DsiRNA-mediated silencing of Ceratitis capitata transformer or transformer-2 leads to masculinization of XX embryos and systemic gene silencing in ovaries.

Ceratitis capitata (medfly), a major agricultural pest, is predominantly controlled using chemical insecticides, which pose environmental risks. Ecosustainable alternatives, such as the Sterile Insect Technique (SIT), rely on the mass release of sterile male-only progeny. However, sexing of male offspring requires the elimination of females during development. To overcome the loss of 50% of the progeny, the sex reversal of females into XX fertile males at embryonic stages by dsRNA injections was effective but not scalable. This study demonstrates the efficacy of Dicer-substrate small interferi…

Synthesis and Structure-Activity Relationship of Phosphonate Esters with (S)‑Cinerolone and (S)‑Jasmololone as Irreversible Inhibitors of the GDSL Esterase/Lipase TcGLIP to Study Pyrethrin Biosynthesis.

TcGLIP, a GDSL esterase/lipase family enzyme found widely across species, synthesizes natural insecticides pyrethrins with (1R,3R)-chrysanthemoyl CoA and (S)-rethrolones as substrates in the Dalmatian Daisy Tanacetum cinerariifolium. We previously reported that alkyl- or benzyl-substituted phosphonate esters with (S)-pyrethrolone blocked TcGLIP at nanomolar ranges, but the mechanism of enzyme-substrate interactions remains elusive. Notably, it is unclear whether the conjugated diene of (S)-pyrethrolonyl moiety is essential for TcGLIP inhibition and whether such probes are effective in blocking…

Occurrence of current-use pesticides in sediment cores from lakes and peatlands in pristine mountain areas of Brazilian national parks.

This study assessed the occurrence of current-use pesticides in sediment cores from six lakes and peatlands in high-altitude (1952-2374 m) pristine areas within two southeastern Brazilian National Parks: Itatiaia National Park (IT-NP) and Serra dos Órgãos National Park (SO-NP). We sampled three sediment cores from lakes at IT-NP and three from peatlands at SO-NP, totaling 60 subsamples. Among the 38 current-use pesticides assessed, 17 were found in at least one sample, with 14 - including herbicides, insecticides, fungicides, and acaricides - identified in both parks. The most frequently detec…

Pesticides detected in two urban areas have implications for local butterfly conservation.

Human-managed green spaces in urban landscapes have become important focal points for insect conservation, partly because of the desirable i … ited States), asking if larval host plants for butterflies in the two regions contained herbicides, insecticides, and fungicides. We assayed 336 individual plants in 19 genera, including woody and herbaceous plants. Pesticide presence was ubiquitous: only 22 samples had no detectable levels of pesticides; the median number of compounds detected in the other 31 …

High prevalence of knockdown resistance mutations, genetic clade diversity, and detection of Acinetobacter species in head lice (Pediculus humanus capitis) infesting children in a Thai orphanage: A comprehensive survey.

Pediculosis, caused by the head louse (Pediculus humanus capitis), remains a public health challenge among school-aged children. The emergence of resistance to pyrethroid insecticides associated with mutations in the knockdown resistance (kdr) genes may hinder control efforts. Furthermore, head lice have been implicated as potential vectors for bacterial pathogens, raising concerns about their role in disease transmission. Head lice collected from children in an orphanage were analyzed for genetic diversity, kdr-mutation resistance, and the presence of bacterial pathogens. A total of 107 lice …

Monarch Butterflies in Western North America: A Holistic Review of Population Trends, Ecology, Stressors, Resilience and Adaptation.

Monarch butterfly populations in western North America suffered a substantial decline, from millions of butterflies overwintering in California in the 1980s to less than 400,000 at the beginning of the 21st century. The introduction of neonicotinoid insecticides in the mid-1990s and their subsequent widespread use appears to be the most likely major factor behind this sudden decline. Habitat loss and unfavorable climates (high temperatures, aridity, and winter storms) have also played important and ongoing roles. These factors kept overwintering populations stable but below 300,000 during 2001…

Simultaneous determination of twelve neonicotinoids and six metabolites in human urine with isotope-dilution UPLC-Q-Orbitrap HRMS.

The extensive global use of neonicotinoid insecticides (NNIs) has led to widespread human exposure, necessitating the development of effective methods for large-scale biomonitoring. However, current methods are inadequate in simultaneously and accurately detecting various NNIs or their metabolites (m-NNIs). In this study, we aimed to establish a robust method using solid-phase extraction (SPE)-ultra high performance liquid chromatography tandem Q-Orbitrap high resolution mass spectrometry (UPLC-Q-Orbitrap HRMS) for the simultaneous determination of 12 NNIs and 6 m-NNIs in human urine. Samples …

Situational analysis of pesticide poisoning and perceptions of autoinjector devices in rural communities in Sri Lanka - a study protocol.

Intentional and unintentional pesticide poisoning is an important public health problem, especially in low- and middle-income countries. Individuals who have been exposed to toxic pesticides, particularly organophosphorus insecticides, need early treatment. Atropine autoinjector devices offer a potential solution, allowing storage of effective treatment near agricultural workers' fields and homes that could be reached within minutes by the worker or fellow villagers to provide first-line emergency care. Here we present the design of a qualitative, formative study that will constitute the first…

Concentration-dependent effects of spinetoram on nontarget freshwater microalgae: A comparative study on Chlorella vulgaris and Microcystis aeruginosa.

The rising global demand for agricultural products is leading to the widespread application of pesticides, such as spinetoram, resulting in environmental pollution and ecotoxicity to nontarget organisms in aquatic ecosystems. This research focused on assessing the toxicity of spinetoram at various concentrations (0, 0.01, 0.1, 0.5, 1.0, and 3.0 mg L-1) on two common freshwater microalgae, Chlorella vulgaris and Microcystis aeruginosa, to shed light on the ecotoxicological effects of insecticides. Our findings demonstrate that M. aeruginosa is more sensitive to spinetoram than is C. vulgaris, w…

Assessing the effects of a commercial fungicide and an herbicide, alone and in combination, on Apis mellifera: Insights from biomarkers and cognitive analysis.

Agrochemicals play a vital role in protecting crops and enhancing agricultural production by reducing threats from pests, pathogens and weeds. The toxicological status of honey bees can be influenced by a number of factors, including pesticides. While extensive research has focused on the lethal and sublethal effects of insecticides on individual bees and colonies, it is important to recognise that fungicides and herbicides can also affect bees' health. Unfortunately, in the field, honey bees are exposed to mixtures of compounds rather than single substances. This study aimed to evaluate the e…

Was the massive increase in use of teratogenic agrichemicals in western states (USA) associated with declines in wild ruminant populations between 1994 and 2013?

Population declines were documented in multiple ruminant species in Montana and surrounding states starting in 1995. While weather, food sources, and predation certainly contributed, the declines were often attributed, at least partly, to unexplained factors. Use of teratogenic agrichemicals, notably neonicotinoid insecticides, fungicides, and glyphosate-based herbicides, massively increased regionally in 1994-96. The question explored in this review is whether this vastly increased use of these teratogenic pesticides might have contributed to observed population declines. We provide reference…

Sublethal pesticide exposure in non-target terrestrial ecosystems: From known effects on individuals to potential consequences on trophic interactions and network functioning.

Over the last decades, the intensification of agriculture has resulted in an increasing use of pesticides, which has led to widespread contamination of non-target ecosystems in agricultural landscapes. Plants and arthropods inhabiting these systems are therefore chronically exposed to, at least, low levels of pesticides through direct pesticide drift, but also through the contamination of their nutrient sources (e.g. soil water or host/prey tissues). Pesticides (herbicides, acaricides/insecticides and fungicides) are chemical substances used to control pests, such as weeds, phytophagous arthro…

Identification and Spatiotemporal Expression of a Putative New GABA Receptor Subunit in the Human Body Louse Pediculus humanus humanus.

The human louse (Pediculus humanus) is an obligatory blood feeding ectoparasite with two ecotypes: the human body louse (Pediculus humanus humanus), a competent vector of several bacterial pathogens, and the human head louse (Pediculus humanus capitis), responsible for pediculosis and affecting millions of people around the globe. GABA (γ-aminobutyric acid) receptors, members of the cys-loop ligand gated ion channel superfamily, are among the main pharmacological targets for insecticides. In insects, there are four subunits of GABA receptors: resistant-to-dieldrin (RDL), glycin-like receptor o…

Evaluation of Yellow Fever Virus Infection in Aedes aegypti Mosquitoes from Pakistan with Distinct Knockdown Resistance Genotypes.

BACKGROUND: Yellow fever (YF) is an acute hemorrhagic disease endemic to Africa and Latin America; however, no cases have been reported in Asian regions with high Aedes aegypti infestation. Factors such as environmental conditions and genetic variations in the yellow fever virus (YFV) strains and mosquito populations may explain this absence. Mosquito populations have undergone strong selective pressure owing to the excessive use of insecticides. This pressure has led to the spread of alterations, such as knockdown-resistant mutations (kdr), which, while conferring resistance to pyrethroids, a…
