A High-Resolution Stereo-Seq Spatial Transcriptomic Resource for Adult Holstein Cattle Liver.

The bovine liver is a highly compartmentalized organ that plays essential roles in continuous gluconeogenesis and nitrogen recycling; however, its spatial molecular architecture has remained largely uncharacterized due to the limitations of traditional bulk and single-cell approaches. To address this gap, Spatial Enhanced Resolution Omics-sequencing (Stereo-seq) was utilized to generate a subcellular-resolution (500 nm) transcriptomic map of an adult Holstein cattle liver, and a refined reference-guided workflow was implemented to overcome standard annotation limitations in livestock. Raw sequ

The Gastro-Circadian Metabolic Axis: A Comprehensive Framework for Chronotherapy in Gastroenterology.

Circadian rhythms exert fundamental control over gastrointestinal and metabolic physiology, governing 24-hour patterns of motility, secretion, nutrient absorption, microbial activity, immune regulation, and hepatic metabolism. Accumulating evidence indicates that the gastrointestinal tract is not an isolated system but is tightly integrated with systemic metabolic and neuroendocrine networks, forming a coordinated gastro-circadian metabolic axis (GCMA). This axis links molecular clocks in the gut, liver, adipose tissue, and skeletal muscle with rhythmic inputs from the gut microbiome, feeding-

Molecular and functional consequences of the Cdc42 C81Y mutation in immune-associated malignancy.

Cell division control protein 42 homolog (Cdc42) is a small Rho GTPase that cycles between active GTP-bound and inactive GDP-bound states to regulate cytoskeletal dynamics and immune signalling. Pathogenic variants of Cdc42 have been linked to rare immunological disorders, including a recently described C81Y mutation associated with primary immunodeficiency (PID) and Hodgkin lymphoma. Although C81Y shares phenotypic similarities with the non-oncogenic C81F variant, it is uniquely associated with thrombocytopenia, chronic inflammation, and malignancy, suggesting distinct molecular consequences

Natural product-based Ebola virus entry inhibitors targeting the viral glycoprotein: A combined computational and experimental study.

Recurrent epidemics of hemorrhagic fever caused by Ebola virus frequently exceed 50% in fatalities making it one of the most dangerous human infections. The absence of effective anti-EBOV medications and their high transmissibility still make it an ongoing health concern worldwide. The Ebola glycoprotein (EBOV-GP) is a class I fusion protein that mediates viral attachment and membrane fusion. Thus, EBOV-GP is a prime target for treatment as it is directly involved in early infection. In this context, the aim of this study is to identify new effective natural-sourced inhibitors of EBOV-GP by ut

HIF-2α drives osteoarthritis progression via suppression of the HDAC4-ATF4-CHOP signaling axis.

Hypoxia-inducible factor-2α (HIF-2α) is a key regulator of cellular adaptation to hypoxia, but its role in osteoarthritis (OA) remains incompletely defined. This study aimed to investigate the contribution of HIF-2α to OA pathogenesis and the underlying molecular mechanisms. Human cartilage specimens were collected to examine HIF-2α expression and components of the histone deacetylase 4 (HDAC4) signaling pathway using Western blotting, quantitative real-time PCR, and immunohistochemistry. An in vitro OA model was established in chondrocytes using interleukin-1β (IL-1β), followed by HIF-2α knoc

Designing of a conserved subunit multiepitope vaccine candidate against Francisella tularensis Schu S4 using immunoinformatics.

Francisella tularensis Schu S4 is the most virulent strain responsible for tularemia, a highly infectious zoonotic disease. Despite its potential as a pathogen of concern, no effective vaccine has been developed for this pathogen. In this study, two multi-epitope subunit vaccine candidates were designed targeting the virulence factors DacD and DsbA to reduce disease severity caused by F. tularensis Schu S4. Using a comprehensive immunoinformatics approach, the most efficient B- and T-cell epitopes were identified from conserved sequences and linked with adjuvants and linkers to generate two va

Conformations and sequence determinants in the lipid binding of an adhesive peptide derived from Vibrio cholerae biofilms.

Surface adhesion is critical to the survival of pathogenic bacteria both in natural niches and during infections, often via forming matrix-embedded communities called biofilms. Vibrio cholerae, the causal agent of pandemic cholera, is capable of forming biofilms adhering to both biotic and abiotic surfaces and the biofilm lifestyle has been implicated in promoting the survival of V. cholerae both in the natural reservoir and during host colonization. Previously, a 57-amino acid loop in the biofilm-specific adhesin Bap1 (Bap1-57aa) has been identified as a key contributor to the adhesion of V.
