Disorganized muscle protein-1 (DIM-1) of filarial parasite Brugia malayi: cDNA cloning, expression, purification, structural modeling and its potential as vaccine candidate for human filarial infection.

We have recently identified disorganized muscle protein-1 (DIM-1) in one of the proinflammatory fractions of the human filaria Brugia malayi adult worm. The present study was undertaken to characterize B. malayi DIM-1 (DIM-1bm) and explore its vaccine potential. In this study we cloned and expressed the DIM-1bm gene, investigated its sequence homology with other nematodes, constructed in silico structural model, purified the recombinant DIM-1bm (rDIM-1bm) protein, and studied the effect of immunization with rDIM-1bm on the establishment of B. malayi infection in Mastomys coucha. DIM-1bm showed

BtcA, A class IA type III chaperone, interacts with the BteA N-terminal domain through a globular/non-globular mechanism.

Bordetella pertussis, the etiological agent of "whooping cough" disease, utilizes the type III secretion system (T3SS) to deliver a 69 kDa cytotoxic effector protein, BteA, directly into the host cells. As with other T3SS effectors, prior to its secretion BteA binds BtcA, a 13.9 kDa protein predicted to act as a T3SS class IA chaperone. While this interaction had been characterized for such effector-chaperone pairs in other pathogens, it has yet to be fully investigated in Bordetella. Here we provide the first biochemical proof that BtcA is indeed a class IA chaperone, responsible for the bind

Oxidation of annelated diarylamines: analysis of reaction pathways to nitroxide diradical and spirocyclic products.

Oxidation of diaryldiamine 2, a tetrahydrodiazapentacene derivative, provides diarylnitroxide diradical 1 accompanied by an intermediate nitroxide monoradical and a multitude of isolable diamagnetic products. DFT-computed tensors for EPR spectra and paramagnetic (1)H NMR isotropic shifts for nitroxide diradical 1 show good agreement with the experimental EPR spectra in rigid matrices and paramagnetic (1)H NMR spectra in solution, respectively. Examination of the diamagnetic products elucidates their formation via distinct pathways involving C-O bond-forming reactions, including Baeyer-Villiger

Recognition and characterization of TGF-beta receptor interacting protein 1 (TRIP-1) containing WD40 repeats from Clonorchis sinensis by bioinformatics, cloning, and expression in Escherichia coli.

A cDNA clone encoding a homologue of transforming growth factor beta (TGF-beta) receptor interacting protein 1 (TRIP-1) was recognized and isolated from full-length cDNA plasmid library of Clonorchis sinensis adult. TRIP-1 is a bifunctional molecule in all eukaryote, which modulates the signaling pathway of TGF-beta as a phosphorylation substrate of TGF-beta type II receptor kinase and controls ribosome assembly and mRNA translation as p36 subunit of the eukaryotic translation initiation factor 3. The structural and immunological characteristics of TRIP-1 from C. sinensis (CsTRIP-1) were analy

Crystal structure of PhnH: an essential component of carbon-phosphorus lyase in Escherichia coli.

Organophosphonates are reduced forms of phosphorous that are characterized by the presence of a stable carbon-phosphorus (C-P) bond, which resists chemical hydrolysis, thermal decomposition, and photolysis. The chemically inert nature of the C-P bond has raised environmental concerns as toxic phosphonates accumulate in a number of ecosystems. Carbon-phosphorous lyase (CP lyase) is a multienzyme pathway encoded by the phn operon in gram-negative bacteria. In Escherichia coli 14 cistrons comprise the operon (phnCDEFGHIJKLMNOP) and collectively allow the internalization and degradation of phospho

Molecular characterization of BjussuSP-I, a new thrombin-like enzyme with procoagulant and kallikrein-like activity isolated from Bothrops jararacussu snake venom.

A thrombin-like enzyme, named BjussuSP-I, isolated from Bothrops jararacussu snake venom, is an acidic single-chain glycoprotein with M(r)=61,000, pI approximately 3.8 and 6% sugar. BjussuSP-I shows high proteolytic activity upon synthetic substrates, such as S-2238 and S-2288. It also shows procoagulant and kallikrein-like activity, but is unable to act on platelets and plasmin. These activities are inhibited by specific inhibitors of this class of enzymes. The complete cDNA sequence of BjussuSP-I with 696bp encodes open reading frames of 232 amino acid residues, which conserve the common dom

Helicobacter pylori expresses an autolytic enzyme: gene identification, cloning, and theoretical protein structure.

Helicobacter pylori is an important pathogen of the gastric system. The clinical outcome of infection is thought to be correlated with some genetic features of the bacterium. However, due to the extreme genetic variability of this organism, it is hard to draw definitive conclusions concerning its virulence factors. Here we describe a novel H. pylori gene which expresses an autolytic enzyme that is also capable of degrading the cell walls of both gram-positive and gram-negative bacteria. We designated this gene lys. We found this gene and observed its expression in a number of unrelated clinica

New 1 alpha,25-dihydroxy vitamin D3 analogues with side chains attached to C-18: synthesis and biological activity.

A new class of analogues of 1 alpha,25-dihydroxy vitamin D3 has been synthesised, in which the side chain (C-23 to C-27) has been removed and where new hydroxylated side chains have been attached to the C-18 methyl group. These analogues show antiproliferative activity in U937 and HaCaT cells comparable to that of 1 alpha,25-dihydroxy vitamin D3. Lack of calcemic activity makes these analogues potentially useful in the treatment of proliferative diseases.

A model of scorpion toxin binding to voltage-gated K+ channels.

Mutational studies have identified part of the S5-S6 loop of voltage-dependent K+ channels (P region) responsible for tetraethylammonium (TEA) block and permeation properties. Several scorpion peptide toxins-charybdotoxin (ChTX), kaliotoxin (KITX), and agitoxin (AgTX)-also block the channel with high affinity and specificity. Here, we examine the interaction predicted when the toxins are docked onto the molecular model of the K+ channel pore that we recently proposed. Docking with the model of the Kv1.3 channel started by location of Lys-27 side chain into the central axis of the pore, followe

X-ray structure of the magnesium(II)-pyrophosphate complex of the truncated head of Dictyostelium discoideum myosin to 2.7 A resolution.

The structure of the magnesium pyrophosphate complex of the truncated head of Dictyostelium myosin has been determined by molecular replacement at 2.7 A resolution and refined to a crystallographic R-factor of 16.0%. The crystals belong to the orthorhombic space group P2(1)2(1)2, where a = 105.2 A, b = 182.1 A, and c = 54.5 A. The conformation of the protein around the magnesium pyrophosphate is very similar to that seen when magnesium ADP-beryllium fluoride binds in the active site. The latter complex mimics the binding of ATP prior to hydrolysis. The pyrophosphate molecule occupies the beta-

Transglutaminase-mediated stiffening of the glomerular basement membrane mitigates pressure-induced reductions in molecular sieving coefficient by reducing compression.

Proteinuria, the presence of high molecular weight proteins in the urine, is a primary indicator of chronic kidney disease. Proteinuria results from increased molecular permeability of the glomerular filtration barrier combined with saturation or defects in tubular protein reabsorption. Any solute that passes into the glomerular filtrate traverses the glomerular endothelium, the glomerular basement membrane, and the podocyte slit diaphragm. Damage to any layer of the filter has reciprocal effects on other layers to increase glomerular permeability. The GBM is thought to act as a compressible u

Zexieyin formula attenuates Alzheimer's disease via suppressing A1 astrocyte activation: A serum pharmacochemistry and network pharmacology study.

BACKGROUND: Disruption of the blood-brain barrier (BBB) is a critical pathological event in Alzheimer's disease (AD) progression. The ZeXieYin Formula (ZXYF), as described in the ancient Chinese medical text Huangdi Neijing, has shown multitarget neuroprotective effects and promising pharmacokinetics in preclinical studies with transgenic AD rodent models. Despite these findings, the exact molecular mechanisms by which its bioactive components interact with BBB regulatory pathways are not fully understood, necessitating comprehensive analysis through integrated systems pharmacology approaches.

Molecular water pumps.

There is good evidence that cotransporters of the symport type behave as molecular water pumps, in which a water flux is coupled to the substrate fluxes. The free energy stored in the substrate gradients is utilized, by a mechanism within the protein, for the transport of water. Accordingly, the water flux is secondary active and can proceed uphill against the water chemical potential difference. The effect has been recognized in all symports studied so far (Table 1). It has been studied in details for the K+/Cl- cotransporter in the choroid plexus epithelium, the H+/lactate cotransporter in t

Molecular point-of-care: technological evolution, clinical applications, and future perspectives.

The landscape of clinical diagnostics is undergoing a fundamental shift from centralized laboratories toward decentralized point-of-care testing (POCT) models. Conventional molecular diagnostics often suffer from prolonged turnaround times, high operational costs, and a strict reliance on sophisticated infrastructure. To overcome these limitations, modern POCT platforms have deeply integrated advanced molecular techniques. These include diverse isothermal nucleic acid amplification methods (such as LAMP, RPA, and RCA) and highly specific CRISPR/Cas recognition systems. Furthermore, coupling th

MAPA: A Semantic Network Framework for Functional Module Discovery and Interpretation in Multi-Omics Data.

Multi-omics technologies generate high-dimensional molecular signatures that provide unprecedented opportunities to uncover biological mechanisms. However, translating complex molecular alterations into coherent and interpretable functional insights remains a major challenge. Existing module discovery methods can identify groups of related features, but often lack direct biological interpretability, whereas pathway-based approaches frequently yield redundant results that complicate interpretation. Here, we present MAPA (Modular Analysis and Phenotype-informed Annotation using large language mo

NLR inflammasome pathways: key targets for pathogenesis and therapy of metabolic diseases.

The NOD-like receptor (NLR) inflammasome system is an evolutionarily conserved intracellular surveillance network that responds to pathogen-associated molecular patterns (PAMPs), damage-associated molecular patterns (DAMPs), and metabolism-associated molecular patterns (MAMPs). Beyond the well-characterized NLRP3 inflammasome, accumulating evidence suggests that NLRP1, NLRP6, NLRC5, NLRP12, and NLRP2 are also implicated in metabolic disorders, including obesity, type 2 diabetes mellitus (T2DM), atherosclerosis, and metabolic dysfunction-associated steatotic liver disease (MASLD). Trajectories of late-life depression: insights from molecular imaging.

Late-life depression is associated with greater disability, suicide risk and mortality than depression in mid-life, and is a risk factor/prodrome for neurodegenerative diseases such as Alzheimer's disease and Parkinson's disease. Many depressed, older adults fail to respond to first line antidepressant treatment, experience relapse and exhibit persistent symptoms, including anxiety, apathy and cognitive impairment, that may reflect underlying neurodegenerative processes. Advances in molecular imaging, particularly positron emission tomography (PET) allow direct in-vivo investigation of neurobi

Aging is not a disease: an evolutionary and comparative biological reappraisal.

The question of whether aging should be classified as a disease has gained prominence in geroscience, fueled by advances in molecular biology and the aspiration to develop interventions that mitigate age-associated functional decline. However, evolutionary models describe aging as an emergent consequence of declining selection gradients and life-history trade-offs rather than as a deviation from species-typical function. Comparative data across taxa reveal substantial heterogeneity in aging trajectories, challenging the assumption of a uniform pathological pattern. At the molecular level, proc
