Biological therapy of strontium-substituted bioglass for soft tissue wound-healing: responses to oxidative stress in ovariectomised rats.

New synthetic biomaterials are constantly being developed for wound repair and regeneration. Bioactive glasses (BG) containing strontium have shown successful applications in tissue engineering account of their biocompatibility and the positive biological effects after implantation. This study aimed to assess whether BG-Sr was accepted by the host tissue and to characterize oxidative stress biomarker and antioxidant enzyme profiles during muscle and skin healing. Wistar rats were divided into five groups (six animals per group): the group (I) was used as negative control (T), after ovariectomy…

Organ-specific colonization and niche remodeling in a human tissue model of metastasis.

For metastatic colonization to occur, disseminated tumor cells must survive, adapt to, and remodel distant microenvironments in an organ-specific manner. We established a human multitissue model of cancer spread, with engineered bone and lung linked by vascular flow containing circulating cancer cells. Parental MDA-MB-231 cells extravasated toward both tissues, remodeled their niches, and acquired transcriptional programs reflecting adaptation to the local microenvironment, particularly upon homing to bone. Tissue-specific colonization by the bone- and lung-tropic MDA-MB-231 derivatives was qu

Exosomal delivery of NF-κB inhibitor delays LPS-induced preterm birth and modulates fetal immune cell profile in mouse models.

Accumulation of immune cells and activation of the pro-inflammatory transcription factor NF-κB in feto-maternal uterine tissues is a key feature of preterm birth (PTB) pathophysiology. Reduction of the fetal inflammatory response and NF-κB activation are key strategies to minimize infection-associated PTB. Therefore, we engineered extracellular vesicles (exosomes) to contain an NF-κB inhibitor, termed super-repressor (SR) IκBα. Treatment with SR exosomes (1 × 1010 per intraperitoneal injection) after lipopolysaccharide (LPS) challenge on gestation day 15 (E15) prolonged gestation by over 24 ho

Engineered Charge Redistribution of Gp2 Proteins through Guided Diversity for Improved PET Imaging of Epidermal Growth Factor Receptor.

The Gp2 domain is a protein scaffold for synthetic ligand engineering. However, the native protein function results in a heterogeneous distribution of charge on the conserved surface, which may hinder further development and utility. We aim to modulate charge, without diminishing function, which is challenging in small proteins where each mutation is a significant fraction of protein structure. We constructed rationally guided combinatorial libraries with charge-neutralizing or charge-flipping mutations and sorted them, via yeast display and flow cytometry, for stability and target binding. Simulations of transcatheter aortic valve implantation: implications for aortic root rupture.

Aortic root rupture is one of the most severe complications of transcatheter aortic valve implantation (TAVI). The mechanism of this adverse event remains mostly unknown. The purpose of this study was to obtain a better understanding of the biomechanical interaction between the tissue and stent for patients with a high risk of aortic rupture. We simulated the stent deployment process of three TAVI patients with high aortic rupture risk using finite element method. The first case was a retrospective analysis of an aortic rupture case, while the other two cases were prospective studies, which en

From Magnetic Moment to Magnetic Particle Imaging: A Comprehensive Review on MPI Technology, Tracer Design and Biological Applications.

Background/Objectives: Magnetic nanoparticles have emerged as powerful tools for biomedical imaging, targeted drug delivery, and hyperthermia therapy. Magnetic particle imaging (MPI) is among the most promising technologies built around its properties: a radiation-free, quantitative tomographic modality that detects superparamagnetic iron oxide nanoparticles (SPIONs) directly against a biologically silent background. This review synthesizes MPI's physical principles, nanoparticle design strategies, and preclinical applications within the broader landscape of magnetic material engineering for b

Multi-emissive metal nanoclusters: luminescence origin, tailoring strategies, and biomedical applications.

Multicolor (multi-emissive) luminescent materials are attracting increasing attention because, compared with single-emission systems, they can deliver two or more emission bands under the same or different excitation conditions, enabling ratiometric readouts that suppress environmental and instrumental interference and improve quantitative reliability in complex bioenvironments. Ligand-protected metal nanoclusters (MNCs), consisting of several to hundreds of metal atoms, provide a particularly powerful single-component platform because their ultrasmall, molecule-like electronic structures and

Optimised amine density via plasma treatment for covalent immobilisation of AMPs to promote osteoblasts and enhance antimicrobial activity.

Bacterial infections remain a leading cause of failure in biomedical implants. To mitigate this, surface modification strategies have been developed, commonly relying on bactericidal agents such as antibiotics to eliminate attached bacteria. However, the rise of antibiotic resistance has significantly reduced the long-term success of these approaches. Antimicrobial peptides (AMPs), such as KR-12, have emerged as promising alternatives due to their strong bactericidal activity and low risk of inducing resistance. This study presents a surface engineering strategy that combines the antimicrobial

Dynamic coordination engineering of gallium biomaterials: A biological accessibility framework for therapeutic biofunction.

Gallium (Ga)-based biomaterials have emerged as versatile therapeutic platforms owing to the iron-mimicking properties and broad biological activities of gallium. Coordination engineering regulates Ga speciation, stability, transport, and therapeutic activation; however, optimizing individual physicochemical properties does not necessarily improve biological performance. Excessive coordination stability may restrict ligand exchange and therapeutic activation, whereas insufficient stability may cause premature dissociation, nonspecific exposure, and reduced target delivery. In this Review, we p

Multi-cellular engineered living systems to assess reproductive toxicology.

Toxicants and some drugs can negatively impact reproductive health. Many toxicants haven't been tested due to lack of available models. The impact of many drugs taken during pregnancy to address maternal health may adversely affect fetal development with life-long effects and clinical trials do not examine toxicity effects on the maternal-fetal interface, requiring indirect assessment of safety and efficacy. Due to current gaps in reproductive toxicological knowledge and limitations of animal models, multi-cellular engineered living systems may provide solutions for modeling reproductive physi
