Acquisition of pig intramuscular preadipocytes through dedifferentiation of mature adipocytes and establishment of optimal induction conditions.

Intramuscular fat deposition is a major contributing factor to variations in pork quality. Understanding the mechanisms driving the differentiation and metabolism of muscle-derived adipocytes is important for regulating the fat deposition in muscle. Studies on intramuscular adipocytes commonly involve stromal-vascular (SV) cell cultures, which contain preadipocytes but also several other types of primordial cells. Hence, it is crucial to obtain pure intramuscular preadipocytes for investigating adipocyte differentiation and metabolism in muscle tissue. In this study, we established cultures of…

Higher levels of ATGL are associated with exercise-induced enhancement of lipolysis in rat epididymal adipocytes.

BACKGROUND: In adipose cells, adipose triglyceride lipase (ATGL) catalyzes the first step in adipocyte triacylglyceride hydrolysis, thereby regulating both basal and hormone-stimulated lipolysis. However, little is known about the molecular mechanism(s) underlying habitual exercise-induced adaptive modulation of ATGL in white adipocytes via alteration in transcription regulator and lipolytic cofactors. METHODOLOGY/PRINCIPAL RESULTS: Male Wistar rats were randomly divided into 2 groups a sedentary control group (CG) and a habitual exercise group (EG). The EG was subjected to running on a tread…

Rimming of adipocytes by neoplastic lymphocytes: a histopathologic feature not restricted to subcutaneous T-cell lymphoma.

Subcutaneous panniculitis-like T-cell lymphoma (SPTCL) is a rare cytotoxic T-cell lymphoma of the skin presenting with histopathologic features simulating those of a lobular panniculitis. The presence of neoplastic T-lymphocytes forming a rim around the individual fat cells in the subcutaneous lobules, so-called "rimming" of adipocytes, is considered a characteristic morphologic feature of this type of cutaneous lymphoma. In this study we reviewed a series of 45 biopsy specimens of primary and secondary cutaneous B- and T-cell lymphomas and one of myeloid leukemia involving the subcutaneous ti…

Recombinant human FIZZ3/resistin stimulates lipolysis in cultured human adipocytes, mouse adipose explants, and normal mice.

Human FIZZ3 (hFIZZ3) was identified as an ortholog of mouse resistin (mResistin), an adipocyte-specific secreted factor linked to insulin resistance in rodents. Unlike mResistin, hFIZZ3 is expressed in macrophages and monocytes, but is undetectable in adipose tissue. The profound macrophage infiltration of adipose that occurs during obesity suggests that hFIZZ3 may play an important role in adipocyte biology. Using a recombinant protein produced in Escherichia coli, we report here that chronic treatment of cultured human adipocytes with hFIZZ3 results in hypotropic cells with smaller lipid dro…

CB1- and CB2-cannabinoid receptor-independent lipolysis induced by WIN 55,212-2 in male rat adipocytes.

The expression of genes encoding the cannabinoid CB(1) and CB(2) receptors and fatty acid amide hydrolase (FAAH) and the lipolytic activity of cannabinoid agonists were investigated in rat adipose tissue.RT-PCR studies indicated that the genes encoding CB(1) and CB(2) receptors and FAAH are not expressed in epididymal adipocytes. In functional studies, the non-selective cannabinoid receptor agonist WIN 55,212-2 concentration-dependently (0.01-30 micro M) induced glycerol release above baseline ( E(max) 96.1+/-6.2% of isoprenaline-induced lipolytic response). The selective CB(2) agonist JWH-015…

Isoproterenol inhibits resistin gene expression through a G(S)-protein-coupled pathway in 3T3-L1 adipocytes.

Resistin was recently identified as a hormone secreted by adipocytes which leads to insulin resistance in vivo and in vitro and might therefore be an important link between obesity and diabetes. To clarify the regulation of resistin gene expression, 3T3-L1 adipocytes were treated with various agents known to modulate insulin sensitivity, and resistin mRNA was measured by quantitative real-time reverse transcription-polymerase chain reaction. Interestingly, isoproterenol treatment reduced the level of resistin mRNA to 20% of non-treated control cells. This effect was dose-dependent with signifi…

Insulin binding and glucose transport in adipocytes in neonatal streptozocin-injected rat model of diabetes mellitus.

The neonatal streptozocin (STZ)-injected rat (NSIR) model of diabetes mellitus resembles human non-insulin-dependent diabetes mellitus (NIDDM) with respect to abnormalities in insulin secretory responses. The suggestion that insulin deficiency leads to insulin resistance, a prominent feature of human NIDDM, led us to examine insulin binding and glucose transport in the NSIR during the development of hyperglycemia. Male Wistar rats were injected at 2 days of age with STZ (90 mg/kg i.p.) or vehicle alone. Mild insulin deficiency, reflected by minimally decreased fed plasma insulin concentrations

Are the metabolic characteristics of congenital intraspinal lipoma cells identical to, or different from normal adipocytes?

Congenital intraspinal lipomas are frequently responsible for progressive neurological deficits caused by distortion or compression of the nervous system. Since fat metabolism in these lesions has not been previously studied, the aim of this study was to determine whether intraspinal lipoma cells behave like lipomas or like normal adipocytes. In 11 patients, intraspinal lipoma cells were compared with normal adipocytes isolated from adjacent subcutaneous adipose tissue for the following parameters: lipoprotein lipase (LPL), lipogenesis from U14C glucose, beta-receptor number, adenylate cyclase…

Effect of p-chlorophenoxyisobutyrate on the antilipolytic action of insulin and insulin binding in isolated adipocytes.

The present study was undertaken to investigate the potentiation by p-chlorophenoxyisobutyrate (CPIB) of the antilipolytic effect of insulin in isolated adipocytes from rats fed a (1) sucrose diet, (2) glycerol-lard diet, or (3) chow diet. CPIB supplementation in the diet consistently resulted in decreased serum triglyceride levels in rats from the three dietary groups. The catecholamine-stimulated glycerol release was significantly depressed to a greater extent by insulin when the fat cells were obtained from rats given CPIB compared to those without drug treatment. The enhanced insulin sensi…

Engineering 3D adipose cell models by support bath bioprinting of mature adipocytes in gellan gum hydrogels.

Mature adipocytes (ACs) are essential for modeling human adipose tissue physiology in vitro, yet their large size, buoyancy, and shear sensitivity make them challenging to incorporate into engineered three-dimensional (3D) constructs. This study introduces an agarose support-bath (SB) extrusion workflow for bioprinting geometrically defined adipose constructs using a gellan gum (GG) bioink. We examined the viscosity of GG solutions and SBs to identify a low-shear printability window suitable for lipid-laden cells. SB printing significantly enhanced filament stability, pore uniformity, and cons…

Palmitoleic acid n7 improves glucose uptake and metabolism and attenuates oxidative stress in human visceral adipocytes.

PURPOSE: Obesity-associated dysfunction of visceral white adipose tissue (vWAT) is characterized by impaired glucose handling, altered adipokine secretion, and redox imbalance, contributing to metabolic deterioration. Palmitoleic acid (16:1n-7), a monounsaturated fatty acid, has been implicated in regulation of glucose and lipid metabolism in rodent and cellular models; however, its effects on human adipocytes under metabolically compromised conditions remain incompletely understood. Here, we investigated the effects of in vitro treatment with 16:1n-7 on glucose uptake and metabolism, adipokin…

Bioprinting functional fat - adipocytes as the next frontier in tissue engineering, regenerative medicine, and cultivated meat.

Adipose tissue is a vital component in both regenerative medicine and cultivated meat, yet the selection of the cellular starting material remains an underexplored determinant of adipose construct performance in bioprinting. This mini-review explores next-generation adipose tissue engineering, emphasizing bioprinting functional fat with adipocytes at the intersection of these two fields. While current adipose bioprinting strategies predominantly rely on adipose-derived stem or stromal cells for their expandability and manufacturability, mature adipocytes (ACs) and microfat emerge as alternativ…

In Vitro Evaluation of Mechanical Fat Processing Devices: Impact on Adipocytes and Adipose-Derived Stem Cells.

In the fat grafting procedure, the new European Regulations and the necessity to perform minimal manipulation processing require the verification of security and performance levels of each device and, in this particular context, functional and tissue responses. The aim of this study is to evaluate the integrity of adipocytes and adipose-derived stromal/stem cells (ADSCs) after their processing using two new mechanical devices operating with a closed system. In this study, two new fat processing systems were assessed. We also considered the biological results of the processing and compared them…

Cancer-Associated Adipocytes in Human Breast Cancer: An Observational Histopathological Study of Dedifferentiation and Stromal Transition.

BACKGROUND: Adipose tissue is a major stromal component of the breast cancer (BC) tumor microenvironment (TME), playing a crucial role in BC progression. Cancer-associated adipocytes (CAAs), located at the invasive tumor front, undergo significant morphological and functional alterations. This observational case-control study investigated the dedifferentiation trajectory of CAAs and its impact on BC progression. METHODS: Paired tumor and distant normal adipose tissues from 20 BC patients were analyzed. Histological and immunohistochemical analyses were performed to assess morphological change…

Fat's all, folks: culturing and manipulating peri-prostatic adipocytes to probe impacts on prostate cancer biology.

Obesity, officially recognised as a global epidemic by the World Health Organization, will soon overtake smoking as the largest preventable risk factor for cancer. By 2035, more than half the world's population is expected to be overweight or obese with a significant increase in obesity-related health expenditures. However, despite the increase in prevalence and the overall lower life expectancy associated with obesity, mechanisms underpinning obesity-driven diseases are not well understood. Adipocytes pose many challenges for in vitro culture due to their poor cell-to-surface attachment and l…

Imeglimin enhances GLUT4-mediated glucose uptake and is more effective than metformin in 3T3-L1 adipocytes.

Imeglimin, a novel oral antidiabetic agent derived from metformin, improves insulin secretion and sensitivity in patients with type 2 diabetes. Recent clinical clamp studies have identified adipose tissue as a potential target of imeglimin; however, its direct effects on adipocytes are unclear. In this study, differentiated 3T3-L1 adipocytes were treated with imeglimin or metformin. We then examined glucose concentration in the culture medium, glucose uptake, plasma membrane glucose transporter expression, and the effects of signaling inhibitors. Imeglimin enhanced glucose uptake in a dose-dep…

Not just fillers: the emerging role of bone marrow adipocytes in bone and bone marrow homeostasis.

Bone marrow adipose tissue (BMAT) is a phylogenetically conserved fat depot occupying a substantial fraction of the medullary cavity in vertebrates, yet its functional significance has only recently begun to be appreciated. Long dismissed as an inert filler, BMAT is now recognized as a dynamic and metabolically active compartment participating in the regulation of bone remodeling and hematopoiesis. Bone marrow adipocytes (BMAds) originate from skeletal stem cells (SSCs) and are anatomically and functionally distinct from peripheral white adipocytes, exhibiting a unique lipid metabolism and red…
