Clinical characteristics and survival of trisomy 13 in a medical center in Taiwan, 1985-2004.

BACKGROUND: This study investigated the survival and natural history of trisomy 13 in a series of patients, comparing the management and outcome before and after the implementation of Taiwan's National Health Insurance program (NHI). METHODS: A total of 28 cases of trisomy 13 seen at Mackay Memorial Hospital, Taipei, Taiwan, from 1985 to 2004 were retrospectively reviewed. Survival and management before (12 cases) and after (16 cases) the implementation of National Health Insurance were compared, and structural defects, imaging findings, and cytogenetic results were analyzed. The cases that w

3D pattern of brain abnormalities in Williams syndrome visualized using tensor-based morphometry.

UNLABELLED: Williams syndrome (WS) is a neurodevelopmental disorder associated with deletion of approximately 20 contiguous genes in chromosome band 7q11.23. Individuals with WS exhibit mild to moderate mental retardation, but are relatively more proficient in specific language and musical abilities. We used tensor-based morphometry (TBM) to visualize the complex pattern of gray/white matter reductions in WS, based on fluid registration of structural brain images. METHODS: 3D T1-weighted brain MRIs of 41 WS subjects (age [mean+/-SD]: 29.2+/-9.2 years; 23F/18M) and 39 age-matched healthy contr

The association of abnormal alpha-fetoprotein and adverse pregnancy outcome: does increased fetal surveillance affect pregnancy outcome?

OBJECTIVE: Our purpose was to investigate whether adverse outcomes associated with elevated maternal serum alpha-fetoprotein levels may be prevented by intensive antenatal monitoring. STUDY DESIGN: Records of patients with elevated maternal serum alpha-fetoprotein values of > or =2.0 multiples of the median between 1995 and 1999 were reviewed. Pregnancy histories were analyzed to determine whether intensive antenatal monitoring (twice-weekly nonstress tests and determinations of the amniotic fluid index) would have detected the adverse outcomes when routine obstetric care would have missed th

Alzheimer's disease: preliminary study of spatial distribution at birth place.

Alzheimer's disease (AD) is a neurodegenerative disorder which is characterized by a progressive loss of memory and the alteration of cognitive functions. At least three chromosomal segments have been associated with early-onset AD in genetic linkage studies. These results argue for a certain degree of heterogeneity in the genetic origin of some forms of AD, although environmental risk factors cannot be ruled out in late-onset AD. In this preliminary study, we analyzed the geographical distribution of the birth places of a sample of 235 AD cases born in a defined region of Quebec (Canada), bet

Expression of the retinoblastoma susceptibility gene in childhood rhabdomyosarcomas.

BACKGROUND: Alveolar and embryonal rhabdomyosarcomas are soft-tissue tumors that occur mainly in childhood. The more aggressive alveolar subtype has been found to possess a characteristic chromosomal abnormality located near the retinoblastoma susceptibility gene (RB1). RB1 is a tumor suppressor gene implicated in the development of retinoblastoma and other, unrelated malignancies, especially osteogenic sarcomas and other second malignancies in retinoblastoma survivors. PURPOSE: The goals of our study were (a) to determine whether abnormalities of RB1 occur routinely in sporadic rhabdomyosarc

Inherited XX sex reversal in the cocker spaniel dog.

Nine XX true hermaphrodites and two XX males were discovered in a family of American cocker spaniels. The true hermaphrodites were partially-masculinized females with ovotestes; the XX males had malformed male external genitalia and cryptorchid aspermatogenic testes. Wolffian and Mullerian duct derivatives were present in both true hermaphrodites and XX males. All four sires of sex-reversed dogs were normal XY males; five of the dams were anatomically normal females and one was an XX true hermaphrodite. A second true hermaphrodite reproduced as a female, producing anatomically normal offspring

Genetic diseases underlying a spectrum of fetal effusions.

OBJECTIVES: Nonimmune hydrops fetalis is well understood to be heterogenous and the common endpoint of many genetic diseases. However, less is known about the prevalence and presenting features of genetic diseases that underlie other types of fetal effusions such as single effusions, leaving uncertainty in clinical practice about optimal approaches to testing and counseling for these pregnancies. We aimed to determine the diagnostic yield of exome sequencing by type of fetal effusion and presence of concurrent structural abnormalities and to identify the unique presenting features of underlyin

Human cytomegalovirus antagonizes SMC5/6 driven genome silencing via UL35 instituted proteasomal degradation of SLF2.

Recent studies demonstrated that the structural maintenance of chromosomes complex 5/6 (SMC5/6) acts as an important antiviral restriction factor that silences episomal DNA. Consequently, viruses have evolved strategies to antagonize this defence system. Interestingly, it was shown that SMC5/6-mediated silencing of extrachromosomal DNA depends on its recruitment to PML nuclear bodies which is mediated by a protein termed SMC5/6 localization factor 2 (SLF2). Since human cytomegalovirus (HCMV) dissociates PML nuclear bodies (PML-NBs) during the first hours of infection, we asked for the fate of

Genome-wide characterization of the AP2/ERF gene family in Scutellaria baicalensis Georgi reveals root-preferential and MeJA-responsive candidate genes.

APETALA2/ethylene-responsive factor (AP2/ERF) transcription factors play central roles in plant growth, development and environmental responses. Scutellaria baicalensis is a medicinal plant that predominantly accumulates pharmacologically important flavones in its roots, and methyl jasmonate (MeJA) induced these flavones accumulation. However, the AP2/ERF family has not been systematically characterized in S. baicalensis, and members showing root-preferential expression and MeJA responsiveness remain unknown. Here, we identified 137 SbAP2/ERF genes at the genome-wide level and investigated the

SNP-Based Chromosomal Microarray Analysis in the Era of Optical Genome Mapping: An Enriched Case-Series Evaluating Copy-Neutral Events.

BACKGROUND/OBJECTIVES: Chromosomal microarray analysis (CMA) is an essential tool in modern cytogenetics for detecting copy number alterations and copy-neutral loss of heterozygosity (CN-LOH). As optical genome mapping (OGM) emerges as a potential replacement for traditional cytogenetic methods, the extent to which CMA remains necessary in routine diagnostic workflows remains to be elucidated. METHODS: We retrospectively reviewed 53 primary neoplastic cases, selected from a larger cohort of 327 hematologic malignancy specimens, in which CMA identified one or more CN-LOH events. Event size, ge

Computational QSAR and structure-based identification of plerixafor-derived PIM-1 kinase inhibitors in diffuse large B-Cell lymphoma.

INTRODUCTION: Diffuse large B-cell lymphoma (DLBCL) is the most common and aggressive subtype of non-Hodgkin lymphoma, with a substantial proportion of patients developing resistance to standard chemotherapy. Chromosomal translocations resulting in overexpression of the serine/threonine kinase Proviral Integration site for Moloney murine leukemia virus (PIM-1) contribute to disease progression, therapeutic resistance, and poor clinical outcomes, establishing PIM-1 as a promising molecular target. Unlike many kinases, the ATP-binding site of PIM-1 lacks a backbone hydrogen bond donor within the

Ovarian Precancerous Lesions and Their Molecular Landscape: A Structured Scoping Review Focused on STIC and Related Precursors of Epithelial Ovarian Cancer.

Ovarian cancer remains one of the deadliest gynecologic malignancies because of late-stage diagnosis, biological heterogeneity, and the lack of effective early detection strategies. Epithelial ovarian cancer (EOC) comprises several histotypes, including high-grade serous carcinoma (HGSC), low-grade serous carcinoma (LGSC), endometrioid ovarian carcinoma (ENOC), clear cell ovarian carcinoma (CCOC), and mucinous ovarian carcinoma (MOC), each with distinct molecular pathways and precursor lesions. In recent years, serous tubal intraepithelial carcinoma (STIC) has emerged as the principal precurso

Genome sequencing reveals the role of rare genomic variants in Chinese patients with symptomatic intracranial atherosclerotic disease.

OBJECTIVES: The predisposition of intracranial atherosclerotic disease (ICAD) to East Asians over Caucasians infers a genetic basis which, however, remains largely unknown. Higher prevalence of vascular risk factors (VRFs) in Chinese over Caucasian patients who had a stroke, and shared risk factors of ICAD with other stroke subtypes indicate genes related to VRFs and/or other stroke subtypes may also contribute to ICAD. METHODS: Unrelated symptomatic patients with ICAD were recruited for genome sequencing (GS, 60-fold). Rare and potentially deleterious single-nucleotide variants (SNVs) and sm

Three-dimensional coculture provides an improved in vitro model for papillary renal cell carcinoma.

Papillary renal cell carcinoma (pRCC) represents the second most common kidney cancer and can be distinguished from other types based on its unique histological architecture and specific pattern of genomic alterations. Sporadic type 1 pRCC is almost universally driven by focal or chromosomal amplification of the receptor tyrosine kinase MET, although the specific mode of its activation is unclear. Although the MET receptors found in human tumor specimens appear highly active, those found on the surface of in vitro-cultured tumor cells are only weakly activated in the absence of exogenous hepat
