Barriers in managing maternal schistosomiasis: a qualitative study of healthcare providers in Tanzania.

BACKGROUND: Maternal schistosomiasis threatens the health of pregnant women and their foetuses in endemic regions. Although praziquantel is recommended for treatment, its utilization remains low. This study explored the experiences of healthcare providers (HCPs) with schistosomiasis, particularly maternal schistosomiasis, and their perceived management barriers in Tanzania. METHODS: This qualitative case study was conducted in October 2024 with nine purposefully selected HCPs. In-depth interviews guided by a semi-structured tool analysed HCPs' experiences with schistosomiasis, maternal schist

Travelers Returning to Denmark with Hypereosinophilia and Ancylostoma Infections Detected Using Broad-Specificity Amplicon-Based Next-Generation Sequencing.

In nonendemic countries such as Denmark, hookworm infections in travelers may be rare and difficult to detect because of low-grade shedding of parasite eggs and a lack of clinical suspicion and relevant diagnostic capacity. Two cases of hookworm infection observed in 2025 that were detected only through amplicon-based next-generation sequencing (NGS) of fecal DNA are presented in the current study. Both infections were characterized by hypereosinophilia and acquired during 2-week holidays in Asia. Case 1 was infected with Ancylostoma ceylanicum during a holiday in the Philippines, and Case 2 a

The therapeutic potential of green synthesized zinc oxide nanoparticles in murine schistosomiasis.

Schistosomiasis remains a major neglected tropical disease, affecting approximately 600 million individuals worldwide and accounting for nearly 500,000 deaths annually. The principal causative species-Schistosoma haematobium, S. mansoni, and S. japonicum-drive significant morbidity through hepatomegaly, splenomegaly, and progressive hepatic fibrosis. Praziquantel (PZQ) remains the cornerstone of treatment; however, its limited efficacy against immature worms and eggs, combined with concerns over emerging drug resistance, underscores the urgent need for novel therapeutic alternatives. This stud

The process of residual calcification following antiparasitic treatment in the pig model of neurocysticercosis is dynamic.

BACKGROUND: Calcified neurocysticercosis (NCC), the end stage of brain cysts of the pork tapeworm Taenia solium is a common cause of epilepsy. Calcified NCC lesions are not inert and represent potential epileptogenic foci. Understanding the mechanisms of residual calcification in NCC is hindered by the difficulty of accessing human brain biopsies. Since cyst degeneration can be induced by antiparasitic treatment (APT) in NCC-infected pigs, this study assessed the residual calcification process in this model at three time points after APT. METHODS/PRINCIPAL FINDINGS: Fifteen naturally infected

Combination therapy with 3-Hydroxybenzaldehyde and Albendazole modulates mitochondrial protein expression in astrocytes after Angiostrongylus cantonensis infection.

BACKGROUND: Angiostrongylus cantonensis is a zoonotic nematode that causes eosinophilic meningitis and central nervous system injury in humans; 3-hydroxybenzaldehyde (3-HBA) is a benzaldehyde compound that exhibits antioxidant and anti-inflammatory activities. Brain injury promotes Ca²⁺ influx and mitochondrial Ca²⁺ loading via voltage-dependent anion channel 1 (VDAC1) and the mitochondrial Ca²⁺ uniporter (MCU), leading to mitochondrial dysfunction and cytochrome c-mediated apoptosis. METHODOLOGY/PRINCIPAL FINDINGS: This study aimed to evaluate the therapeutic effects of 3-HBA combined with a

The first molecular detection of benzimidazole resistance in Haemonchus contortus from sheep in Hejing and Minfeng counties of Southern Xinjiang.

To understand the benzimidazole (BZ) resistance of Haemonchus contortus in Southern Xinjiang, three single nucleotide polymorphisms (SNPs) designated as F167Y, E198A, and F200Y, in the isotype-1 β-tubulin gene which are associated with BZ resistance, were investigated for H. contortus populations from sheep in Hejing and Minfeng counties of Southern Xinjiang. In brief, a total of 190 H. contortus adults were collected from 52 out of 70 slaughtered sheep in city abattoirs across two regions in Southern Xinjiang. The species identity of each adult worm was confirmed by PCR amplification of ITS-2

Improving the Cost-efficiency of Preventive Chemotherapy: Impact of New Diagnostics on Stopping Decisions for Control of Schistosomiasis.

BACKGROUND: Control of schistosomiasis (SCH) relies on the regular distribution of preventive chemotherapy (PC) over many years. For the sake of sustainable SCH control, a decision must be made at some stage to scale down or stop PC. These "stopping decisions" are based on population surveys that assess whether infection levels are sufficiently low. However, the limited sensitivity of the currently used diagnostic (Kato-Katz [KK]) to detect low-intensity infections is a concern. Therefore, the use of new, more sensitive, molecular diagnostics has been proposed. METHODS: Through statistical an

Modeling Approaches to Predicting Persistent Hotspots in SCORE Studies for Gaining Control of Schistosomiasis Mansoni in Kenya and Tanzania.

BACKGROUND: Some villages, labeled "persistent hotspots (PHS)," fail to respond adequately in regard to prevalence and intensity of infection to mass drug administration (MDA) for schistosomiasis. Early identification of PHS, for example, before initiating or after 1 or 2 years of MDA could help guide programmatic decision making. METHODS: In a study with multiple rounds of MDA, data collected before the third MDA were used to predict PHS. We assessed 6 predictive approaches using data from before MDA and after 2 rounds of annual MDA from Kenya and Tanzania. RESULTS: Generalized linear model

Village Response to Mass Drug Administration for Schistosomiasis in Mwanza Region, Northwestern Tanzania: Are We Missing Socioeconomic, Cultural, and Political Dimensions?

Praziquantel (PZQ)-based mass drug administration (MDA) is the main approach for controlling schistosomiasis in endemic areas. Interventions such as provision and use of clean and safe water, minimizing contacts with infested water, disposal of human waste in latrines, and snail control provide additional key interventions to break the transmission cycle and could complement and perhaps sustain the benefits of MDA. However, all interventions deployed need to be accepted by the targeted communities. A qualitative study was conducted to examine factors that might differentiate villages which did

Exploring the complex associations over time among albendazole treatment, cyst evolution, and seizure outcomes in neurocysticercosis.

OBJECTIVE: To develop a causal model for the occurrence of neurocysticercosis (NC)-related seizures and test hypotheses generated from the model. METHODS: We used data from a randomized controlled trial comparing albendazole with placebo among patients newly diagnosed with NC. Based on our causal model, we explored the associations among albendazole treatment, NC cyst evolution, and seizure outcomes over 24 months of follow-up using generalized linear mixed effect models. RESULTS: We included 153 participants, of whom 51% received albendazole. The association between seizure outcomes and tre

Levamisole-induced vasculitis.

Levamisole is a veterinary antihelminthic, chemotherapeutic agent, and immunomodulator that also is used as an adulterant and cutting agent in cocaine distribution. This drug may potentiate the sympathomimetic actions of cocaine and can cause neutropenia, agranulocytosis, purpuric retiform lesions, and skin necrosis. This article describes two cases of suspected levamisole-induced vasculitis. No standardized diagnostic or treatment algorithm exists for this challenging condition. Diagnosis and treatment require a multidisciplinary team approach.

Praziquantel Resistance in the Zoonotic Cestode Dipylidium caninum.

Dipylidium caninum is a cosmopolitan cestode infecting dogs, cats, and humans. Praziquantel is a highly effective cestocidal drug and resistance in adult cestodes has not been reported. From 2016 to 2018, a population of dogs with cestode infections that could not be eliminated despite multiple treatments with praziquantel or epsiprantel was identified. Cases of D. caninum were clinically resistant to praziquantel and could not be resolved despite increasing the dose, frequency, and duration of treatment. Resistant isolates were identified and characterized by sequencing the 28S, 12S, and volt

Toxicology of Avermectins and Milbemycins (Macrocyclic Lactones) and the Role of P-Glycoprotein in Dogs and Cats.

Overdoses of macrocyclic lactones in dogs and cats can result in such signs as tremors, ataxia, seizures, coma, and blindness. Dogs with the ABCB1-1Δ gene defect are predisposed to macrocyclic lactone toxicosis at lower dosages than dogs without the defect. Intravenous lipid emulsion therapy has been suggested for treatment of macrocyclic lactone toxicosis but evidence of efficacy is limited. Initial decontamination and supportive care remain the mainstays of therapy for macrocyclic lactone toxicosis.
