complete ophthalmological assessment, in both patients and carriers, spectral-domain Optical Coherence Tomography scans of the peri-papillary retinal nerve fiber layer were performed. RESULTS: Molecular analysis revealed a missense mutation which has not been reported before. Besides patients with ARSACS, who also presented additional ophthalmological abnormali

Identification of α-fodrin as an autoantigen in experimental coronavirus retinopathy (ECOR).

The coronavirus, mouse hepatitis virus (MHV), JHM strain induces a biphasic disease in BALB/c mice that consists of an acute retinitis followed by progression to a chronic retinal degeneration with autoimmune reactivity. Retinal degeneration resistant CD-1 mice do not develop either the late phase or autoimmune reactivity. A mouse RPE/choroid DNA expression library was screened using sera from virus infected BALB/c mice. Two clones were identified, villin-2 protein and α-fodrin protein. α-Fodrin protein was used for further analysis and western blot reactivity was seen only in sera from virus

Glutamate levels in aqueous humor of patients with retinal artery occlusion.

PURPOSE: We analyzed the levels of glutamate and other amino acids in aqueous humor of patients with retinal artery occlusion (RAO) to determine whether glutamate is associated with retinal ischemia in RAO. METHODS: Aqueous humor samples were obtained from nine RAO patients by paracentesis performed as an emergent therapeutic intervention and from nine cataract patients without other ocular disease who served as controls. Aspartic acid, glutamate, taurine, and alanine concentrations were determined using high-performance liquid chromatography. RESULTS: The glutamate level in aqueous humor of

Lipid hydroperoxide formation in the retina: correlation with retinal degeneration and light damage in a rat model of Smith-Lemli-Opitz syndrome.

The Smith-Lemli-Opitz syndrome (SLOS) is an autosomal recessive disease presenting with multiple congenital anomalies, caused by a defect in cholesterol biosynthesis that results in abnormally elevated levels of 7-dehydrocholesterol (7DHC). Progressive retinal degeneration has been demonstrated in a rat model of SLOS, which is markedly exacerbated by intense light, far more so than occurs in normal albino rats under the same conditions. Herein, we demonstrate that, by six postnatal weeks, retinas in the SLOS rat model contain levels of lipid hydroperoxides (LPOs) comparable to those found in l

Controversies in the treatment of cytomegalovirus retinitis: foscarnet versus ganciclovir.

Cytomegalovirus (CMV) retinitis is the most common intraocular infection in patients with AIDS. Untreated, CMV retinitis is a binding disease. Ganciclovir, a nucleoside analog, and foscarnet, a pyrophosphate analog, are both effective in controlling CMV retinitis. A randomized, controlled, comparative trial of foscarnet and ganciclovir demonstrated that they were equivalent in terms of controlling CMV retinits, but that foscarnet was associated with a longer survival, possibly due to an antiretroviral effect of foscarnet. However, foscarnet was less well tolerated than ganciclovir, primarily d

[Proliferative retinopathy in chronic myeloid leukemia].

BACKGROUND: In 50% of patients with acute and chronic myelocytic leukemia retinal alterations occur which are similar to those of nonproliferative diabetic retinopathy or hypertensive retinopathy. Retinal neovascularizations, however, are rarely seen and only found in patients with the chronic disease. PATIENTS AND METHODS: We report on the clinical course of three patients with advanced stage of proliferative retinopathy in CML. In two patients, the diagnosis of CML was established after ocular involvement. Therapy was planned according to the principles, which have proven effective in other

Fibrosis-related biomarkers and large and small vessel disease: the Cardiovascular Health Study.

OBJECTIVE: Fibrosis has been implicated in a number of pathological, organ-based conditions of the liver, kidney, heart, and lungs. The objective of this study was to determine whether biomarkers of fibrosis are associated with vascular disease in the large and/or small vessels. METHODS: We evaluated the associations of two circulating biomarkers of fibrosis, transforming growth factor-β (TGF-β) and procollagen type III N-terminal propeptide (PIIINP), with incident peripheral artery disease (PAD) and subclinical macrovascular (carotid intima-media thickness, flow-mediated vasodilation, ankle-

[Diabetes and peripheral arterial occlusive disease: therapeutic potential and pro-angiogenic strategies].

Cardiovascular complications are the leading cause of morbidity and mortality in patients with diabetes mellitus; up to 80% of deaths in patients with diabetes are closely associated with vascular disease. The ability of the organism to form a collateral network of blood vessels constitutes an important response to vascular occlusive disease and determines to a large part the clinical consequences and severity of tissue ischemia. The development of new vessels is significantly reduced in diabetic patients with coronary or peripheral artery disease. This probably contributes to the severe cours

[Adequacy of prescribing the disaggregants in vascular pathology of the eye fundus and in primary open-angle glaucoma].

The functional activity of blood platelets was studied in 936 patients with vascular pathology of the eye fundus (central chorioretinal dystrophy--CCRD; preproliferative diabetic retinopathy--PPDR; proliferative diabetic retinopathy--PDR; hypertensive retinopathy--HP; thrombosis of retinal veins--TRV; and primary open angle glaucoma--POAG) by using 2 methods: the routine one, which does not expose the real process of platelets' aggregation in the blood bed, and a method, which is closer to the conditions in vivo. The non-physiological ("tube"-based) methods of determination of platelets' aggre

Vitreoretinal traction and perimacular retinal folds in the eyes of deliberately traumatized children.

The pathophysiology of perimacular folds in eyes of deliberately traumatized children is disputed. The authors reviewed the clinical and forensic records and systemic and ocular findings at autopsy of three children with perimacular retinal folds who died after being violently shaken. Two of the children suffered direct head trauma in addition to being shaken; one patient was violently shaken without any physical or forensic evidence of direct head trauma. No direct ocular trauma was detected. In each case, the vitreous had partially separated from the retina but remained attached to the inter

Acute multifocal inner retinitis.

Two patients developed acute changes in vision two to four weeks after a febrile illness. On ophthalmic examination, each patient had bilateral vitreitis without anterior segment inflammation and multiple, bilateral, round, yellow-white inner retinal lesions that were located in the posterior pole and midperiphery. Laboratory tests did not contribute to a diagnosis. Symptomatic visual loss was caused by neuroretinitis and serous retinal detachment in one patient and by an occluded branch retinal artery in the other. The multifocal retinal lesions resolved gradually without treatment over sever

Congenital retinal fold as a sign of dominant exudative vitreoretinopathy.

Four eyes of three patients from two large families with dominant exudative vitreoretinopathy showed an elevated retinal fold extending from the posterior pole toward the inferotemporal periphery of the fundus. A congenital retinal fold (ablatio falciformis congenita) has to be considered as a sign rather than a diagnosis, and can be caused by a few underlying disorders, to which dominant exudative vitreoretinopathy can be added. There are no indications that the hyaloid system is involved in the pathogenesis of a retinal fold in the latter condition. The formation of a retinal fold in dominan

Feline histoplasmosis with ocular involvement.

A 12-year-old, castrated male cat with a history of respiratory disease developed bilateral endophthalmitis, retinal detachment, and granulomatous choroiditis, and unilateral glaucoma, and was killed. Other lesions included granulomatous optic neuritis, myositis involving extraocular muscles, and focal retinitis. Light and electron microscopy showed many intracellular organisms, interpreted to be Histoplasma. Granulomatous inflammatory lesions and organisms were present in lung, liver, lymphoid tissues, and adrenals.

Proliferative diabetic retinopathy in patients with defects of platelet function.

A 71-year-old woman had von Willebrand's disease, an inheritable abnormality of platelet activity, and developed diabetic retinal neovascularization. A 59-year-old man had a myeoloproliferative disorder with thrombocythemia but may have had retinal changes before the development of his platelet dysfunction. The occurrence of diabetic proliferative retinopathy in patients with reduced platelet activity suggests that platelet induced microcirculatory abnormalities may not be required for neovascularization.
