JAK-1 inhibitors in the management of atopic dermatitis in patients over 65 years old: a descriptive cohort study.

Atopic dermatitis (AD) can affect older adults. Janus kinase inhibitors (JAK-1 inhibitors), such as upadacitinib and abrocitinib, have emerged as effective treatments for moderate-to-severe AD. However, data on their safety and efficacy in older adults remain limited due to underrepresentation in clinical trials. This retrospective cross-sectional study examined the efficacy and safety of oral JAK-1 inhibitors in patients aged 65 years or older. Data was extracted from electronic medical records. Disease control was assessed using the Atopic Dermatitis Control Tool (ADCT), while adverse events

61Cu-NODAGA Prostate-specific Membrane Antigen Imaging and Therapy for Prostate Cancer: Phase 1 Trial of a New Class of 61Cu-labeled PET Radiotracers.

Background Prostate-specific membrane antigen (PSMA)-targeted PET has revolutionized prostate cancer imaging, but the sensitivity at low prostate-specific antigen levels is lacking. Copper 61 (61Cu) is a positron emitter with favorable physical characteristics that allow for delayed imaging, which may result in improved sensitivity. Purpose To evaluate the safety, dosimetry, optimal imaging parameters, and initial efficacy of a novel 61Cu-labeled PSMA-targeting radiotracer, 61Cu-1,4,7-triazacyclononane,1-glutaric acid-4,7-acetic acid (NODAGA)-PSMA for imaging and therapy (I&T) (hereafter, 61Cu

Infectious complications in CLL/SLL patients receiving Bruton's Tyrosine Kinase inhibitors - systematic review and meta-analysis of randomized controlled trials.

The development of Bruton Tyrosine Kinase inhibitors (BTKis) has revolutionized the management of chronic lymphocytic leukemia/small lymphocytic lymphoma (CLL/SLL). However, increased infection rates have been reported in patients receiving BTKis in multiple clinical trials. This study aimed to evaluate the risk of infections associated with BTKis compared to other therapeutic regimens in CLL/SLL patients. A systematic review and meta-analysis of randomized controlled trials (RCTs) was conducted. We included trials comparing BTKi-containing regimens (e.g., BTKi alone or combined with anti-CD20

Structure-Based Drug Design of Novel Heterocyclic Scaffolds as TgCDPK1 Inhibitors.

Toxoplasmosis is caused by the protozoan parasite Toxoplasma gondii and poses grave health concern for immunocompromised patients. T. gondii has a family of calcium dependent protein kinases (CDPKs) that control a variety of critical processes. Among these, TgCDPK1 is required for parasite motility, cell invasion, and egress and hence is essential both for in vitro growth of T. gondii and to cause infections in animals. Using existing X-ray cocrystal structures of pyrazolopyrimidine (PP) inhibitors bound to TgCDPK1, six new chemical series of inhibitors are rationally designed. The synthesis o

Efficacy of Azacitidine Plus Venetoclax in Acute Myeloid Leukemia Transformed from Myelodysplastic Syndrome after Failure of Azacitidine Monotherapy.

Objective Patients with acute myeloid leukemia (AML) transformed from myelodysplastic syndrome (MDS) have a poor prognosis, including those treated with azacitidine during the MDS phase; there is no standard for the care of these patients. Recently, azacitidine plus venetoclax (AZA/VEN) was reported to prolong the survival in treatment-naïve AML patients compared with AZA monotherapy. However, the results of AZA/VEN for AML transformed from MDS, particularly after AZA monotherapy, remain unclear. The present study therefore compared the clinical results of AZA/VEN treatment in these patients.

Genotyping and treatment issues with 'unusual' HCV 1 subtypes.

Unusual non 1a/1b HCV 1 subtypes are mostly prevalent in Sub-Saharan Africa. These variants harbour resistance associated substitutions as natural polymorphisms in the NS5A region and are difficult to characterize and treat. We report the HCV genome sequences from two African subjects with unusual subtype 1 g and 1e treated with the pangenotypic regimen sofosbuvir/velpatasvir with failure in the latter. Sequence analysis of different regions of the HCV genome in a subtype 1 reference panel and use of multiple subtyping tools showed that the NS5A region is the most suitable to detect these unus
