Myeloid-Derived Suppressor Cells in Bladder Cancer: An Emerging Target.

Bladder cancer remains a prevalent and challenging malignancy. Myeloid-derived suppressor cells (MDSCs) have emerged as key contributors to the immunosuppressive tumor microenvironment, facilitating tumor progression, immune evasion, and resistance to therapies. This review explores the role of MDSC in bladder cancer, highlighting their involvement in immune regulation; tumor progression; and resistance to therapies such as bacillus Calmette-Guérin (BCG) therapy, chemotherapy, and immune checkpoint inhibitors (ICIs). We also discuss their potential as biomarkers and therapeutic targets, with c…

A red cell membrane-camouflaged nanoreactor for enhanced starvation/chemodynamic/ion interference therapy for breast cancer.

In this study, a multifunctional Cu-doped CaO2 nanoreactor loaded with GOx and camouflaged with a folic acid-modified cell membranewas developed for breast cancer treatment. The as-developed composite nanoreactor showed a synergistic effect on calcium overload to damage mitochondria, thus killing tumor cells to achieve ion interference therapy (IIT). The loaded GOx could deplete glucose to "starve" tumor cells. The H2O2 released by CaO2 decomposition and enzyme catalytic reactions from GOx could not only be highly toxic in the tumor microenvironment but also enhance the efficiency of chemodyna…

Bacterial immunotherapy: is it a weapon in our arsenal in the fight against cancer?

Advances in understanding the genetic basis of cancer have driven alternative treatment approaches. Recent findings have demonstrated the po … stics. Genetically modified bacteria has reduced inherent toxicity and enhanced specificity towards tumor microenvironment. This anti- tumor activity of bacteria is attributed to its toxins and other active components from the cell membrane, cell wall and spores. Furthermore, bacterial genes can be regulated to express and deliver cytokines, antibodies and cancer therap …

Tumorigenic functions of serglycin: Regulatory roles in epithelial to mesenchymal transition and oncogenic signaling.

Numerous studies point out serglycin as an important regulator of tumorigenesis in a variety of malignancies. Serglycin expression correlate … , evoking cell adhesion and signaling. Serglycin also creates a pro-inflammatory and pro-angiogenic tumor microenvironment by regulating the secretion of proteolytic enzymes, IL-8, TGFβ2, CCL2, VEGF and HGF. Hence, serglycin activates multiple signaling cascades that drive angiogenesis, tumor cell growth, epithelial to mesenchymal transition, cancer cell stemness and me …

Exosome-mediated cell-cell communication in tumor progression.

Exosomes, which are 30-150 nm lipid bilayer vehicles, have been recognized as one of the most crucial components of the tumor microenvironment. Exosomes transfer specific lipid, nucleic acids, proteins and other bioactive molecules from the donor cells to the recipient cells. Accumulating evidence has suggested that cancer cells and the tumor associated stromal cells can release and receive exosomes, inside of which the components and amounts are greatly changed. Pioneering studies have revealed that these exosomes play essential roles in tumor progression. Here we summarize the recent advance…

A syngeneic orthotopic mouse model of metastatic colorectal cancer.

Colorectal cancer (CRC) development is initiated in the colon-rectum sections of the gut, by the emergence of a primary tumor. CRC slowly progresses to a multiple locations metastatic disease, involving secondary tumors arising in various organs, such as the liver. Mouse models have been developed to investigate the immune response, locally generated in primary tumors. Classically, tumors are implanted under the skin, for practical reasons and simplicity of monitoring. However, the skin location does not necessarily recapitulate the tumor immune microenvironment (TME) that would normally be ge

Advances in Research on the B-Lineage Transcription Factor EBF1 in Solid Tumors.

Early B-cell factor 1 (EBF1) is a crucial transcription factor that governs the development and differentiation of B lymphocytes. Furthermore, it is essential in developing multiple organs and tissues. The functional dysregulation of EBF1 expression is intricately associated with the occurrence, recurrence, and treatment resistance of B-lineage acute lymphoblastic leukemia. In recent years, EBF1 has demonstrated a more intricate and multifaceted role in solid tumors. It does not immutably adhere to the conventional classification of tumor suppressor genes. On the contrary, EBF1 is a flexible r

Immunosuppressive Extracellular Vesicles as a Linking Factor in the Development of Tumor and Endometriotic Lesions in the Gynecologic Tract.

Both gynecological tumors and endometriosis require for their development a favorable environment, termed in the case of tumors a "pre-metastatic niche" and in case of endometriosis a "pro-endometriotic niche". This is characterized by chronic inflammation and immunosuppression that support the further progression of initial lesions. This microenvironment is established and shaped in the course of a vivid cross-talk between the tumor or endometrial cells with other stromal, endothelial and immune cells. There is emerging evidence that extracellular vesicles (EVs) play a key role in this cellul

Reprogramming tumor-associated macrophages in breast cancer: from myeloid plasticity and tumor-promoting programs to CAR-macrophage therapy.

Breast cancer develops within spatially organized, multicellular microenvironments in which particular tumor-associated macrophage (TAM) states can influence immune access and stromal remodeling. Conventional M1/M2 classification and total macrophage density fail to capture the developmental, transcriptional, metabolic, and spatial heterogeneity that determines TAM function. This macrophage-focused review examines breast cancer within its multicellular microenvironment, integrating lineage-tracing, single-cell, spatial, mechanistic, and clinical evidence while distinguishing breast-specific fi

Lineage plasticity-driven immunometabolic state transitions in advanced prostate cancer: biomarkers and therapeutic stratification.

Advanced prostate cancer is commonly described as immunologically cold, yet that label obscures a disease continually remodelled by treatment. Androgen-receptor (AR) pathway inhibition, chemotherapy, DNA-damage-directed therapy and radioligand treatment select resistant clones while altering lineage identity, metabolic dependence, antigen presentation and metastatic niches. AR-high, AR-low, double-negative, amphicrine and treatment-emergent neuroendocrine phenotypes are therefore better viewed as evolving immunometabolic states than as fixed histologies. Here, we develop a lineage-metabolism-i

Longitudinally Monitoring Prostate Cancer Bone Metastasis and Treatment Response via Circulating Extracellular Vesicles.

Lethal prostate cancer (PCa) is dominated by treatment-refractory progression in bone. In this study, we used plasma-derived extracellular vesicles (EVs) for monitoring the tumour and therapy-induced changes in the bone tumour microenvironment (bone-TME) in men with bone metastatic PCa (M1b PCa). Transcriptome analysis of differentially expressed genes in plasma EVs from patients with M1b and localized (M0) PCa, identified a 'bone metastasis signature' (BMS) that reflects tumour-induced bone formation. In preclinical studies, the BMS was enriched in plasma EVs from mice inoculated with bone-me

Bionic Copper-Piceatannol for Polyamine Depletion-Synergized Radioimmunotherapy.

Elevated intracellular glutathione, intratumoral hypoxia, and excessive polyamines severely compromise the efficacy of radiotherapy (RT) by attenuating radiation damage and maintaining an immunosuppressive tumor microenvironment (ITME). Strategies that concurrently address these obstacles are urgently needed to enhance RT outcomes. Herein, we fabricated a biomimetic nanoplatform (PCuP) via the self-assembly of copper ions and piceatannol into CuP nanometal-polyphenol cores, followed by surface camouflage with platelet membranes (PM). Owing to the platelet membrane coating, PCuP preferentially …

Recapitulating patient-to-patient colorectal cancer tumor heterogeneity using patient-derived xenograft cells in an engineered tissue model.

Establishing in vitro cancer models that more closely recapitulate patient tumor microenvironmental heterogeneity, including variations in stromal cells and mechanical properties that influence colorectal cancer (CRC) progression, is crucial for advancing CRC research. This study evaluated the ability of 3D engineered CRC-PDX (3D-eCRC-PDX) tissues to recapitulate the heterogeneity found between patient-derived xenograft (PDX) tumors from three CRC patients (stage II, III-B, and IV). To form the 3D-eCRC-PDX tissues, CRC-PDX tumor cells were encapsulated in PEG-fibrinogen hydrogels and maintaine…

Small Extracellular Vesicles-Derived Circ6718 Unlocks Stromal Remodeling and Serves as a Biomarker in Gastric Cancer.

Small extracellular vesicles (sEVs)-derived circular RNA (circRNA) serves as a crucial biomarker for diagnosing gastric cancer and as a key regulator of tumor progression, orchestrating intercellular crosstalk within the tumor microenvironment (TME). In gastric cancer (GC), tissue-derived mesenchymal stem cells (GC-MSCs) critically drive tumor progression; however, the interplay between sEVs and circRNA in GC-MSCs remains incompletely understood. We identified sEVs-hsa_circ_0006718 (circ6718) as significantly upregulated in gastric cancer patients. Elevated levels of sEVs-circ6718 correlated w…

Metabolic regulation of tumor-associated macrophage function and immunotherapy in cancer.

Tumor-associated macrophages (TAMs), essential components of the tumor immune microenvironment (TIME), undergo metabolic reprogramming as part of functional adaptation. Tumor cells modulate TAMs through multiple mechanisms, including metabolic cross-feeding, cytokine production, extracellular vesicles, tumor-derived proteins (such as GRP78) and pathogen-associated patterns (such as Lipopolysaccharide) signaling mediators. In turn, metabolic alterations in TAMs fine-tune TAM function via intricate signaling networks with outcomes that vary across cancer types. These functional and phenotypic sh
