Efficacy and safety of patisiran for ATTRv-PN: a systematic review and meta-analysis.

BACKGROUND: Hereditary transthyretin amyloidosis (ATTRv; v for variant) with polyneuropathy is a rare, progressive, and fatal autosomal dominant disorder. Therapies such as liver transplantation and TTR stabilizations have limitations. Patisiran is a small interfering RNA (siRNA), offering potential as a genetic-level therapy for hereditary transthyretin amyloidosis with polyneuropathy (ATTRv-PN). However, evidence on patisiran's efficacy and safety for ATTRv-PN remains limited. OBJECTIVES: This study aimed to further clarify patisiran's efficacy and safety for ATTRv-PN by meta-analysis. DES…

Real-World Safety of Niraparib for Maintenance Treatment of Ovarian Cancer in Canada.

Niraparib was recently funded in Canada for the maintenance treatment of ovarian cancer following platinum-based chemotherapy. However, the drug's safety profile in the real world remains uncertain. We conducted a cohort study to describe the patient population using niraparib and the proportion that experienced adverse events between June 2019 and December 2022 in four Canadian provinces (Ontario, Alberta, British Columbia [BC], and Quebec). We used administrative data and electronic medical records from Ontario Health, Alberta Health Services, and BC Cancer, and registry data from Exactis In…

Using Vaccine Safety Data to Demonstrate the Potential of Pooled Data Analysis.

In Canada, vaccine safety studies are often conducted at the provincial/territorial level where the primary data on vaccination reside. Combining health services data from multiple jurisdictions using a pooled data analytic approach would reduce the amount of time needed to detect vaccine safety signals. To determine the difference in the time it would take to identify safety signals using different proportions of the Canadian population, we conducted power and sample size calculations for a hypothetical self-controlled case series-based surveillance analysis. We used scenarios modeled after t…

Safety and Efficacy of Immunization with a Late-Liver-Stage Attenuated Malaria Parasite.

BACKGROUND: Currently licensed and approved malaria subunit vaccines provide modest, short-lived protection against malaria. Immunization with live-attenuated Plasmodium falciparum malaria parasites is an alternative vaccination strategy that has potential to improve protection. METHODS: We conducted a double-blind, controlled clinical trial to evaluate the safety, side-effect profile, and efficacy of immunization, by means of mosquito bites, with a second-generation genetically attenuated parasite (GA2) - a mei2 single knockout P. falciparum NF54 parasite (sporozoite form) with extended deve…

Efficacy and safety of antithrombin supplementation in neonates and infants on a continuous heparin infusion.

BACKGROUND: Antithrombin (AT) is a natural anticoagulant and potent inhibitor of several coagulation proteins, including activated factor X (FXa) and FIIa. The therapeutic activity of heparin depends on the presence of AT. Levels of plasma AT are low in neonates and young infants compared to those in adults. Exogenous AT supplementation is postulated to enhance the activity of heparin and facilitate attainment of therapeutic anticoagulation in infants. OBJECTIVES: To describe the efficacy and safety of AT administration in infants on a continuous heparin infusion. METHODS: Retrospective coho…

To avoid or not to avoid: impact of self-compassion on safety behaviors in social situations.

BACKGROUND/OBJECTIVES: Safety behaviors are commonly used to decrease anxiety in social settings but maintain anxiety. Self-compassion has been shown to reduce anxiety and rumination, but the impact on safety behaviors has not been examined. For the present studies, it was hypothesized that inducing self-compassion would lead to lower safety behaviors compared to controls. METHODS: In Study 1 (N = 390), participants with elevated social anxiety recalled a distressing social situation, were randomly assigned to a self-compassionate (n = 186) or control (n = 204) writing exercise, and then repo…

Effectiveness, safety, and patient satisfaction of carboxytherapy as an adjunctive treatment for periorbital hyperpigmentation.

INTRODUCTION: Dark under-eye circles or periorbital hyperpigmentation constitute a prevalent and challenging cosmetic problem with diverse etiologies and types. While modifying exacerbating habits can provide partial relief for the pigmentary and vascular factors associated with this condition, and despite the abundance of available treatment options, there is currently a lack of gold-standard evidence-based treatments proposed for curing this disorder. OBJECTIVES: This study aimed to assess the safety and effectiveness of carboxytherapy in treating periorbital hyperpigmentation. MATERIAL AN…
