Novel oral delivery system using modified inulin for enhanced liver cancer therapy.

Hepatocellular carcinoma (HCC) remains a leading cause of cancer-related deaths worldwide. Although advanced oral anticancer drugs, such as sorafenib and lenvatinib, are available for treating HCC, their effectiveness is limited by significant side effects and the requirement for optimal liver function, restricting their broader use. Furthermore, these drugs cannot be used in combination, leading to suboptimal treatment outcomes. This study introduces a novel therapeutic approach by repurposing two existing drugs, niclosamide and disulfiram, as a combination therapy to treat HCC. Despite their

Pharmaceutically engineered olfactory-masking agents: Innovating oral drug delivery through multisensory excipient design.

Enhancing patient adherence to oral medication administration is still exceedingly difficult, especially for sensitive, elderly, and pediatric groups. Techniques for disguising taste are widely known, but olfactory modulation, which is a crucial factor in determining palatability and adherence, has received little attention. This review integrates concepts from formulation science, medicinal chemistry, and sensory neuroscience to examine the new subject of scent-masking through creative excipient techniques. We explore the physiological processes implicated in scent-triggered aversion and offe

Non-intravitreal anti-vascular endothelial growth factor delivery for ocular neovascular diseases: Current status and future directions.

Intravitreal anti-vascular endothelial growth factor (VEGF) therapy has fundamentally transformed the management of retinal and choroidal neovascular diseases. Despite its proven efficacy, long-term reliance on repeated intravitreal injections imposes a substantial burden on patients and healthcare systems. Real-world studies consistently demonstrate reduced adherence and inferior visual outcomes compared with randomized clinical trials. These limitations have driven interest in alternative, non-intravitreal routes of anti-VEGF delivery aimed at reducing treatment burden while maintaining ther

Tumor Assembloids as Three-Dimensional Platforms for Modeling Drug Delivery Barriers: Construction Strategies, Applications, and Translational Challenges.

A major limitation of conventional two and three-dimensional preclinical in vitro cancer models is their inability to reproduce the drug-delivery barriers. Tumor assembloids, which integrate patient-derived cancer cells with stromal, endothelial, and immune components in three-dimensional architectures, provide a manipulable framework for simulating these multicellular impediments. This review specifically examines the application of tumor assembloids to investigate drug delivery constraints, including stromal exclusion, vascular transport, immune-mediated resistance, penetration gradients, an

Chrono-Nanomedicine at the Barrier: Circadian Rhythms Influence the Delivery of Donepezil Across the BCSFB.

To reach the target tissue within the central nervous system (CNS), drugs, such as donepezil (DNPZ), must overcome naturally occurring barriers. Despite its substantial pharmacological relevance, the blood-cerebrospinal fluid barrier (BCSFB) remains insufficiently studied. The BCSFB harbors a functional molecular clock that regulates, for instance, the expression of membrane transporters. Thus, there has been an increasing interest in exploring chronotherapeutic strategies to enhance cerebral drug delivery. Additionally, nanotechnology constitutes a well-established approach for improving tiss

Dual drug-loaded nanofibrous scaffolds for targeting dormant breast cancer cells: design, mechanisms, and translational perspectives.

Breast cancer is the most commonly diagnosed malignancy among women worldwide. Recurrence associated with maintenance therapy may promote the development of multidrug resistance, while dormant breast cancer cells (DBCCs) within the tumor microenvironment (TME) are inherently resistant to conventional therapies. These factors collectively contribute to late-stage relapse and metastasis. Nanofibers (NFs) have recently emerged as valuable platforms for formulating biomedical and healthcare applications, including tissue engineering, wound healing, and post-surgical cancer management. In particula

Streptavidin functionalized hyaluronic acid hydrogels for controlled and customizable drug delivery.

Hyaluronic acid (HA)-based hydrogels have emerged as promising materials for tissue repair given their ability to rapidly gel in situ to precisely fill tissue defects, their mechanical tunability and customizable degradation kinetics, and their capacity to deliver therapeutic agents to the site of injury. HA hydrogels can also be used to encapsulate therapeutics for local delivery; however, release through diffusion may be too rapid to achieve a therapeutic window for many applications. To overcome this limitation, we functionalized norbornene-modified HA (NorHA) hydrogels with streptavidin us

Intestinal microenvironment-modulating nanomedicines for inflammatory bowel disease.

Inflammatory bowel diseases present a highly restrictive intestinal microenvironment for drug delivery, in which epithelial injury, chronic inflammation, and microbial dysbiosis collectively impair transport, retention, and therapeutic efficacy. Excessive reactive oxygen species (ROS), mucus barrier disruption, altered luminal acidity, hydrogen sulfide accumulation, and abnormal enzymatic activity destabilize therapeutic cargos and limit access to inflamed tissues. These pathological features have motivated the development of nanocarriers designed either to directly modulate biochemical abnorm
