Polymorphism in xeroderma pigmentosum complementation group C codon 939 and aflatoxin B1-related hepatocellular carcinoma in the Guangxi population.

UNLABELLED: Genetic polymorphisms in DNA repair genes may influence individual variations in DNA repair capacity, and this may be associated with the risk and outcome of hepatocellular carcinoma (HCC) related to aflatoxin B1 (AFB1) exposure. In this study, we focused on the polymorphism of xeroderma pigmentosum complementation group C (XPC) codon 939 (rs#2228001), which is involved in nucleotide excision repair. We conducted a case-control study including 1156 HCC cases and 1402 controls without any evidence of hepatic disease to evaluate the associations between this polymorphism and HCC risk

Spermatotoxic effect of aflatoxin B1 in rat: extrusion of outer dense fibres and associated axonemal microtubule doublets of sperm flagellum.

Male Wistar rats were treated with aflatoxin B1 (AFB1). Live as well as methanol-fixed cauda epididymal spermatozoa were stained with acridi … uded. This sperm abnormality is reported for the first time as produced by a dietary toxin. Dietary aflatoxins, therefore, could also be contributory factors for the deterioration of the reproductive health of men.

Hepatitis B virus infection contributes to oxidative stress in a population exposed to aflatoxin B1 and high-risk for hepatocellular carcinoma.

Biomarkers of hepatitis B virus (HBV) infection, aflatoxin B1 (AFB1) exposure and oxidative stress were detected in 71 hepatocellular carcinoma (HCC) patients and 694 controls from southern China. Plasma level of AFB1-albumin-adducts (AAA) and protein carbonyl content (PCC) were significantly higher in the 71 HCC cases than in any age/gender matched HBV sero-status groups (p<0.001). HCC patients positive for the p53-249 G-T mutation had a marginally higher level of PCC than those negative for the mutation (p=0.077). HBV infection had a prominent influence on the association between AFB1 exposu

Hepatocellular carcinoma and polymorphisms in carcinogen-metabolizing and DNA repair enzymes in a population with aflatoxin exposure and hepatitis B virus endemicity.

High rates of hepatocellular carcinoma (HCC) in The Gambia, West Africa, are primarily due to a high prevalence of chronic hepatitis B virus infection and heavy aflatoxin exposure via groundnut consumption. We investigated genetic polymorphisms in carcinogen-metabolizing (GSTM1, GSTT1, HYL1*2) and DNA repair (XRCC1) enzymes in a hospital-based case-control study. Incident HCC cases (n = 216) were compared with frequency-matched controls (n = 408) with no clinically apparent liver disease. Although the prevalence of variant genotypes was generally low, in multivariable analysis (adjusting for d

Palladium catalyzed kinetic and dynamic kinetic asymmetric transformations of gamma-acyloxybutenolides. Enantioselective total synthesis of (+)-Aflatoxin B1 and B2a.

The reaction of gamma-tert-butoxycarbonyloxy-2-butenolide with phenol nucleophiles in the presence of a Pd(0) complex with chiral ligands may be performed under conditions that favor either a kinetic resolution or a kinetic asymmetric transformation (KAT) or dynamic kinetic asymmetric transformation (DYKAT). Performing the reaction at high concentration (0.5 M) in the presence of a carbonate base favors the former, i.e., KAT; whereas, running the reaction at 0.1M in the presence of tetra-n-butylammonium chloride favors the DYKAT process. Syntheses of aflatoxin B(1) and B(2a) employs the DYKAT

Altered DNA mutation spectrum in aflatoxin b1-treated transgenic mice that express the hepatitis B virus x protein.

Humans chronically infected with hepatitis B virus (HBV) are at further risk of liver cancer upon exposure to dietary aflatoxin B1 (AFB1), a carcinogenic product of the mold Aspergillus flavus. For the present study, we utilized double-transgenic mice (ATX mice) that express the HBV X protein (HBx) and possess a bacteriophage lambda transgene to evaluate the in vivo effect of HBx expression on AFB1-induced DNA mutations. The expression of HBx correlated with a 24% increase in mutation frequency overall and an approximately twofold increase in the incidence of G/C-to-T/A transversion mutations

Aflatoxin B1-induced rat hepatic hyperplastic nodules do not exhibit a site-specific mutation within the p53 gene.

The weight of accumulated evidence suggests a role for the p53 tumor suppressor gene in the development of human hepatocellular carcinoma (HCC). Most striking is an apparent mutational specificity at codon 249 of the human gene. Aflatoxin B1 (AFB) is a liver-specific carcinogen which causes G to T substitutions. This transversion was detected at codon 249 in about 50% of the analyzed HCC tumors from African and Asian patients. In these geographic regions aflatoxin exposure and hepatitis B viral infection are risk factors for HCC. In contrast to the human data, no mutations at codon 249 were de

Hepatic protection by 16, 16-dimethyl prostaglandin E2 (DMPG) against acute aflatoxin B1-induced injury in the rat.

Studies were conducted to assess the possible protective action of 16,16-dimethyl prostaglandin E2 (DMPG) against acute aflatoxin B1 (AFB1) induced hepatic injury in the rat. Evaluation of liver damage by histopathologic techniques and clinical chemistry indicated that hepatic necrosis was ameliorated by treatment with DMPG even though binding of radiolabeled (3H)-AFB1 to hepatic DNA was unaffected by this prostaglandin. However, DMPG did not protect rats against AFB1-induced mortality. These data suggest that hepatic protection by DMPG was due to mechanisms other than an interference with the

The effect of inducers of mixed-function oxidases on hepatic microsome-mediated aflatoxin B1 transformation in C3H/10T1/2 cells.

The potent hepatotoxin and hepatocarcinogen aflatoxin B1 (AFB1) is metabolized by different forms of cytochrome P450 associated with the hepatic mixed-function oxidase system. C3H/10T1/2(10T1/2) cells, which have limited inherent capacity to metabolize AFB1, were treated with AFB1 in the presence of hepatic microsomes isolated from chemically treated rats to investigate the effects of the induction of specific cytochromes P450 on AFB1-mediated toxicity and transformation. Relative to uninduced microsomes, phenobarbital (PB) treatment induced AFB1-DNA binding (essentially representing the forma

Efficacy evaluation of a fermented milk containing inactivated biomasses of Lacticaseibacillus rhamnosus and Saccharomyces cerevisiae to ameliorate the toxic effects of dietary aflatoxins and fumonisins in pigs: a pilot study.

Inactivated microorganisms may reduce mycotoxins' bioavailability, although in vivo evidence is limited especially in farm animals. This pilot study evaluated whether a fermented milk (FM) containing heat-inactivated biomasses of Lacticaseibacillus rhamnosus and Saccharomyces cerevisiae at 1% (w/w) mitigated the toxic effects of dietary aflatoxins (AFs) and fumonisins (FBs) in weaned piglets. Regional prevalence of aflatoxin M1 in milk and milk-based products (2021-2025): Contamination drivers, detection advances, and mitigation strategies.

Aflatoxin M1 (AFM1), a hydroxylated metabolite of aflatoxin B1, is a heat-stable mycotoxin commonly detected in milk and dairy products, posing a significant food safety concern. This review synthesizes data from 98 studies across 53 countries (2021-2025) to evaluate the global occurrence of AFM1 in milk, cheese, and yogurt relative to regulatory limits. Contamination patterns show marked regional variability, with higher exceedances in South Asia and Africa than in Europe and East Asia. Key drivers include contaminated feed, climatic conditions, and seasonal variability. From a food chemistry

Proximity-inducible CRISPR/Cas13a trans-cleavage activity and hydrogel-assisted paper-based distance sensor for the detection of aflatoxin M1.

Accurate and rapid determination of aflatoxin M1 (AFM1) in food is crucial for ensuring the safety of the human diet. In view of this, we proposed a proximity-inducible Cas13a activity and hydrogel-assisted paper-based distance sensor for the detection of aflatoxin M1. The single-stranded DNA (ssDNA) and AFM1 aptamer were split into two parts and linked to the terminus of two separate DNA activator strands, respectively. By introducing AFM1, it stimulated these two DNA activator strands to assemble with CRISPR RNA (crRNA) through spatial proximity mechanism between the target and these two DNA

l-Aspartic acid alleviates aflatoxin B1-induced liver injury by regulating intestinal microflora homeostasis and bile acid metabolism.

BACKGROUND: Dietary aflatoxin B1 (AFB1) contamination poses a significant threat to livestock health and food safety by causing severe liver injury. l-Aspartic acid (l-Asp) is known for its antioxidant and anti-inflammatory properties but its potential to mitigate AFB1-induced hepatotoxicity and associated intestinal dysbiosis in rabbits remains unexplored. To address this, 35-day-old rabbits (n = 50) were assigned randomly to five groups: a control group, an AFB1 (0.3 mg kg-1 bodyweight) group, and three groups receiving dietary l-Asp supplementation (0.5%, 1.0%, or 1.5%) for 21 days, with AF

Plant growth regulator diethyl aminoethyl hexanoate suppresses Aspergillus flavus growth and aflatoxin biosynthesis through disruption of redox homeostasis.

Aflatoxin B₁ (AFB₁), produced by Aspergillus flavus, is a major threat to food safety and human health. In this study, we report that diethyl aminoethyl hexanoate (DA-6), a plant growth regulator, exhibits strong antifungal and anti-aflatoxigenic activities against A. flavus. DA-6 significantly inhibited mycelial growth, conidial germination, and sclerotia formation in a dose-dependent manner, while also effectively suppressing fungal colonization in stored rice bran and maize. Microscopic and biochemical analyses revealed that DA-6 disrupted plasma membrane integrity, leading to cytoplasmic l

Simultaneous determination and degradation of aflatoxin B1 using a covalent-organic framework initiated hydrogel and aptamer-based colorimetric sensing platform.

As the most dangerous mycotoxin, aflatoxin B1 (AFB1) has caused some food safety issues to be concerned. In this study, a simultaneous detection and degradation method towards AFB1 was established. Covalent-organic frameworks (COFs) were firstly synthesized and directly in situ deposited on the stainless-steel mesh, which would trigger the free-radical polymerization of acrylamide to form a hydrogel coating. The synthesized Pt@Au nanoparticles as a colorimetric signal probe exhibited the high peroxidase catalytic activity (Km = 0.14 mM, Vmax = 4.57 × 10-8 mol (L s)-1, Kcat = 1.40 × 106 s-1). C

Persistent luminescent nanoparticle-based immunochromatographic assay integrated with a smartphone device for simultaneous detection of aflatoxin B1 and T-2 toxin.

Conventional multicolor fluorescent labeling has inconsistent signal and low quantum yield, restricting sensitivity for low-abundance mycotoxin detection. This study developed a dual-channel immunochromatographic assay (ICA) strip based on color-coding properties of persistent luminescent nanoparticles (PLNPs). By precisely controlling the type of doping ions in Zn2GeO₄ (ZGO) and optimizing the reaction pH, PLNPs with intense green and blue emission were successfully synthesized. The as-prepared PLNPs exhibit high quantum yield and robust optical stability, which effectively reduce background

A Cross-Sectional Association Between Serum Aflatoxin and Micronutrient Status Among Children Aged 6-24 Months in Rural Tanzania.

Micronutrient deficiencies are a significant public health problem, particularly affecting children under five, caused by inadequate intake of micronutrient-rich foods or environmental factors like aflatoxin exposure. Three hundred sixty-nine children aged 6-24 months from Tanzania's Babati and Hanang districts participated in this study. Serum aflatoxin albumin adduct (AF-alb) levels were assessed as measures of aflatoxin exposure. Haemoglobin levels, serum ferritin, C-reactive protein, zinc and vitamins A, B9, and B12 were assessed to determine anaemia and deficiencies in iron, zinc and vita
