A preparation was developed that allows for the recording of single-unit chemoreceptor activity from mouse carotid body in vitro. An anesthetized mouse was decapitated, and each carotid body was harvested, along with the sinus nerve, glossopharyngeal nerve, and petrosal ganglia. After exposure to collagenase/trypsin, the cleaned complex was transferred to a recording chamber where it was superfused with oxygenated saline. The ganglia was searched for evoked or spontaneous unit activity by using a glass suction electrode. Single-unit action potentials were 57 +/- 10 (SE) (n = 16) standard devia

Many free nerve endings of small-diameter afferents (A-delta or C nerve fibres) respond to craniofacial noxious stimuli and a number of neurochemicals are involved in their activation or sensitization. The small-diameter nociceptive afferents project to the trigeminal (V) brainstem complex where they can excite nociceptive neurones that have been categorized as either nociceptive-specific (NS) or wide dynamic range (WDR). These neurones project to other brainstem regions or to the contralateral thalamus. The lateral and medial thalamus contain NS and WDR neurones which have properties and conn

We describe a method for characterizing and studying afferent input to the central nervous system arising from deep axial structures of the neck during defined cervical vertebral movement. Multiple or single unit recordings of afferent activity arising from identified receptive fields in these tissues can now be studied in situ while simultaneously inducing 'natural' stimulation of mechanoreceptors during well defined movements of the intact vertebral column. When combined with existing strategies for extracellular and intracellular recordings of neurones, the methods described here will allow

Proprioception-related somatosensory evoked potentials (SEPs) to passive flexion movement of the middle finger at proximal interphalangeal joint were recorded in 7 patients with myoclonus of cortical origin who demonstrated enlarged electrical SEPs (giant SEPs). In 3 out of the 7 patients, the proprioception-related SEPs were also enlarged. The remaining 4 patients showed giant electrical SEPs without enhancement of proprioception-related SEPs. Long loop electromyographic response was recorded during the resting condition in all of the 3 patients with enlarged proprioception-related SEPs. We h

The head of adult hagfishes (jawless craniates, Myxinoidea) of the family Eptatretidae displays a number of skin grooves of uncertain origin. These grooves have been homologized to the neuromast lines of other craniates, and they are innervated by two ganglionated cranial nerves that have been interpreted as lateral line nerves. The grooves do not, however, contain the compound receptors that are typical of a lateral line (i.e. neuromasts or electroreceptors), and both their development and function have remained enigmatic. To elucidate the embryonic origin of the grooves (which should develop

We studied the ability of patients with cerebellar degeneration to perceive differences in kinaesthetic stimuli and compared it with that of normal subjects. All participants were tested for duration, amplitude and velocity sensation. In separate experiments, the responses of muscle spindle afferents and slowly adapting cutaneous mechanoreceptors to the kinaesthetic stimuli were recorded. The performance of patients with cerebellar degeneration was significantly worse than that of normal subjects on the tasks testing for duration and velocity perception. Although both spindle afferents and slo

In leech, the central projections of peripheral sensory neurons segregate into specific axonal tracts, which are distinguished by differential expression of surface antigens recognized by the monoclonal antibodies Lan3-2 and Lan4-2. Lan3-2 recognizes an epitope expressed on axons that segregate into three distinct axon fascicles. In contrast, the Lan4-2-positive axons selectively project into only one of the Lan3-2-positive axon tracts. These observations provide evidence for a hierarchy of guidance cues mediating specific pathway formation in this system. Since the Lan3-2 antibody has been sh

Dorsal root entry zone (DREZ) lesions were used to treat intractable pain due to deafferentation in 78 patients managed between 1981 and 1988. Etiology of pain included avulsion of brachial or lumbosacral plexuses (27 cases), spinal cord injury (20 cases), amputation (nine cases), post-herpetic neuralgia (16 cases), and cauda equina injury (six cases). Three different lesioning techniques were employed: a radiofrequency (rf) method using a 0.5 X 2-mm stainless steel electrode with control of electric current and duration (Group 1: 21 cases); the CO2 laser (Group 2: 20 cases); and an rf method,

Interaural phase and intensity are cues by which the barn owl determines, respectively, the azimuth and elevation of a sound source. Physiological studies indicate that phase and intensity are processed independently in the auditory brainstem of the barn owl. The phases of spectral components of a sound are encoded in nucleus magnocellularis (NM), one of the two cochlear nuclei. NM projects solely and bilaterally to nucleus laminaris (NL), wherein interaural phase difference is computed. The other cochlear nucleus, nucleus angularis (NA), encodes the amplitudes of spectral components of sounds

The purpose of these experiments is to see if reflex nervous hyperventilation obtained by passive limb movements in man is a consequence of the activation of a direct ascending nervous pathway from mechanoreceptors of moving limbs to respiratory centres, or if it is the consequence of a primary enhanced cardiodynamic activity. Contemporaneous measurements of pulmonary ventilation and cardiac output (the latter with the trasthoracic bioimpedance method) were carried out in subjects whose ankles were passively moved at an angle of 10 degrees and a frequency of 1 Hz, each test lasting three breat

The afferent renal nerves enhance sympathetic activity in the one-kidney, one-clip hypertensive rat. We have also found adenosine-sensitive nerve endings in the renal pelvis that, when stimulated, increase sympathetic activity producing hypertension. To determine whether adenosine, which is excreted when renal blood flow is reduced, activates the afferent renal nerves in one-kidney, one-clip hypertension, urinary adenosine concentration was lowered by infusing adenosine deaminase into the renal artery. Urinary adenosine concentration was threefold greater in one-kidney, one-clip hypertensive a

The interpeduncular nucleus (IPN) is a midbrain structure that receives its major afferents from the medial habenulae via the fasciculi retroflexi. Among the axons projecting to the IPN is a population of substance P (SP)-containing axons. The IPN has been subdivided into the central, dorsal, intermediate, rostral, and lateral subnuclei using cytoarchitectonic criteria. The distribution of SP among these subnuclei was determined by using Sternberger's ('79) peroxidase-antiperoxidase technique. In the normal IPN the rostral subnucleus can be subdivided into two sectors on the basis of SP conten

The present study was undertaken in order to determine the subnuclear origin of olivary parasagittal zones in the uvula of the rat cerebellum. Microinjections of horseradish peroxidase were made in different sublobules of the uvula ( sublobules IXa, b, and c of Larsell , 1952) and in different mediolateral positions within these sublobules . The results suggest that different specific regions of the contralateral inferior olive project to each of the parasagittal zones found within the uvula. Injections in the region of the most medial parasagittal zone result in the retrograde labeling of cel

The primary sensory neurons in the olfactory and vomeronasal mucosae develop outside the neuraxis from ectodermal placodes, and their axons enter the central nervous system (CNS) during embryonic life, to terminate in the olfactory and accessory olfactory bulbs. The sensory neurons are unique in that they are produced and differentiate continuously from a stem cell in the sensory epithelium, throughout the life of the animal. After loss of sensory neurons following injury, the stem cell is able to increase its rate of division and replace the lost neurons. Sensory neurons thus formed during ad

The hypothesis was tested which states that the somatic deafferentation syndrome is a visually prompted response to sensorimotor loss. The dorsal roots, C5-T2, were bilaterally cut in a strain of rats known to be genetically blind. These complete dorsal rhizotomies left the forelimbs totally anesthetic, analgesic and paretic. Contact and visual placing reactions were absent, and responses to pinprick or pinch were absent. Self-mutilation limited to the distal digits appeared on the first or second postoperative days and then progressed proximally. The forelimbs were symmetrically affected, and

The cerebellar afferent projection from the trigeminal sensory nuclei (TSN) was studied by means of retrograde axonal transport of horseradish peroxidase (HRP). The projection is almost exclusively ipsilateral. Three cortical regions, viz., the intermediate-lateral part of lobulus simplex with the adjacent area of lobule V, the rostralmost folia of the paramedian lobule with the surrounding parts of crus I and II, and lobule IX, especially its rostral two folia, are the main targets for the cerebellar afferent fibres. A few fibres reach also the other cerebellar regions, as shown in Fig. 3. Mo

The organization of certain of the major afferents to the hippocampus and dentate gyrus has been studied in normal and reeler mutant mice using the autoradiographic and the anterograde degeneration methods. The distribution of the hippocampal and dentate afferents which arise in the medial and lateral parts of the entorhinal cortex and the hippocampus of both sides, has been found to be generally similar to that previously described in the rat, but there are a few minor differences that are discussed in the text. Despite the marked ectopia of many of the neurons in the hippocampal formation in

Integration of our own morphological observations into recent ultrastructural, biochemical, and neuropharmacological results on the carotid body led to a new hypothesis on chemoreceptor function: (i) Glomus cells with small dense-cored vesicles (type IB cells) that store norepinephrine are chemoreceptors. (ii) Glomus cells with large dense-cored vesicles (type IA), which are postsynaptic to the other glomus cell type and presynaptic to afferent nerve endings, are dopaminergic interneurons that suppress the afferent discharge frequency during normoxia by releasing dopamine. (iii) The hypoxic st
