Dual-Strategy Design of Heterojunction-Enhanced Piezoelectric Hydrogels for Periodontitis Treatment.

Periodontitis is a widespread chronic inflammatory disease that threatens oral and systemic health by sustaining inflammation and accelerating alveolar bone loss. Although bioelectrical modulation can promote bone repair under inflammatory conditions, the coordinated regulation of electrical signaling, immunoregulation, and osteogenesis within an inflamed microenvironment remains a central challenge. Here, we report a multifunctional piezoelectric hydrogel system that combines heterojunction enhancement with dual-salt synergy. In situ construction of ZnO/ZnS heterojunctions with oxygen vacanci

Circuit Survival in Children with Hemolytic Uremic Syndrome Requiring Continuous Renal Replacement Therapy.

INTRODUCTION: Hemolytic uremic syndrome (HUS) is a significant cause of acute kidney injury (AKI) in children that is caused by Shiga toxin-producing Escherichia coli. Continuous renal replacement therapy (CRRT) is frequently used for managing AKI in critically ill pediatric patients. The filter lifespan is a crucial factor affecting its effectiveness and cost. This study aimed to investigate the impact of HUS on the lifespan of CRRT filters compared with that of other etiologies. MATERIALS AND METHODS: This retrospective longitudinal study was conducted in a pediatric intensive care unit in

Molecular Portraits of Aqueous Humor in Primary and Pseudoexfoliative Open-Angle Glaucoma.

Glaucoma is a neurodegenerative disease that affects the optic nerve and retinal ganglion cells. The two most common forms, primary open-angle glaucoma (POAG) and pseudoexfoliative glaucoma (PEG), are associated with ocular hypertension caused by impaired aqueous humor (AH) outflow but have specific ocular and systemic features that may determine their diagnosis and treatment. To identify these features, we compared AH composition in patients using a multiomics approach. Total reflection X-ray fluorescence analysis revealed an increase in zinc and calcium concentrations more pronounced in PEG.

Lipid synthesis seems to drive proliferation in Men1 mouse adrenals and human adrenocortical cell lines.

Adrenocortical carcinoma (ACC) is a devastating disease with few effective treatments. The underlying molecular pathways remain largely unknown. To identify potential pathways and drivers relevant to ACC pathogenesis, we utilized histologically normal adrenals from heterozygous multiple endocrine neoplasia type 1 (Men1) mice to study early adrenocortical tumorigenesis. Employing mass spectrometry-based proteomic profiling, we identified 681 proteins, of which 52 displayed significant differential regulation in the adrenal tissues of heterozygous Men1 mice in comparison with their wild-type cou

The cysteine metabolic regulation through CSE-transsulfuration pathway is essential in iron-induced oxidative damage of heart.

OBJECTIVES: Iron overload (IO) cardiomyopathy is a major cause of mortality in patients with iron overload disorders. This study investigates the role of cystathionine γ-lyase (CSE), a key enzyme in the transsulfuration pathway for cysteine and hydrogen sulfide (H2S) production, in iron-induced oxidative cardiac injury. METHODS: We investigated the effects of CSE on oxidative stress, metabolic dysregulation, and cardiac remodeling using in vivo mouse models of chronic iron overload and in vitro ferric citrate (FAC)-treated cardiomyocytes. RESULTS: Genetic deletion of Cth (encoding CSE) in mi

Iron-Sulfur Cluster Targeting: A Novel Perspective for Treating Alcohol-Related Lipid Metabolic Diseases.

Iron-sulfur (Fe-S) clusters are pivotal molecular cofactors bridging inorganic chemistry and organic life processes, with their tightly regulated biosynthesis and function underpinning cellular metabolic homeostasis. Chronic alcohol-induced metabolic stress impairs Fe-S cluster stability and biosynthesis via reactive oxygen species (ROS)-mediated oxidative damage and direct acetaldehyde toxicity, culminating in functional failure of this core cofactor. Current evidence is synthesized herein to delineate the hierarchical pathogenic cascade triggered by Fe-S cluster dysfunction. At the cellular

Sulfation of Sugarcane-Bagasse Xylan: Process Optimization, Physicochemical Properties and In Vitro Anticoagulant Activity.

Xylan sulfates are established anticoagulant polysaccharides, but their preparation requires balancing sulfate substitution and product recovery. Sugarcane-bagasse xylan was sulfated using sulfur trioxide-pyridine complex (SO3·Py), 4-dimethylaminopyridine (DMAP), and 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride (EDCI). An entropy-weighted five-factor Box-Behnken design jointly optimized degree of substitution (DS) and isolated, DS-adjusted yield, assigning respective weights of 0.570 and 0.430. Pareto analysis identified a trade-off between increasing DS and decreasing yield, w

Regulation of Glycerol-3-Phosphate Dehydrogenase by Hypoxia-Inducible Factor-1 in WSSV-Infected White Shrimp Litopenaeus vannamei.

Hypoxia-inducible factor-1 (HIF-1) induces the Warburg effect in the Litopenaeus vannamei shrimp infected with white spot syndrome virus (WSSV) to satisfy the energetic and biosynthetic demands required for viral replication. Glycerol-3-phosphate dehydrogenase (GPDH) is a key enzyme linking carbohydrate metabolism, lipid biosynthesis, and cellular redox balance; however, its role during WSSV infection remains poorly understood. In this study, the complete coding sequences of the cytoplasmic (LvGPDHc) and mitochondrial (LvGPDHm) GPDH isoforms were characterized from the hepatopancreas of L. van

Programmable PNA-nanoparticle hybrids as nanoscale recognition architectures for amplification-free nucleic acid recognition.

Rapid, field-deployable nucleic acid diagnostics require robust recognition elements and simple signal transduction. We present a programmable peptide nucleic acid-gold nanoparticle (PNA-AuNP) platform for amplification-free detection of Schistosoma spp. DNA. Thiolated PNAs were conjugated to citrate-stabilized AuNPs, forming a nanointerface that translates sequence-specific hybridization into a plasmonic colorimetric signal. Structural and thermodynamic validation confirmed duplex integrity: X-ray photoelectron spectroscopy revealed N 1s (+0.4 eV) and O 1s carbonyl (-0.2 eV) shifts, while FTI

Whole Genome Characterisation of Non-O1/Non-O139 Vibrio cholerae in Wastewater Treatment Plants in the Tshwane District, South Africa.

Non-O1/non-O139 Vibrio cholerae (NOVC) isolates are non-toxigenic and regarded as etiological agents of infrequent but mild-to-severe human gastroenteritis. In response to the 2022-2023 cholera outbreak in Tshwane, this study investigated the occurrence of NOVC isolates in wastewater within the Tshwane district, South Africa. A total of 341 wastewater samples were screened using Thiosulfate citrate bile salts sucrose (TCBS) media, with subsequent confirmation of Vibrio cholerae (V. cholerae) isolates by multiplex-PCR assays. Rep-PCR was performed to determine the genetic relatedness of the iso

Interplay of Oxidative Stress, Autophagy, and Rubicon in Ovarian Follicle Dynamics: Orchestrating Ovarian Aging.

Organ functions generally decline with age, but the ovary is a prototypical organ that undergoes functional loss over time. Autophagy plays a crucial role in maintaining organ homeostasis, and age-related upregulation of the autophagy inhibitor protein, Rubicon, has been linked to cellular and tissue dysfunction. This review describes how granulosa cell autophagy supports follicular growth and oocyte selection and maturation by regulating cellular energy metabolism and protein quality control. We then introduce the role of selective autophagy, including mitophagy or lipophagy, in steroidogenes

Optimization of permeabilized brain tissue preparation to improve the analysis of mitochondrial oxidative capacities in specific subregions of the rat brain.

BACKGROUND: As the major energy producer of cerebral tissue, mitochondria play key roles in brain physiology and physiopathology. Yet, the fine details of the functioning of mitochondrial oxidative phosphorylation in this organ are still scattered with grey area. This is partly due to the heterogeneity of this tissue that challenges our abilities to study specific cerebral subregions. In the last decades, cerebral mitochondria have largely been studied as a single entity by isolating mitochondria from large sections of brain. Given the evidence that these organelles must adapt to brain areas f

Extracellular catalysis of environmental substrates by Shewanella oneidensis MR-1 occurs via active sites on the C-terminal domains of MtrC.

The Gram-negative Shewanellaceae family is well known for its ability to transfer catabolically derived electrons to extracellular terminal electron acceptors through electron conduits that permeate the outer membrane. The primary conduit is MtrCAB, a trimeric porin-cytochrome complex that contains the cell surface exposed decaheme cytochrome MtrC. This donates electrons to extracellular substrates, including OmcA, soluble metals, organic electron shuttles, and insoluble metal oxides. However, it is not clear whether this broad substrate specificity requires specific sites for binding and redu

Metabolomic and Proteomic Profiling of Serum-Derived Extracellular Vesicles from Early-Stage Amyotrophic Lateral Sclerosis Patients.

The identification of reliable biomarkers for amyotrophic lateral sclerosis (ALS) is an unmet medical need for the development of diagnostic and therapeutic strategies. Brain-derived extracellular vesicles (EVs) have been described in peripheral blood serum and used as a direct readout of the status of the central nervous system. Here, we aimed to explore exosome-enriched EVs (referred to simply as EVs) from ALS patients via omics analysis at an early disease stage. Serum EVs were obtained from 9 healthy controls and 9 ALS patients. After EV purification, proteomic (LC‒MS/MS followed by TimsTO

Metabolic Astrocytic Support with Decanoic Acid Enhances Energy Metabolism in Alzheimer's Disease Models.

Alzheimer's disease (AD) is increasingly recognized as a disorder of cerebral energy metabolism, where impaired glucose utilization contributes to disease pathology. Medium-chain fatty acids (MCFAs), such as decanoic acid (C10), have emerged as promising metabolic substrates due to their ability to bypass glycolytic deficits and support mitochondrial function. In this study, we investigated the metabolic impact of C10 in the 5xFAD mouse model of AD and in human induced pluripotent stem cell (hiPSC)-derived astrocytes carrying familial AD mutations. Utilizing stable 13C-labeled metabolic tracer
