Association between XPF polymorphisms and cancer risk: a meta-analysis.

BACKGROUND: Xeroderma pigmentosum complementation group F (XPF or ERCC4) plays a key role in DNA repair that protects against genetic instability and carcinogenesis. A series of epidemiological studies have examined associations between XPF polymorphisms and cancer risk, but the findings remain inconclusive. METHODOLOGY/PRINCIPAL FINDINGS: In this meta-analysis of 47,639 cancer cases and 51,915 controls, by searching three electronic databases (i.e., MEDLINE, EMBASE and CNKI), we summarized 43 case-control studies from 29 publications on four commonly studied polymorphisms of XPF (i.e., rs180

Irritable bowel syndrome.

INTRODUCTION: The prevalence of irritable bowel syndrome (IBS) varies depending on the criteria used to diagnose it, but it ranges from about 5% to 20%. IBS is associated with abnormal gastrointestinal motor function and enhanced visceral perception, as well as psychosocial and genetic factors. People with IBS often have other bodily and psychiatric symptoms, and have an increased likelihood of having unnecessary surgery compared with people without IBS. METHODS AND OUTCOMES: We conducted a systematic review and aimed to answer the following clinical question: What are the effects of treatmen

Developments in the discovery of drugs for spinal muscular atrophy: successful beginnings and future prospects.

INTRODUCTION: Spinal muscular atrophy (SMA) is an autosomal recessive disease caused by mutations in a gene that produces a protein called survival motor neuron (SMN). SMN has an important role in snRNP assembly in all cells but that may not be its only role; the reasons for SMN deficiency resulting in neuromuscular dysfunction and motor neuron degeneration remain active areas of research. Besides increasing SMN, compensating for SMN deficiencies or neuroprotection may be therapeutic options for SMA. Age of onset and the rate of disease progression are variable and therapeutic strategies shoul

The role of proteomic research in vascular disease.

INTRODUCTION: Arterial diseases including atherosclerosis, intimal hyperplasia and aneurysms have been shown to be a product of genotype and environment. Gene expression pathways rely on protein translation to generate target effects. As a result of alternative splicing and post-translational modifications, one gene does not code for a single protein but for many. Proteomic studies allow quantification of these proteins in a biological system and determination of altered protein expression in disease. Proteomics is a powerful and expanding field of investigation which in combination with other

Risk of pancreatic cancer in workers exposed to chlorinated hydrocarbon solvents and related compounds: a meta-analysis.

This is a meta-analysis of occupational exposures to chlorinated hydrocarbon (CHC) solvents and pancreatic cancer, based primarily on studies that addressed exposure directly (agent studies) and secondarily on studies that reported data without verification of individual CHC exposures (job title studies), all of which were listed in databases for the period January 1969 to May 1998. Standardized extraction of data and double-checking of consistency of data extraction by five extractors were done. Simple random models estimated meta-relative risks. Suggestive weak excesses were found for trichl

Homogeneity of 16S-23S ribosomal intergenic spacer regions of Tropheryma whippelii in Swiss patients with Whipple's disease.

The current genetic strategies used to identify Tropheryma whippelii, the putative agent of Whipple's disease, are based on PCR-mediated amplification of a part of its 16S rRNA gene (16S rDNA). Because there is very little intraspecies variation in these molecules, they are not suitable as targets for epidemiologic investigations. However, the intergenic spacer region between the 16S and 23S rDNAs is usually much more variable and has repeatedly been used for epidemiologic purposes. We have therefore amplified the spacer region of T. whippelii directly from clinical specimens from nine indepen

High incidence of colorectal cancer in Japanese individuals with hypertension.

The association between hypertension and the incidence of colorectal cancer remains controversial. This study aimed to clarify this relationship in a Japanese population using two large-scale cohort datasets. We conducted a population-based retrospective cohort study using the Shizuoka Kokuho Database, which contains insurance claims from the Shizuoka region and medical checkup data collected between 2012 and 2022. Propensity score matching was used to compare colorectal cancer incidence between normotensive and hypertensive groups, and cumulative incidence was analyzed using Gray's test, with

Neonatal-onset familial hemophagocytic lymphohistiocytosis type 2: a case report.

BACKGROUND: Familial hemophagocytic lymphohistiocytosis type 2 (FHL2) is a rare autosomal recessive disorder primarily caused by mutations in the perforin-1 (PRF1) gene, leading to immune system hyperactivation and multi-organ dysfunction. FHL2 typically presents in infancy, with neonatal onset being extremely rare and particularly difficult to recognize early due to substantial clinical overlap with sepsis. Given its rapid progression, early detection is critical for effective diagnosis and management. CASE SUMMARY: This case report describes a neonate with FHL2 who presented on day 8 after

Genome-Wide Association Study of Vertical Jump Performance Among Elite Badminton Players.

Vertical jump performance is known to be a moderately heritable trait. However, previous studies on sport genetics have largely relied on candidate-gene approaches, which do not adequately reflect the polygenic nature of explosive performance, particularly among elite badminton players. Therefore, the aim of the present study was to identify genetic variants associated with lower-limb explosive performance, assessed via vertical jump measures, among elite Turkish badminton players using a genome-wide association study (GWAS) approach. The present study included 90 elite male (n = 47) and femal

Genetic Diversity and Evolutionary Dynamics of Feline Panleukopenia Virus in China: Phylogenetic Analysis and Substitution Patterns in NS1 and VP2 Proteins.

Feline panleukopenia virus (FPLV) is the primary causative agent of a highly contagious and often fatal disease affecting domestic cats and other felids. The increasing isolation of species-specific FPLV variants from multiple host species has garnered considerable attention, highlighting the need to investigate their genetic diversity. In this study, three FPLV isolates were obtained and phylogenetically classified into two distinct FPLV-China groups within separate clusters. Compared to the prototype FPLV (M38246.1), these isolates exhibited seven amino acid substitutions in the NS1 (n = 6)

Biomarker Research in NSAID Hypersensitivity: A Scoping Review and Evidence Map.

Background/Objectives: Nonsteroidal anti-inflammatory drug (NSAID) hypersensitivity comprises clinically distinct phenotypes, with different implications for diagnosis and future drug use. However, the biomarker literature in this field remains heterogeneous and difficult to interpret as a whole. We aimed to map how NSAID hypersensitivity biomarker studies are distributed across harmonized phenotype categories, biomarker classes, intended uses, and translational validation stages. Methods: We conducted a scoping review with systematic literature searching and descriptive evidence mapping. Engl

First-in-human nuclease-free homologous recombination-dependent gene editing in pediatric patients with methylmalonic acidemia: results of a phase 1/2 study.

Gene-based editing can potentially correct the genetic defect in methylmalonic acidemia (MMA). SUNRISE, a first-in-human phase 1/2 open-label study, evaluated the safety/tolerability (primary endpoints) of liver-targeted hLB-001 in four pediatric participants (ages 20-114 months) with mitochondrial methylmalonyl-CoA mutase (MMUT)-deficient MMA. We designed a single-infusion adeno-associated viral capsid (hLB-001) to nondisruptively integrate functional MMUT at the 3' end of the albumin (ALB) locus to produce both albumin and MMUT. All four participants experienced at least one treatment-emerge
