[Beryllium--underestimated occupational health hazard in Poland].

Beryllium is a metal used in different branches of industry. Inhalation is the most common way of exposure. Beryllium shows a high affinity to the lung tissue. It may act as toxic, carcinogenic and immunological agent. Chronic beryllium disease (CDB) is very difficult to diagnose because of its similarity to sarcoidosis. The lymphocyte proliferation assay is a very helpful test in CBD diagnosis. It seems reasonable to include this test into the body of the standard diagnostic procedures in workers exposed to beryllium. Early detection of hypersensitivity to beryllium, before CBD symptoms occur

The carcinogenicity of chrysotile asbestos.

In in vitro test systems, chrysotile is markedly toxic, causes chromosomal aberrations, and is capable of inducing morphological and preneoplastic transformation. In carefully designed animal experiments, chrysotile produces lung cancer and mesothelioma as effectively as do the amphiboles tested. Human population studies do not refute these experimental results. Chrysotile asbestos is carcinogenic to humans, especially for the induction of lung cancer and mesothelioma in exposed populations. For cancers of other sites, with the exception of laryngeal and possibly gastrointestinal cancer, the e

Lacticaseibacillus rhamnosus GG enhances gefitinib efficacy and reduces treatment-related toxicity via propionyl-L-carnitine regulation.

Gefitinib improves outcomes in patients with EGFR-mutant non-small cell lung cancer; however, treatment-related gastrointestinal and hepatic toxicity can limit its long-term use. Here, we investigate whether probiotic strains can improve gefitinib efficacy while reducing treatment-associated toxicity. Among the tested strains, Lacticaseibacillus rhamnosus GG (LGG) shows the strongest enhancement of gefitinib efficacy while reducing intestinal and hepatic injury without altering systemic gefitinib exposure. Metabolomic analyses identify propionyl-L-carnitine as an LGG-associated metabolite dete

Clinical Features of Hypo- and Hypervitaminosis of Fat-Soluble Vitamins in Pediatric Patients.

Fat-soluble vitamins (A, D, E, and K) are crucial for pediatric health, contributing to normal cellular function, growth, immune defense, and development. Unlike water-soluble vitamins, they are absorbed with dietary fats and stored in the liver and adipose tissue, leading to risks of deficiency (hypovitaminosis) and toxicity (hypervitaminosis) in certain physiological and pathological conditions. This narrative review aimed to summarize the clinical manifestations, diagnostic considerations, and management of hypo- and hypervitaminosis of fat-soluble vitamins in pediatric populations. A compr

Synergistic toxicity of methylmercury and cadmium through NRF2 suppression and mercury retention.

Methylmercury (MeHg) is a potent environmental toxicant that frequently coexists with other heavy metals, raising concerns about combined toxic effects. Increasing evidence indicates that co-exposure to multiple metals can lead to synergistic or greater-than-additive effects; however, the molecular mechanisms underlying such interactions remain poorly understood. Among the tested metals, only co-exposure with Cd markedly enhanced MeHg cytotoxicity. Here, our objective was to evaluate the impact of MeHg co-exposure on cytotoxicity of various heavy metals in HeLa cells. We used cell viability as

The role of stem cells in the pharmacological assessment of nanoparticles.

Stem cells aid in the pharmacological assessment of nanoparticles by serving as a human-relevant, dynamic, and scalable model. Their integration into nanoparticle research enhances toxicity screening and therapeutic optimization by evaluating absorption, distribution, metabolism, excretion and toxicity (ADMET) parameters across models ranging from simple in vitro cultures to complex organoids. They provide insights into how nanoparticles interact with biological systems, guiding the rational design of safer, more effective nanomedicines. Conventional in vitro and animal models often fall short

Mechanism of reproductive toxicity of BHT on male Ruditapes philippinarum: epigenetic - testis development and oxidative damage - cell death.

Synthetic phenolic antioxidants (SPAs) are widely utilized in industrial production and consumer goods owing to their excellent antioxidant properties. However, their potential for multiple toxic effects, coupled with the increasing levels of environmental pollution, has heightened public concern. Currently, there is a relative scarcity of study on the reproductive toxicology of SPAs in hydrobios, with a particular dearth of studies in invertebrates. This study investigates the reproductive toxicity of the typical SPAs butylhydroxytoluene (BHT) in male clams Ruditapes philippinarum across dist

Non-Targeted Analysis (NTA) of Plasma and Liver from Sprague Dawley Rats Exposed to Perfluorohexanesulfonamide (PFHxSA), a Precursor to Perfluorohexane Sulfonic Acid (PFHxS).

High-resolution accurate mass non-targeted analysis (NTA) is a useful discovery tool for metabolite characterization of in vivo dosing studies since it enables detection of both predicted and unexpected biotransformation products. We used NTA to investigate biotransformation of perfluorohexanesulfonamide (PFHxSA) in plasma and liver from male and female Sprague Dawley rats after a 5-day repeat exposure study. PFHxSA is an emerging per- and polyfluoroalkyl substance (PFAS) with unknown toxicity and a potentially reactive headgroup. NTA revealed the presence of predicted in vivo biotransformatio

Acute Toxicity of Commercial Wildfire Retardants to Two Daphniid Species (Ceriodaphnia dubia and Daphnia magna).

In the face of global climate change, there has been an increase in wildfires around the world, highlighting the need for improved firefighting techniques, such as the use of fire retardants (FRs). These products can enter aquatic systems directly or through runoff, posing potential risks to aquatic biota. In this study, the acute toxicity (24-h/48-h EC50) of three distinct FRs (N-Borate, N-Phosphate+, and N-Phosphate-) was assessed on the immobility of freshwater microcrustaceans Ceriodaphnia dubia and Daphnia magna. The toxicity of the FRs varied up to two orders of magnitude, all of which p

Toxicity assessment of di(2-ethylhexyl) phthalate using zebrafish embryos: Cardiotoxic potential.

Plasticizers are considered as newly emerged contaminants. They are added to plastics to increase their flexibility and softness. Phthalate plasticizers including the Di-2-ethylhexyl phthalates (DEHP) are toxic and induce adverse effects on the different organization levels of the environment. In the current study, we investigated the potential toxicity of DEHP using Zebrafish as a biological model. Five ascending concentrations of DEHP were tested in embryos throughout 96 hpf: 0.0086, 0.086, 0.86, 8.6, and 86 mg/L. Embryotoxicity assessments revealed limited lethal effects on DEHP-exposed emb

Mycotoxins: Emerging toxic mechanisms, and unanswered research questions.

Recently, a series of toxic mechanisms have been explored in mycotoxins. Emerging evidence show that mycotoxins may induce human neurodegenerative diseases (ND); however, this idea is still unproven. Besides to identify this hypothesis, some questions, for example, how the mycotoxins induce this disease and what the molecular mechanism is, as well as whether the brain-gut axis is involved in this context, should be answered. Very recent studies further reported an "immune evasion" mechanism in trichothecenes; moreover, hypoxia seems to play important function in this process; nevertheless, whe

A cytotoxicity and mechanistic investigation of mono- and di-chloro naphthalenes.

Polychlorinated naphthalenes (PCNs) were characterized as persistent organic pollutants (POPs) that were widely distributed in the environment. Although the striking in vivo toxicity of these pollutants towards both animals and humans was well documented, their cytotoxicity and mechanism of action have not been extensively investigated. In this study, the in vitro antiproliferative activity of mono- and di-chloronaphthalenes as representative PCNs were evaluated and the results indicated strong growth inhibitory effects against mammalian cells, especially the human breast MCF-10A cell and huma

Phloroglucinol Derivative Carbomer Hydrogel Accelerates MRSA-Infected Wounds' Healing.

Globally, wound infection is considered to be one of the major healthcare problems, with bacterial infections being the most critical threat, leading to poor and delayed wound healing, and even death. As a superbug, methicillin-resistant Staphylococcus aureus (MRSA) causes a profound hazard to public health safety, prompting us to search for alternative treatment approaches. Herein, the MTT test and Hoechst/propidium iodide (PI) staining demonstrated that PD was slightly less toxic to human fibroblasts including Human keratinocytes (HaCaT) cell line than Silver sulfadiazine (SSD), and Vancomyc

Evaluation of phytotoxicity and cytotoxicity of industrial catalyst components (Fe, Cu, Ni, Rh and Pd): A case of lethal toxicity of a rhodium salt in terrestrial plants.

Until recently, chemical derivatives of platinum group metals have not been in a systematic direct contact with living organisms. The situation has changed dramatically due to anthropogenic activity, which has led to significant redistribution of these metals in the biosphere. Millions of modern cars are equipped with automotive catalytic converters, which contain rhodium, palladium and platinum as active elements. Everyday usage of catalytic technologies promotes the propagation of catalyst components in the environment. Nevertheless, we still have not accumulated profound information on poss
