Limbal autograft transplantation for ocular surface disorders.

Limbal autograft transplantation is presented in 26 consecutive cases comprising both acute and chronic chemical injury (20 cases), thermal burns (2 cases), contact lens-induced keratopathy (3 cases), and ocular surface failure after multiple surgical procedures (1 case), with follow-up ranging from 2 to 45 months (mean, 18 months). The operative technique usually involved transfer of two free grafts of limbal tissue from the uninjured or less injured donor eye to the severely injured recipient eye, the latter having been prepared by limited conjunctival research and superficial dissection of

Animal models in lymph node transfer surgery: A systematic review.

BACKGROUND AND AIM: Lymph node transfer surgery (LNTS) is indicated in secondary lymphedema (LE) patients who do not respond to conservative therapy. Animal models are the spearhead of LE research and were used to pioneer most of the surgical interventions currently in practice. We conducted a systematic review of the literature to explore animal models dedicated to LNTS to compare different species, techniques, and outcomes. METHODS: Four databases were searched: PubMed, Cumulative Index of Nursing and Allied Health Literature, Scopus, and Web of Science. We used the Preferred Reporting Item

Delineation of the antibody light chain biases and kinetics following primary and secondary influenza virus infections of ferrets.

The ferret is considered the "gold standard" animal model for influenza virus research. However, the mechanisms of the ferret humoral immune responses remain understudied. Here, the kinetic profile of the influenza A or B virus hemagglutinin (HA)-specific primary antibody response was tracked until the contraction phase. Additionally, the acute humoral response following a secondary infection with a homosubtypic H1N1 influenza A virus was evaluated. In particular, the HA-binding reactivity in serum was quantified and the number of HA-specific antibody-secreting cells was evaluated in different

Reconceptualizing Aplastic Anemia-Seed, Worm, Soil.

BACKGROUND: Aplastic anemia (AA) encompasses a group of hematological syndromes often misdiagnosed, resulting in a decrease in the overall blood cell count and representing a form of bone marrow failure. The disease is defined by damage to hematopoietic stem cells due to various physical, chemical, and biological factors. The diagnosis of the disease is based on clinical presentation, as the underlying etiology remains poorly understood, which complicates treatment options. Understanding bone marrow failure disorders began with cytomorphological studies, which led to the identification of AA s

The role of heterogeneous nuclear ribonucleoproteins in mammalian spermatogenesis: mechanisms and clinical implications.

In brief: As a major family of RNA binding proteins, heterogeneous nuclear ribonucleoproteins (hnRNPs) are involved in the entire RNA metabolism process and have been increasingly recognized as essential regulators of spermatogenesis. This review systematically summarizes the latest advances in the functions, molecular mechanisms, and clinical applications of hnRNPs in mammalian spermatogenesis. Abstract: Spermatogenesis in mammals is a complex and highly orchestrated process of cellular development that necessitates precise regulation at the transcriptional, post-transcriptional, and translat

Detection of mitochondrial bioenergetics using a novel bimodal 3D microelectrode array (MEA)-based biosensor.

Developing new label-free paradigms for functional assays in biomedical research has the potential to catalyze efforts in drug discovery and improve the understanding of complex disorders. Mitochondria are an essential organelle in nearly every eukaryotic organism that perform vital functions such as adenosine triphosphate (ATP) production, redox signaling, reactive oxygen species (ROS) homeostasis and regulation of programmed cell death. These activities are regulated by electrophysiological processes that occur in the inner mitochondrial membrane (IMM) and outer mitochondrial membrane (OMM)

Metabolomic Insights into Lysosomal Storage Diseases: An Untargeted View.

Lysosomal Storage Diseases (LSDs) include roughly 70 inherited metabolic disorders, most of which are expressed in an autosomal recessive pattern. These conditions arise from mutations in genes encoding lysosomal enzymes, leading to intracellular buildup of substrates and subsequent lysosomal dysfunction. LSDs can be broadly classified based on the nature of the stored substrate, encompassing Sphingolipidoses, Mucopolysaccharidoses, Lysosomal Glycogen Storage Disease, Oligosaccharidoses, Mucolipidoses, and Lysosomal Proteinoses. Although individually rare, LSDs collectively affect approximatel

Status Epilepticus in Children With Hemophagocytic Lymphohistiocytosis: Literature Review.

Hemophagocytic lymphohistiocytosis (HLH) is a life-threatening disorder involving systemic inflammation and multi-organ damage, which often affects the central nervous system (CNS). Familial HLH, which is caused by mutations in genes such as PRF1 and UNC13D, impairs the function of cytotoxic cells, leading to uncontrolled immune activation. CNS involvement occurs in 30% to 73% of cases and manifests as seizures, encephalopathy or status epilepticus, which often mimics other neurologic conditions. This article presents the case of a 28-month-old boy with PRF1-related HLH who developed super-ref

Insights Into the Antibiofilm Activity of Carbon Quantum Dots Against a Panel of Different Microorganisms: A Review.

Biofilm-associated infections account for a majority of chronic bacterial diseases. These infections also present a therapeutic challenge due to their intrinsic tolerance to conventional antibiotics. Carbon dots (CDs) have emerged as a versatile nanoplatform with the potential to combat these resilient structures. This review critically evaluates the burgeoning field of CD-based antibiofilm agents. We move beyond a catalog of studies to analyze how the physicochemical properties of CDs dictate their mechanisms of action against a panel of microorganisms. The antibiofilm activity of CDs is mult

CRISPR/Cas9 and iPSC-Based Therapeutic Approaches in Alzheimer's Disease.

Alzheimer's disease (AD), the leading cause of dementia, remains poorly understood despite decades of intensive research, which continues to hinder the development of effective treatments. As a complex multifactorial disorder, AD lacks a cure to halt the progressive neurodegeneration, and the precise mechanisms underlying its onset and progression remain elusive, limiting therapeutic options. Due to the challenges of studying neuronal cells in vivo, technologies such as clustered regularly interspaced short palindromic repeats/CRISPR-associated protein 9 (CRISPR/Cas9) and human-induced pluripo

Prominin-2/FBXO22/BACH1 axis protects bone marrow mesenchymal stem cells against TBHP-induced ferroptosis and ameliorates intervertebral disc degeneration.

BACKGROUND: Our preliminary research has revealed that Prominin-2 overexpression effectively guarded against oxidative stress (OS)-induced ferroptosis by decreasing BTB and CNC homolog 1 (BACH1) expression, thus promoting bone marrow mesenchymal stem cells (BMSCs) survival under the OS microenvironments in degenerative discs. METHODS: In this study, we probed how Prominin-2 controls the BACH1 expression in OS-induced BMSC ferroptosis. We then evaluated the efficiency of targeted Prominin-2/BACH1 pathway in BMSCs in treating degenerative nucleus pulposus cells (NPCs) and intervertebral disc de

Application of novel strategies in chronic wound management with focusing on pressure ulcers: new perspective.

Invading blood cells, extracellular tissue, and soluble mediators all play important roles in the wound-healing process. There is a substantial global burden of disease and mortality attributable to skin defects that do not heal. About 1% to 2% of the population in industrialized nations suffers from chronic wounds that don't heal, despite healthcare breakthroughs; this condition is very costly, costing about $25 billion each year in the US alone. Amputation, infection (affecting as many as 25% of chronic wounds), sepsis, and dermal replacements are all consequences of conventional therapeutic

Brain and Immune System: Intercellular Communication During Homeostasis and Neuroimmunomodulation upon Dysfunction.

The review compares the principles of organization of the brain and immune system, two important organs developed over 500 million years in multicellular organisms, including humans. It summarizes the latest results from research in neurosciences and immunology concerning intercellular communication. While in the brain, intercellular communication is primarily based on exchange of electrical signals, this is not the case in the immune system. The question, therefore, arises as to whether nature developed two entirely different systems of organization. It will be demonstrated that a few basic p

Clinical implications of miRNAs in erythropoiesis, anemia, and other hematological disorders.

Erythropoiesis is regulated by the differential expression of many genes. Besides being transcriptionally regulated, these genes are also with the oath of epigenetic regulation by the microRNAs (miRNAs), in particular. Various miRNAs appear to be very important for the normal process of erythropoiesis and various hematological abnormalities in humans. Therefore, the review aims to summarize the significance of miRNAs in erythropoiesis and different hematological diseases with clinical importance. Our analysis indicates that specific miRNAs regulate erythropoiesis in a stage-specific manner fro

Good manufacturing practices production of human placental derived mesenchymal stem cells for therapeutic applications: focus on multiple sclerosis.

BACKGROUND: Among neurological diseases, multiple sclerosis (MS) affects mostly young adults and can cause long-term disability. While most medications with approval from regulatory agencies are very effective in treating MS disease, they are unable to repair the tissue damage found in the central nervous system (CNS). Consequently, Cell-based therapy particularly using mesenchymal stem/stromal cells (MSCs), holds promise for neuroprotection and tissue repair in MS treatment. Furthermore, placenta-derived MSCs (PLMSCs) have shown the potential to treat MS due to their abundance, noninvasive is

Progress and application of adipose-derived stem cells in the treatment of diabetes and its complications.

Diabetes mellitus (DM) is a serious chronic metabolic disease that can lead to many serious complications, such as cardiovascular disease, retinopathy, neuropathy, and kidney disease. Once diagnosed with diabetes, patients need to take oral hypoglycemic drugs or use insulin to control blood sugar and slow down the progression of the disease. This has a significant impact on the daily life of patients, requiring constant monitoring of the side effects of medication. It also imposes a heavy financial burden on individuals, their families, and even society as a whole. Adipose-derived stem cells (
