Regulatory controls of oncotrophoblast proteins and developmental alkaline phosphatases in cancer cells.

The two oncotrophoblast proteins, Regan isoenzyme (placental-type alkaline phosphatase) and human chorionic gonadotrophin, are readily studied oncodevelopmantal gene products in human cancer patients and in three experimental model systems. The latter consists of (a) HeLa sublines TCRC-1 and TCRC-2, which produce Regan and non-Regan isoenzymes, (b) HEp-2 and FL amnion cell lines as models for the reciprocal expression of developmental genes, and (c) modulation in vivo of developmental gene expression in HeLa cells. In the case of the third model, for example, HeLa TCRC-1 cells grow in immunosu

Mediastinal Mature Teratoma With Malignant Carcinomatous Transformation (Somatic-Type Malignancy) With Metastatic Course.

We report the case of a 76-year-old patient presenting with an anterior mediastinal heterogeneous mass. Surgical biopsy revealed a solid and cystic lesion filled with hair. Pathological examination showed an atypical papillary epithelial lining without other germ cell tumor or immature teratoma associated. The final diagnosis was a mature teratoma of the mediastinum with somatic-type malignancy (carcinomatous transformation). After 8-month follow-up, subcutaneous and lymph node metastatic lesions of the carcinomatous component were identified. Subtyping of the malignant component within germ c

HNRNPA2B1 Orchestrates Immune Evasion in Colorectal Cancer by Rewiring Tumor-Immune Cell Interactions and Suppressing CD8+ T-cell Infiltration.

Immune checkpoint blockade (ICB) has transformed colorectal cancer therapy, yet the majority of microsatellite-stable colorectal cancers remain refractory because of insufficient tumor-immune cell cross-talk. Identifying molecular regulators that modulate the tumor immune microenvironment (TIME) is crucial for expanding ICB efficacy. In this study, we identified HNRNPA2B1, an RNA-binding protein prominently upregulated in colorectal cancer, as a key driver of immune evasion. Despite low cytotoxicity to normal cells, HNRNPA2B1 rewired the TIME by suppressing Cxcl9/Cxcl10-Cxcr3 signaling, CD8+ T

High expression of SENP1 correlates with poor prognosis and immune evasion in gastric cancer.

Gastric cancer (GC) remains a highly prevalent malignancy with poor prognosis in advanced stages. SUMOylation, a key post-translational modification, regulates tumor progression and immune responses, yet the role of SUMO-specific protease 1 (SENP1) in GC remains unclear. Here, we show that SENP1 is significantly overexpressed in GC tissues and cell lines, as demonstrated by analyses of TCGA/GEO datasets and by immunohistochemistry, western blotting, and qRT-PCR. Higher SENP1 levels were associated with more advanced disease, undifferentiated histology, systemic dissemination, and worse surviva

Single-cell Atlas reveals core function of CPVL/MSR1 expressing macrophages in the prognosis of triple-negative breast cancer.

BACKGROUND: Triple-negative breast cancer (TNBC) is the most aggressive subtype of breast cancer, with the worst prognosis among all subtypes. The impact of distinct cell subpopulations within the tumor microenvironment (TME) on TNBC patient prognosis has yet to be clarified. METHODS: Utilizing single-cell RNA sequencing (scRNA-seq) integrated with bulk RNA sequencing (bulk RNA-seq), we applied Cox regression models to compute hazard ratios, and cross-validated prognostic scoring using a GLMNET-based Cox model. Cell communication analysis was used to elucidate the potential mechanisms of CPVL

Ultrastructure of cholangiocarcinoma associated with opisthorchiasis.

An electron microscopic study was carried out on eleven surgical liver biopsy specimens obtained from patients with cholangiocarcinoma associated with opisthorchiasis. The tumor cells of histologically well differentiated cholangiocarcinoma had few cytoplasmic organelles. They contained relatively large nuclei, abundant free ribosomes and numerous groups of fine fibrils. Each cell was surrounded by a basement membrane. Numerous long microvilli were seen projecting into the glandular lumen. The moderately differentiated cholangiocarcinomatous cells exhibited increased organelle content, marked

Combination Hormonal Drug Therapy Inhibits In Vitro Growth of Adult Ovarian Granulosa Tumor Cells.

This study investigated the mechanism of action and in vitro efficacy of an anti-hormonal combination drug regimen in treating adult-type granulosa cell tumors of the ovary (AGCT). Using patient-derived and commercial AGCT cell lines, the IC50 values of bicalutamide, anastrozole, and leuprolide acetate were established alone and in combination via cell proliferation assays. Averaged across cell lines, IC50 was determined for bicalutamide (IC50: 7.3 µM) and anastrozole (IC50: 6.2 µM). Minimal effect was seen with leuprolide acetate alone. Suppression of proliferation was increased with the comb

Cytotoxic factors secreted by cells infected by human immunodeficiency virus type I.

Conditioned media from human immunodeficiency virus type I (HIV-1) infected cells were tested for cytotoxic cell-derived factors. The assay used a murine fibroblast cell line which is sensitive to the effects of tumor necrosis factors, but nonpermissive for HIV-1 replication. Cytotoxic activity was detected in cultures of peripheral blood mononuclear cells infected with HIV-1. However, no differences in activity were found in conditioned media from infected lymphoid or monocytoid cell lines compared to their uninfected counterparts. These data suggest that cytotoxic activities of this type are

Optical imaging of progenitor cell homing to patient-derived tumors.

Capitalizing on cellular homing to cancer is a promising strategy for targeting malignant cells for diagnostic, monitoring and therapeutic purposes. Murine C17.2 neural progenitor cells (NPC) demonstrate a tropism for cell line-derived tumors, but their affinity for patient-derived tumors is unknown. We tested the hypothesis that NPC accumulate in patient-derived tumors at levels detectable by optical imaging. Mice bearing solid tumors after transplantation with patient-derived leukemia cells and untransplanted controls received 10(6) fluorescent DiR-labeled NPC daily for 1-4 days, were imaged

Expression level of CD117 (KIT) on ovarian cancer extracellular vesicles correlates with tumor aggressiveness.

Ovarian cancer is known to be the most lethal malignancy among all gynecological cancers affecting a large number of women worldwide. The treatment of ovarian cancer is challenging due to the high recurrence rate of the disease and is further complicated by acquired chemoresistance. Most ovarian cancer deaths are the result of the metastatic spread of drug-resistant cells. The theory of cancer stem cells (CSC) suggests that both tumor initiation and progression are driven by a population of undifferentiated capable of self-renewal, tumor initiation and development of chemoresistance. The CD117

TWIST1 induces expression of discoidin domain receptor 2 to promote ovarian cancer metastasis.

The mesenchymal gene program has been shown to promote the metastatic progression of ovarian cancer; however, specific proteins induced by this program that lead to these metastatic behaviors have not been identified. Using patient derived tumor cells and established human ovarian tumor cell lines, we find that the Epithelial-to-Mesenchymal Transition inducing factor TWIST1 drives expression of discoidin domain receptor 2 (DDR2), a receptor tyrosine kinase (RTK) that recognizes fibrillar collagen as ligand. The expression and action of DDR2 was critical for mesothelial cell clearance, invasion

Hypomethylating agents increase L1 retroelement expression without inducing novel insertions in myeloid malignancies.

Retroelements in the human genome are silenced via multiple mechanisms, including DNA methylation, to prevent their potential mutagenic effect. Retroelement activity, demonstrated by their expression and somatic retrotransposition events, was shown to be deregulated in multiple tumors but not yet in leukemia. We hypothesized that treatment with hypomethylating agents, commonly used in myelodysplastic syndromes and acute myeloid leukemia, could lead to increased retroelement activity and somatic retrotranspositions, thus contributing to disease progression. To address this hypothesis, we induce

Lymphocytic choriomeningitis in laboratory personnel exposed to hamsters inadvertently infected with LCM virus.

Seven of 27 students and staff oa a university laboratory became ill with lymphocytic choriomeningitis (LCM) following exposure over an 18-month period to hamsters implanted with tumors. The clinical symptoms were compatible with LCM infection and all the patients' sera had fluorescent antibody to LCM virus; 5 of them also had complement-fixing (CF) antibodies. Complement-fixing antibodies were found in 9 of 9 hamsters with tumors and 16 of 31 without tumors (excluding 6 hamsters with anticomplementary sera) and in 8 of 14 rabbits, but in none of 49 rats. Lymphocytic choriomeningitis virus was

Scorpion (Androctonus crassicauda) venom limits growth of transformed cells (SH-SY5Y and MCF-7) by cytotoxicity and cell cycle arrest.

The purpose of study was to examine the cytotoxic and anti-cancer properties along with addressing the plausible pathway followed by scorpion venom to reduce cell viability in SH-SY5Y and MCF-7 cells. Following exposure of cells with scorpion venom, cytotoxicity was estimated using MTT and lactate dehydrogenase assays. Apoptotic effects were measured by assessment of mitochondrial membrane potential, reactive nitrogen species, DNA fragmentation, and caspase-3 activity whereas antiproliferative effect was assayed using BrdU incorporation. Our results indicate that scorpion venom causes suppress

Counterintuitive production of tumor-suppressive secretomes from Oct4- and c-Myc-overexpressing tumor cells and MSCs.

Background: Advanced breast cancer frequently metastasizes to bone, but inhibiting tumor progression in chemotherapy may occasionally enhance tumorigenesis. Here, we employed a counterintuitive approach of overexpressing Yamanaka factors (Oct4, c-Myc, Sox2, and Klf4) and examined a conditioned medium (CM)-based treatment option with induced tumor-suppressing cells (iTSCs). Methods:In vitro proliferation and migration assays were conducted using tumor cell lines derived from breast cancer, as well as prostate and pancreatic cancers, and osteosarcoma. The tumor-suppressing capability of iTSC-der

Pterostilbene as a potential novel telomerase inhibitor: molecular docking studies and its in vitro evaluation.

Pterostilbene is a naturally occurring dimethyl ether analog of resveratrol identified in several plant species. Telomerase is important in tumor initiation and cellular immortalization. Given the striking correlations between telomerase activity and proliferation capacity in tumor cells, telomerase had been considered as a potentially important molecular target in cancer therapeutics. Molecular docking studies were performed on pterostilbene with the crystal structure of telomerase (3DU6). Pterostilbene was evaluated for its in vitro cytotoxicity in breast (MCF7) and lung cancer (NCI H-460) c

The anti-aging Klotho protects glioblastoma macrophages from radiotherapy-induced inflammation and predicts immunotherapy response.

Immunosuppressive myeloid cells, such as microglia and macrophages, play a key role in mediating resistance to immunotherapy in glioblastoma patients. Bulk RNA sequencing analysis revealed elevated expression of Klotho (Kl) in gliomas derived from irradiated glioma-bearing mice. Klotho, which encodes an anti-aging protein, was found to be upregulated in glioma-associated microglia/macrophages (GAMs) exhibiting an M1 pro-inflammatory phenotype. This upregulation appeared to enhance the antitumor efficacy of a combination of radiotherapy and dendritic cell (DC) immunotherapy. Furthermore, transc

Oncolytic virus with an immune stimulatory payload for osteosarcoma therapy.

Osteosarcoma (OS) is a highly aggressive bone malignancy that predominantly affects children and young adults. There have been no major changes in patient survival in the past four decades. Therefore, new therapeutic interventions are needed. We have developed an enhanced conditionally replicative canine oncolytic adenovirus, CAV2-AU-M3, armed with an anti-PD1 heavy-chain antibody (HcAb), and evaluated its efficacy against osteosarcoma across four canine cell lines. CAV2-AU-M3 was characterized for its infectivity, lytic properties, and anti-PD1 Ab production in monolayer and spheroid cultures
