The case is a woman in her 60s with a heterozygous germline variant in PALB2 (c.3106G>C, p.Val1036Leu) who developed ovarian cancer. Tumour somatic analysis revealed a 24.7% loss of heterozygosity, commonly associated with homologous recombination deficiency. No other pathogenic DNA damage repair gene mutations were identified, although ATR amplification was observed. The proband has four siblings, all of whom tested positive for the PALB2 c.3106G>C variant. Both of her brothers were diagnosed with prostate cancer. One of her sisters was diagnosed with breast cancer at age 62, while the other

Autosomal recessive congenital ichthyosis (ARCI) is a rare and heterogeneous group of congenital disorders characterized by aberrant skin cornification and diffuse skin scaling. Most neonates with ARCI are collodion babies. Herein, we recruited a Chinese family with one patient, who died as a collodion baby one month after birth. Heterozygous mutations of c.424 C > T and c.1385 C > T in the TGM1 were detected in the proband by Chip capture high-throughput sequencing, and the two alleles were inherited from the mother and father, respectively. While the mother was pregnant with another child, h

Paired box 2 (PAX2)-related disorder, also known as renal coloboma syndrome, is a variably penetrant autosomal dominant condition, associated with renal and ophthalmological abnormalities. We report a child with PAX2-related disorder who presented atypically with acute ataxia on a background of stage 3 chronic kidney disease. Extensive biochemical, radiological and gene agnostic rapid trio exome sequencing was non-diagnostic. Identification of bilateral optic disc colobomas in the proband and his father raised the suspicion of an inherited PAX2-related disorder. No causative variants were iden

This article builds on years of ethnographic conversations I sustained with my father, 89, who lives in Colombia. Soon after getting diagnosed with an incurable Multiple Myeloma-a cancer known for unleashing prolonged and painful agonies-he withdrew from oncology treatments and secured access to euthanasia (assisted-dying) on his own, bypassing medico-insurance guidelines created to regulate this medical practice and prevent abuses. Eight years after withdrawing treatments, my dad is still alive. His case shines a light on how securing access to euthanasia may have had unintended therapeutic e

Axenfeld-Rieger syndrome is a rare autosomal dominant disorder characterized primarily by developmental anomalies of the anterior segment of the eye and systemic manifestations, including craniofacial abnormalities, dental anomalies, and neurological involvement. Although genetic mutations in the forkhead box C1 (FOXC1) or paired-like homeodomain transcription factor 2 (PITX2) have been implicated, the full extent of associated neurological features remains underexplored. We present the case of a 2-year-old boy diagnosed with Axenfeld-Rieger syndrome who exhibited mild facial dysmorphism and r

INTRODUCTION: Pathogenic variants in the KMT5B gene, encoding a lysine methyltransferase involved in chromatin remodeling, have been associated with intellectual disability, autism spectrum disorder, and various craniofacial features. However, the detailed genotype-phenotype correlations have yet to be fully elucidated.
CASE PRESENTATION: We report a four-year-old male patient who presented with developmental delay, impaired social interaction, repetitive behaviors, and language delay. Whole-exome sequencing identified a novel heterozygous frameshift variant in KMT5B (c.618del; p.Glu206Aspfs*

In the paper I analyse Danylo Samoylovich's views on the prevention, course and treatment of the plague. In addition to the traditional method of source analysis, I also use other theoretical tools: anthropology of knowledge, theory of modernisation as well as institutionalisation, medicalisation and professionalisation. Both Russian and Ukrainian authors call Samoylovich the father of national epidemiology. There is no doubt that his accomplishments have been exaggerated in Russia and Ukraine, but in no way does this detract from Samoylovich's significance, both in his contributions to theore

Miliary tuberculosis (TB) is a rare but severe manifestation in infancy. Hypercalcaemia, well documented in granulomatous diseases like TB and sarcoidosis in adults, is infrequently reported in children. Nephrogenic diabetes insipidus induced by hypercalcaemia is a known complication. A female infant with right-sided neck swelling for 1 month (fine-needle aspiration showed tubercular lymphadenitis), on four-drug anti-tubercular therapy (ATT), admitted with complaints of polyuria, polydipsia and progressive weight loss over the last 2 weeks. Her father was on ATT for pulmonary TB. Investigation

Pierre Fauchard, considered the father of dentistry, contributed to the development of different fields of dentistry that we know today. However, the contribution of this important individual to the fields of oral medicine and oral and maxillofacial pathology is unknown. This study aimed to identify Pierre Fauchard's contribution to these areas of dentistry. We focused on "Le Chirurgien Dentiste, or Traité des Dents" in both French and English, looking for information about the oral diseases diagnosed and treated by Fauchard. Information on patient history, disease description, treatment appli

During World War II, millions of people were mistreated and imprisoned in Nazi concentration camps. Due to the antisemitic pressure applied by the Nazi regime, many scientists had to leave Germany, and they immigrated to the United States, Switzerland, Turkey or South America. Alfred Kantorowicz was among those highly educated people who were forced out of their professional career. For a certain period, he had to stay away from the world of research and academia, which were his favorite occupations. However, these unexpected difficulties did not prevent him to pursue his success story with ma

Little is known about clinical documentation for youth exposed to parental justiceinvolvement (e.g., parole, probation, jail, prison). We reviewed the electronic health records of 100 youth with at least one mention of parental incarceration between 2011-2019 from a large Midwestern pediatric hospital-based institution to describe clinical documentation and health characteristics. Within the sample, youth more commonly experienced incarceration of a father-identified figure (68%) as opposed to a mother-identified figure (32%). Seventeen percent (17%) of the youth were between zero and four yea

Autosomal recessive spinocerebellar ataxias (SCARs) are one of the most common neurodegenerative diseases characterized by progressive ataxia. Although SCARs are known to be caused by mutations in multiple genes, there are still many cases that go undiagnosed or are misdiagnosed. In this study, we presented a SCAR patient, and identified a probable novel pathogenic mutation (c.1A>G, p.M1V) in the AFG3L2 start codon. The proband's genotype included heterozygous mutations of the compound AFG3L2 (p.[M1V]; [R632X] (c.[1A>G]; [1894.C>T])), which were inherited from the father (c.1A>G, p.M1V) and mo

Capillary malformation-arteriovenous malformation (CM-AVM) syndrome is characterized by the presence of multiple small (1-2 cm in diameter) capillary malformations of the skin. This disorder has been described as two distinct entities: CM-AVM1 and CM-AVM2. The diagnosis of these disorders has been associated with pathogenic variants in the RASA1 gene for RASA1-CM-AVM, formerly known as CM-AVM1, and, more recently, the EPHB4 genes for EPHB4-CM-AVM, formerly known as CM-AVM2. Affected patients with either type may also have arteriovenous malformations and fistulas, which can cause life-threateni

An infant presents in extremis. After the medical team stabilizes him, the race is on to figure out why he got so sick in the first place. The consulting genetics team thinks that it is unlikely his problems are due to a genetic cause, but his extreme, confounding presentation is enough to justify trio exome sequencing. When the results reveal an unexpected, paternally inherited variant of uncertain significance (VUS) in NOTCH3, fresh questions arise. The infant's presenting symptoms and descriptive diagnoses, including hematemesis, epistaxis, and gastric ulcers, certainly do not fit the mold

BACKGROUND: Mutations in PDE6A and PDE6B are known to cause autosomal recessive RP in humans, On the other hand, mutations in PDE6G are rare but can lead to severe early-onset RP.
CASE PRESENTATION: An 8-year-old Chinese boy was referred to our hospital for poor vision issues. Refraction with cycloplegia showed high hyperopia with astigmatism both eyes. Funduscopic examination revealed typical bone spicule-type pigment deposits in the periphery and midperiphery. The patient was given glasses and a whole exome sequencing containing mitochondrial genes was performed. The results of genetic test

BACKGROUND: This study aims to report the clinical characteristics of a child with autosomal recessive polycystic kidney disease (ARPKD) within a Chinese Zhuang ethnic family.
METHODS: We used whole exome sequencing (WES) in the family to examine the genetic cause of the disease. Candidate pathogenic variants were validated by Sanger sequencing.
RESULTS: We identified previously unreported mutations in the PKHD1 gene of the proband with ARPKD through WES: a splice site mutation c.6809-2A > T, a nonsense mutation c.4192C > T(p.Gln1398Ter), and a missense mutation c.2181T > G(p.Asn727Lys). Her

Recessively inherited limb-girdle muscular dystrophy type 1, caused by mutations in the calpain 3 gene, is the most common limb-girdle muscular dystrophy worldwide. Recently, cases of autosomal dominant calpainopathy have been described. A man was referred to our neurological outpatient clinic at the age of 54 for persistent hyperCKemia (>1000 U/l) associated with muscle fatigue and myalgia. Clinical examination revealed mild proximal weakness in the lower limbs. His brother exhibited a moderate increase in serum creatine kinase levels (up to 2000 U/l) without other signs of myopathy. Their fa

Fain's theoretical conceptualisation of the 'Censorship of the Lover' (1971) offers a framework to investigate the unrepresented traumatic aspects of the hypochondriacal fear of breast cancer. The failure of the maternal role to function as both the mother-of-the-infant and the lover-of-the-father creates considerable deficits in the primal psychosomatic tie. The authors aim to draw attention on to the importance of the mother-of-the-infant facet of the dual maternal function. The threatening repetitive scenario experienced by the hypochondriacal patient is considered as a form of pathological

Mitochondrial DNA mutations such as A3243G or A1555G are widely reported to cause hearing loss, but few reports exist on the A8296G mutation, which can also cause hearing loss. This report presents the case of a patient with the A8296G mutation and severe bilateral sensorineural hearing loss (SNHL) that progressed over two decades. The patient had no history of diabetes, but did have a family history of SNHL in her father and maternal grandmother. She was first diagnosed with SNHL at 45 years of age, and an A8296G mutation was found. The hearing threshold in the low-frequency range of the righ
